Two viruses that cause salivary gland hypertrophy in Glossina pallidipes and Musca domestica are related and form a distinct phylogenetic clade.

Glossina pallidipes and Musca domestica salivary gland hypertrophy viruses (GpSGHV and MdSGHV) replicate in the nucleus of salivary gland cells causing distinct tissue hypertrophy and reduction of host fertility. They share general characteristics with the non-occluded insect nudiviruses, such as being insect-pathogenic, having enveloped, rod-shaped virions, and large circular double-stranded DNA genomes. MdSGHV measures 65x550 nm and contains a 124 279 bp genome (approximately 44 mol% G+C content) that codes for 108 putative open reading frames (ORFs). GpSGHV, measuring 50x1000 nm, contains a

Comparative analysis of the nutritional composition and volatile compounds in male and female adults, nymphs, and molts of Eupolyphaga sinensis Walker.

ABSTRAC: Female adult Eupolyphaga sinensis Walker (FAESW) has traditionally been a food source in Southeast Asian countries such as China and India, due to its rich nutritional content. However, the nutritional value of male adults (MAESW) and its molts (MESW) has hardly been reported. Therefore, this study aims to explore the potential application of MAESW and MESW in food by investigating and comparing their nutritional composition (i.e., protein, amino acids, fatty acids, and essential elements) with traditional sources of nutrition. The protein content of MAESW and MESW was 66.10 ± 0.49% a

Analysis of Microbial Composition of Edible Insect Products Available for Human Consumption within the United States Using Traditional Microbiological Methods and Whole Genome Sequencing.

Edible insects offer a promising protein source for humans, but their food safety risks have not been previously investigated within the United States. Therefore, the aim of this study was to investigate the microbial content of processed edible insect products. A total of eight different types of edible insect products, including diving beetles, silkworms, grasshoppers, Jamaican crickets, mealworms, mole crickets, whole roasted crickets, and 100% pure cricket powder, were purchased from a large online retailer for the analysis. All the products were purchased in August 2022 and examined betwe

Cloning and identification of an oxytocin/vasopressin-like receptor and its ligand from insects.

More than 20 years ago, an oxytocin/vasopressin-like peptide, CLITNCPRGamide, was isolated from the locust, Locusta migratoria [Proux JP, et al. (1987) Identification of an arginine vasopressin-like diuretic hormone from Locusta migratoria. Biochem Biophys Res Commun 149:180-186]. However, no similar peptide could be identified in other insects, nor could its prohormone be cloned, or its physiological actions be established. Here, we report that the recently sequenced genome from the red flour beetle Tribolium castaneum contains a gene coding for an oxytocin/vasopressin-like peptide, identical

Purification, characterization, and primary structure of four depressant insect-selective neurotoxin analogs from scorpion (Buthus sindicus) venom.

Four depressant insect-selective neurotoxin analogs (termed Bs-dprIT1 to 4) from the venom of the scorpion Buthus sindicus were purified to homogeneity in a single step using reverse-phase HPLC. The molecular masses of the purified toxins were 6820.9, 6892.4, 6714.7, and 6657.1 Da, respectively, as determined by mass spectrometry. These long-chain neurotoxins were potent against insects with half lethal dose values of 67, 81, 103, and 78 ng/100 mg larva and 138, 160, 163, and 142 ng/100 mg cockroach, respectively, but were not lethal to mice even at the highest applied dose of 10 microg/20 g m

Integrating Siddha dietary principles with alternative protein systems: A pathway toward sustainable and functional nutrition.

The world food systems are facing the fundamental challenges that are being necessitated by climatic changes, increasing population count, protein demand and the escalating non-communicable diseases. Ecological nutrition approaches should therefore be concerned with ecological stability and functional health outcomes. The South Indian traditional medicine is Siddha medicine, an ancient system that provides a constitution-based dietary system that focuses on the three humours, i.e., Vatham, Pitham and Kabam, and the concept of food as medicine. In line with this, other proteins, such as plant-b

Folding motifs induced and stabilized by distinct cystine frameworks.

Bioactive peptides of different sources and biological functionalities, like endothelins, sarafotoxins, bee and scorpion venom toxins, contain a consensus cystine framework, Cys-(X)1-Cys/Cys-(X)3-Cys, which has been found to induce and stabilize a homologous folding motif named the cystine-stabilized alpha-helix (CSH). This is composed of an alpha-helical segment spanning the Cys-(X)3-Cys sequence portion that is crosslinked by two disulfide bridges to the sequence portion Cys-(X)1-Cys, itself folded in an extended beta-strand type structure. Spore-Associated Proteins Involved in c-di-GMP Synthesis and Degradation of Bacillus anthracis.

