TZ1391: a computationally designed circular mRNA multi-epitope vaccine candidate against Mycobacterium tuberculosis via TLR3 immunomodulation.

Tuberculosis (TB), caused by Mycobacterium tuberculosis, remains a major global health burden due to latent infection, multidrug resistance, and the limited efficacy of the BCG vaccine. To address this challenge, we computationally designed and evaluated a circular mRNA-based multi-epitope vaccine candidate, TZ1391. Five experimentally validated M. tuberculosis antigens (ESAT-6, CFP-10, Ag85B, PPE18, and HspX) were used to predict immunodominant cytotoxic T lymphocyte (CTL), helper T lymphocyte (HTL), and B-cell epitopes. Three vaccine constructs (MTB-C1, MTB-C2, and MTB-C3) were assembled by

Computational design, structural modeling and immune evaluation of an amastin-based multi-epitope vaccine candidate against leishmaniasis.

Leishmaniasis is a neglected tropical disease caused by protozoan parasites of the genus Leishmania. Currently, no licensed human vaccine is available and increasing drug resistance continues to compromise the disease control. The present study aimed to computationally design and evaluate a multi-epitope vaccine candidate targeting the conserved amastin protein of Leishmania donovani (UniProt ID: A5XDA6) using an integrated immunoinformatics approach. Cytotoxic T lymphocyte (CTL), helper T lymphocyte (HTL) and B cell epitopes were predicted and sequentially prioritized on the basis of HLA-bind

The two-faced T cell epitope: examining the host-microbe interface with JanusMatrix.

Advances in the field of T cell immunology have contributed to the understanding that cross-reactivity is an intrinsic characteristic of the T cell receptor (TCR), and that each TCR can potentially interact with many different T cell epitopes. To better define the potential for TCR cross-reactivity between epitopes derived from the human genome, the human microbiome, and human pathogens, we developed a new immunoinformatics tool, JanusMatrix, that represents an extension of the validated T cell epitope mapping tool, EpiMatrix. Initial explorations, summarized in this synopsis, have uncovered w

Differential epitope prediction across diverse circulating variants of SARS-COV-2 in Brazil.

COVID-19, caused by the SARS-COV-2 virus, induces numerous immunological reactions linked to the severity of the clinical condition of those infected. The surface Spike protein (S protein) present in Sars-CoV-2 is responsible for the infection of host cells. This protein presents a high rate of mutations, which can increase virus transmissibility, infectivity, and immune evasion. Therefore, we propose to evaluate, using immunoinformatic techniques, the predicted epitopes for the S protein of seven variants of Sars-CoV-2. MHC class I and II epitopes were predicted and further assessed for their

Identification of conserved cross-species B-cell linear epitopes in human malaria: a subtractive proteomics and immuno-informatics approach targeting merozoite stage proteins.

Human malaria, caused by five Plasmodium species (P. falciparum, P. vivax, P. malariae, P. ovale, and P. knowlesi), remains a significant global health burden. While most interventions target P. falciparum, the species associated with high mortality rates and severe clinical symptoms, non-falciparum species exhibit different transmission dynamics, remain hugely neglected, and pose a significant challenge to malaria elimination efforts. Recent studies have reported the presence of antigens associated with cross-protective immunity, which can potentially disrupt the transmission of various Plasm

Subtractive proteomics-guided vaccine targets identification and designing of multi-epitopes vaccine for immune response instigation against Burkholderia pseudomallei.

In this study, a methodical workflow using subtractive proteomics, vaccine designing, molecular simulation, and agent-based modeling approaches were used to annotate the whole proteome of Burkholderia pseudomallei (strain K96243) for vaccine designing. Among the total 5717 proteins in the whole proteome, 505 were observed to be essential for the pathogen's survival and pathogenesis predicted by the Database of Essential Genes. Among these, 23 vaccine targets were identified, of which fimbrial assembly chaperone (Q63UH5), Outer membrane protein (Q63UH1), and Hemolysin-like protein (Q63UE4) were

A conserved subunit vaccine designed against SARS-CoV-2 variants showed evidence in neutralizing the virus.

