The variability of individual tolerance to methotrexate in cancer patients.

Individual tolerance to single or widely spaced doses of methotrexate was explored in 49 patients with advanced cancer with normal serum creatinine and/or blood urea nitrogen. Methotrexate was given as an intravenous infusion over 1 hour at initial doses of 80-120 mg./m(2) body surface area. The doses were increased by 50% increments every 2 weeks until moderate toxicity occurred, arbitrarily defined as leukopenia <5000/mm.(3), and/or thrombocytopenia <100,000/mm.(3), and/or the appearance of oral mucous or intestinal toxicity. The individual dose required to produce initial evidence of toxicit

An investigation of dental anomalies in individuals with amelogenesis imperfecta.

PURPOSE: To explore the presence of dental anomalies in children with amelogenesis imperfecta (AI) and to describe their distribution across different AI genetic variants. METHODS: This cross-sectional study undertook secondary analysis of data from a primary study of children with AI below 18 years of age. After training and calibration, participants' panoramic radiographs and clinical photographs were assessed for the presence of six dental anomalies (pre-eruptive crown resorption, eruption abnormalities, pulp calcifications, taurodontism, gingival enlargement and anterior open bite). The e

Mapping individual molecular connectomes in Alzheimer's disease.

INTRODUCTION: Mapping individual differences is crucial to improve personalized medicine approaches in Alzheimer's disease (AD), which is characterized by strong inter-individual variability in the accumulation patterns of tau and amyloid beta pathology. METHODS: We assess the progression of AD across the disease continuum by building individual molecular connectomes using longitudinal positron emission tomography (PET) data. RESULTS: We demonstrate that these connectomes constitute a unique fingerprint, capable of identifying a single individual from a large group of subjects. Alterations i

Examining Employment Trajectories of Individuals With Mental Disorders.

OBJECTIVE: This study aimed to explore the employment trajectories of individuals with mental disorders, including those with a substance use disorder, who received individual placement and support (IPS) services. METHODS: A growth mixture modeling analysis of Supported Employment Demonstration study data was used to identify distinct employment trajectories. The analysis focused on determinants of variations in work participation across different trajectories of 1,656 study participants. RESULTS: Five distinct employment trajectories were found, with work history, educational attainment, an

Multiracial individuals' perspectives on participating in genetics research.

The vast majority of genetics research is confined to a relatively narrow subset of the global population, limiting the benefits of this research. Recommendations to ameliorate this issue frequently include calls to recruit diverse populations through community engagement, an approach that has been effective in genetics, but has not explicitly included Multiracial individuals, despite this being the fastest-growing population in the United States. As such, this study explored Multiracial individuals' perspectives on: (1) what "Multiracial community" means; (2) concerns regarding ancestry-based

Needs of professionals working with individuals with FASD in Spain.

Prenatal alcohol exposure is one of the main preventable causes of developmental disorders and health anomalies in children. Fetal Alcohol Spectrum Disorder (FASD) is a diagnostic term that encompasses a range of physical, cognitive, and behavioural impairments. Although this condition is gradually attracting greater attention among professionals, knowledge and awareness of its real impact - and of how to intervene effectively - remains limited. This lack of understanding shapes the work of professionals when they must support individuals affected by FASD. The aim of this study is to explore t

Thyroid Hormone Use in Individuals With Eating Disorders.

OBJECTIVE: Individuals with eating disorders are at an increased risk for misuse of thyroid hormone for several reasons, including the overlap in many of the clinical symptoms between malnutrition and hypothyroidism, changes to the hypothalamic-pituitary-thyroid axis, and increased frequency of compensatory behaviours. The purpose of this descriptive study is to investigate thyroid hormone use in individuals with eating disorders and its impact on thyroid function tests. METHOD: In this retrospective, descriptive study, chart review was used to investigate thyroid hormone use for 341 individu