Bis-(3'-5')-cyclic-dimeric GMP (c-di-GMP) is an important bacterial regulatory signaling molecule affecting biofilm formation, toxin production, motility, and virulence. The genome of Bacillus anthracis, the causative agent of anthrax, is predicted to encode ten putative GGDEF/EAL/HD-GYP-domain containing proteins. Heterologous expression in Bacillus subtilis hosts indicated that there are five active GGDEF domain-containing proteins and four active EAL or HD-GYP domain-containing proteins. Using an mCherry gene fusion-Western blotting approach, the expression of the c-di-GMP-associated protei

The full-length cDNA of anticoagulant protein infestin revealed a novel releasable Kazal domain, a neutrophil elastase inhibitor lacking anticoagulant activity.

Infestins are Kazal-type serine proteinase inhibitors found in the midgut of the Chagas' disease vector, Triatoma infestans. In previous studies, we characterized two double-headed infestins with potent anticoagulant activity; infestin 1-2, which inhibits thrombin and infestin 3-4, a factor XIIa inhibitor. In the present work, we have cloned the full-length cDNA of infestins' precursor. The translated cDNA predicted a polypeptide containing a signal peptide and seven Kazal-type domains, four domains from infestin 1-2 and infestin 3-4, and three new domains. Northern blot analysis confirmed tha

Protein localization and potential function of lipocalin in Reticulitermes speratus queens.

To understand the mechanisms underlying social evolution and caste development in social insects, caste-specific organs and genes should be investigated. In the rhinotermitid termite, Reticulitermes speratus, the lipocalin gene RS008881, which encodes a protein transporter, is expressed in the ovarian accessory glands of primary queens. To obtain additional data on its expression and product localization, we conducted real-time quantitative polymerase chain reaction and protein assays using a peptide antibody. Gene expression analysis of the castes revealed that RS008881 was highly expressed i

Identification, characterization and hypolipidemic effect of novel peptides in protein hydrolysate from Protaetia brevitarsis larvae.

The proteins were mainly derived from Protaetia brevitarsis larval extracts obtained using two empty intestine methods (traditional static method: TSM or salt immersion stress method: SISM) and extraction solvents (water: W or 50 % water-ethanol: W:E), and the proteins were used as objects to investigate the effect of emptying intestine methods on hypolipidemic peptides. The results revealed that the F-2 fractions of protein hydrolysate had stronger in vitro hypolipidemic activity, with the peptides obtained by SISM possessing a stronger cholesterol micelle solubility inhibition rate, especial

Do insect food-derived ecdysteroids appear in the blood of waterfowl, predatory birds, carnivorous mammals and primates?

Arthropod molting hormones, the so-called ecdysteroids were experimentally shown to have anabolic effects in higher vertebrates, promoting energy supply, protein synthesis, and tissue growth. Insectivorous songbirds (Aves: Passeriformes) and bats (Mammalia: Chiroptera) were demonstrated to have naturally acquired ecdysteroids in their blood. However, it is not known if partly insectivorous or artificially insect-fed vertebrates would also have ecdysteroids in their circulation. The aim of this screening study was to clarify this, by collecting blood samples of mammals, birds and reptiles that

Evidence of prior exposure to human bocavirus as determined by a retrospective serological study of 404 serum samples from adults in the United States.

Recently, molecular screening for pathogenic agents has identified a partial genome of a novel parvovirus, called human bocavirus (HBoV). The presence of this newly described parvovirus correlated with upper and lower respiratory tract infections in children. Lower respiratory tract infections are a leading cause of hospital admission in children, and the etiological agent has not been identified in up to 39% of these cases. Using baculovirus expression vectors (BEVs) and an insect cell system, we produced virus-like particles (VLPs) of HBoV. The engineered BEVs express the HBoV capsid protein

Toxins from scratch? Diverse, multimodal gene origins in the predatory robber fly Dasypogon diadema indicate a dynamic venom evolution in dipteran insects.