Novel coronavirus (SARS-CoV-2) leads to coronavirus disease 19 (COVID-19), declared as a pandemic that outbreaks within almost 225 countries worldwide. For the time being, numerous mutations have been reported that led to the generation of numerous variants spread more rapidly. This study aims to establish an efficient multi-epitope subunit vaccine that could elicit both T-cell and B-cell responses sufficient to recognize three confirmed surface proteins of the virus. The sequences of the viral surface proteins, e.g., an envelope protein (E), membrane glycoprotein (M), and S1 and S2 domain of

Potential of a novel flagellin epitope as a broad-spectrum vaccine candidate against enteric fever.

Relentless emergence of antibiotic resistant Salmonella strains, coupled with the drawbacks associated with currently available vaccines against enteric fever, warrants an urgent need to look for new vaccine candidates. Out of the multiple virulence factors harbored by Salmonella, flagella are regarded as one of the most important targets of innate as well as adaptive immune response. Individual Salmonella serotypes alternate between expression of two different antigenic forms encoded by fliC and fljB genes, respectively thereby employing this as a strategy to escape the host immune response.

A computational method to design broad-spectrum T cell-inducing vaccines applied to Betacoronaviruses.

Antigenically diverse pathogens such as coronaviruses pose substantial global health threats, highlighting the need for broad-spectrum vaccines. Here, we introduce Spectravax, a computational method that designs broad-spectrum vaccines accounting for genetic diversity in both host and pathogen populations. Using Spectravax, we designed a nucleocapsid (N) antigen to elicit cross-reactive immune responses to viruses from the Sarbecovirus and Merbecovirus subgenera of Betacoronaviruses. In silico analyses demonstrated superior predicted host and pathogen coverage for Spectravax compared to wild-t

In-silico design of a multi-epitope vaccine targeting conserved transmembrane proteins of Gardnerella vaginalis using immuno-informatics approach.

BACKGROUND: Gardnerella vaginalis is one of the major etiological agents of bacterial vaginosis, which form persistent biofilms and impair the efficacy of antimicrobial agents and trigger recurrent episodes. As no licensed vaccine is currently in existence, computation-aided vaccinology is an affordable method to discover new immunogenic targets. The present study was carried out to develop multi-epitope subunit vaccine against conserved transmembrane proteins of G. vaginalis, utilizing an immune-informatics pipeline. METHODS: Transmembrane proteins were identified as conserved by ClustalW mu

Bioinformatics, expression, and immunogenicity analysis of MPXV A7 protein.

Since the World Health Organization designated the Monkeypox (mpox) outbreak a Public Health Emergency of International Concern (PHEIC) in 2022, monkeypox virus (MPXV) has spread across all six continents. However, the virus's genomic variability and strong adaptability have thus far hindered the development of effective, targeted vaccines. In this study, bioinformatic analysis demonstrated that the A7 protein of MPXV is highly conserved among orthopoxviruses and contains hydrophilic B-cell epitopes. The recombinant A7 protein was expressed using a prokaryotic system, followed by purification

In silico design of a multi-epitope vaccine targeting DENV-1 and DENV-3.

The co-infection of DENV-1 and DENV-3 during endemic outbreaks can be potentially fatal and complicate the diagnostic process. In our study, we have focused on the development of a multiepitope vaccine against DENV-1 and DENV-3 co-infection, utilizing non-structural protein 1 (NS1) and envelope protein (E) as key antigens. B cell and T cell epitopes were predicted for their immunogenicity, antigenicity, and ability to elicit an IFN-γ response. The final construct showed predicted stability (Instability Index: 30.63), antigenicity (0.5509), non-allergenicity, and hydrophilic character (GRAVY: -

Cytotoxic and immunomodulatory activity of CD151-LEL-based peptides in breast cancer and THP-1 cells.

CD151, a cell surface oncoprotein belongs to the tetraspanin family, drives metastasis, drug resistance, and immune escape, and is recognized as a tumor-associated antigen (TAA). In the screening phase, a positive correlation between CD151 expression and amplification of CCND1, ERBB2, and MYC genes as well with breast cancer (BC) metastasis was observed using data from different GEO datasets. This study identified three immunogenic peptides within the large extracellular loop (LEL) of CD151 using in silico approaches: one each of CTL and HTL epitope, which bind strongly to class I and II MHC m

AI-aided design of multi-epitope peptide vaccine elicits cellular and humoral immunity to broad Omicron subvariants.

Peptide vaccines offer a flexible, rapidly updatable platform for responding to SARS-CoV-2 antigenic drift. However, many candidates target a single epitope class, predominantly CD4 +/CD8 + T-cell epitopes or short linear B-cell epitopes, limiting their capacity to induce both high-titer neutralizing antibodies and robust T-cell immunity. Here, to elicit potent humoral and cellular responses, we used AI-aided epitope prediction tools to analyze 18 Omicron subvariants and designed two candidate peptides from the RBD of XBB.1.5, designated LY54-XBB (L455-Y508) and P67-XBB (Y351-K378). In silico

Integrative immunogenomic strategy for designing a multi-epitope vaccine against Zika and Dengue viruses.

Two of the most prevalent illnesses spread by mosquitoes are the Dengue virus (DENV) and the Zika virus (ZIKV), both of which belong to the Flaviviridae family. Co-infection with ZIKV and DENV has been reported worldwide, sometimes with dire consequences. Developing a single vaccine capable of protecting against both pathogens would therefore be highly beneficial. In this research, an in-silico immunology strategy was employed to construct a multi-epitope, multi-pathogen vaccine targeting both viruses. Considering that DENV and ZIKV share the same mosquito vector, Aedes aegypti, and that its s

MIC13 of Toxoplasma gondii: Potential Gene for Vaccine Candidate-An In Silico Approach.

Purpose: Toxoplasmosis, which is the result of infection by Toxoplasma gondii (T. gondii), is a widespread parasitic disease that affects approximately one-third of the global population. Routine medications are not able to eradicate the parasites enclosed in cysts that reside inside the hosts that are infected. MIC13 is believed to have a significant function in facilitating the dissemination of the parasite throughout the host organism. The objective of this research was to utilize immunoinformatics techniques for antigenic analysis and structural prediction of the MIC13 protein, with the go

Bioinformatics analysis to design a multi-epitope mRNA vaccine against S. agalactiae exploiting pathogenic proteins.

Antibiotic resistance in bacterial pathogen infections is a growing global issue that occurs due to their adaptation to changing environmental conditions. Therefore, producing an efficient vaccine as an alternative approach can improve the immune system, eradicate related pathogens, and overcome this growing problem. Streptococcus agalactiae belongs to group B Streptococcus (GBS). Colonization of GBS during pregnancy is a significant risk factor for infants and young children. S. agalactiae infected population exhibits resistance to beta-lactams, including penicillin and the second-line antibi

Designing a multi-epitope subunit vaccine against Toxoplasma gondii through reverse vaccinology approach.

The parasite Toxoplasma gondii, or T. gondii, is zoonotic that both individuals as well as animals can contract resulting in toxoplasmosis, a life-threatening illness. We used an immunoinformatic technique in our research to construct a vaccine with multi-epitopes so that it can decrease the devastating impact caused by this dangerous parasite. In order to construct the vaccine, GRA6 and MIC3 proteins were targeted, which are engaged in T. gondii identification, infection, and immune response. Novel epitopes for linear B lymphocytes (LBL), cytotoxic T lymphocytes (CTL), and helper T lymphocyte

Prediction of an immunogenic peptide ensemble and multi-subunit vaccine for Visceral leishmaniasis using bioinformatics approaches.

Visceral Leishmaniasis (VL) is a neglected tropical disease of public health importance in the Indian subcontinent. Despite consistent elimination initiatives, the disease has not yet been eliminated and there is an increased risk of resurgence from active VL reservoirs including asymptomatic, post kala azar dermatitis leishmaniasis (PKDL) and HIV-VL co-infected individuals. To achieve complete elimination and sustain it in the long term, a prophylactic vaccine, which can elicit long lasting immunity, is desirable. In this study, we employed immunoinformatic tools to design a multi-subunit epi