BACKGROUND: Venoms and the toxins they contain represent molecular adaptations that have evolved on numerous occasions throughout the animal kingdom. However, the processes that shape venom protein evolution are poorly understood because of the scarcity of whole-genome data available for comparative analyses of venomous species. RESULTS: We performed a broad comparative toxicogenomic analysis to gain insight into the genomic mechanisms of venom evolution in robber flies (Asilidae). We first sequenced a high-quality draft genome of the hymenopteran hunting robber fly Dasypogon diadema, analyse

Protein-Level Interactions as Mediators of Sexual Conflict in Ants.

All social insects with obligate reproductive division of labor evolved from strictly monogamous ancestors, but multiple queen-mating (polyandry) arose de novo, in several evolutionarily derived lineages. Polyandrous ant queens are inseminated soon after hatching and store sperm mixtures for a potential reproductive life of decades. However, they cannot re-mate later in life and are thus expected to control the loss of viable sperm because their lifetime reproductive success is ultimately sperm limited. In the leaf-cutting ant Atta colombica,, the survival of newly inseminated sperm is known t

Biochemical responses of Anticarsia gemmatalis (Lepidoptera: Noctuidae) in soybean cultivars sprayed with the protease inhibitor berenil.

The damage caused by Anticarsia gemmatalis motivates this study on the adaptive mechanisms of the insect to soybean. The lipoxygenase pathway produces and releases jasmonic acid, involved in the regulation of the plant defense genes, which encodes protease inhibitor (PI) production. Three soybean cultivars IAC-18, IAC-24, and Foscarin-31 were sprayed with water and berenil, a synthetic inhibitor, at 0.60 and 1.0% (w/v) and then infested with A. gemmatalis larvae. The lipoxygenase (LOX) activity increased in the leaves of Foscarin-31, IAC-18, and IAC-24 by 87, 81, and 78%, respectively, after 2

Discovery of markers of exposure specific to bites of Lutzomyia longipalpis, the vector of Leishmania infantum chagasi in Latin America.

BACKGROUND: Sand flies deliver Leishmania parasites to a host alongside salivary molecules that affect infection outcomes. Though some proteins are immunogenic and have potential as markers of vector exposure, their identity and vector specificity remain elusive. METHODOLOGY/PRINCIPAL FINDINGS: We screened human, dog, and fox sera from endemic areas of visceral leishmaniasis to identify potential markers of specific exposure to saliva of Lutzomyia longipalpis. Human and dog sera were further tested against additional sand fly species. Recombinant proteins of nine transcripts encoding secreted

A luminal proteome of the endoplasmic reticulum and Golgi apparatus reveals a novel modulator of ER stress tolerance in African trypanosomes.

African trypanosomes employ specialized mechanisms of membrane trafficking as a strategy to persist in both the mammalian host and insect vector. Their survival relies on continuous synthesis and surface delivery of extremely abundant surface coat proteins, imposing an extraordinary biosynthetic burden on the secretory pathway. Despite this, their ER and Golgi luminal proteomes remain incompletely characterized. Here, we exploit proximity biotinylation, using the abundant ER chaperone BiP as bait to map the ER proteome in bloodstream and procyclic lifecycle stages. Comparison with BiPN, a trun

Integrated proteomic analysis reveals differential remodelling of defense responses in potato to herbivory by Phthorimaea operculella.

Plants rely on both transcriptional reprogramming and post-translational machinery to regulate their responses to biotic stress. While transcriptome-wide dynamics underlying plant defense against insect herbivores have been extensively documented, systematic investigations into protein-level regulation and integrative analyses of these multi-layered regulatory networks remain limited. In this study, we employed quantitative proteomics and phospho-proteomics to characterize the protein-level responses of potato plants to attacks by Phthorimaea operculella. A total of 60 differentially expressed

Molecular cloning and expression under cold stress of two heat shock proteins from Dastarcus helophoroides (Fairmaire) (Coleoptera: Bothrideridae).

Heat shock proteins (HSPs) maintain cellular homeostasis and regulate immune responses across species. HSP90 stabilises and activates regulatory proteins, while HSP70 facilitates protein folding and prevents aggregation. In this study, the complete cDNA sequences of Dastarcus helophoroides - HSP70, HSP90, and β-actin (used as a reference for real-time quantitative polymerase chain reaction) - were obtained using rapid amplification of cDNA ends-polymerase chain reaction, and the expression of HSP70 and HSP90 under cold stress was analysed. The full-length cDNA of HSP90 contains a 2346-bp open
