Blood recirculating devices, such as ventricular assist devices and prosthetic heart valves, are burdened by thromboembolic complications requiring complex and lifelong anticoagulant therapy with its inherent hemorrhagic risks. Pathologic flow patterns occurring in such devices chronically activate platelets, and the optimization of their thrombogenic performance requires the development of flow-induced platelet activation models. However, existing models are based on empirical correlations using the well-established power law paradigm of constant levels of shear stress during certain exposure

Psychosocial stress promotes and links mood and cardiovascular disorders in a sex-specific manner. However, findings in animal models are equivocal, in some cases opposing human dimorphisms. We examined central nervous system (CNS), behavioral, endocrine, cardiac, and hepatic outcomes in male or female C57Bl/6 mice subjected to chronic social stress (56 days of social isolation, with intermittent social confrontation encounters twice daily throughout the final 20 days). Females exhibited distinct physiological and behavioral changes, including relative weight loss, and increases in coronary re

BACKGROUND AND PURPOSE: Studies in sepsis suggest that mitochondria mediate multiple organ dysfunction, including cardiac failure; however, the underlying molecular mechanisms remain elusive. This study examined changes in mitochondrial membrane integrity, antioxidant activities, and oxidative stress in the heart after infectious challenge (intratracheal Streptococcus pneumoniae, 4 x 10(6) colony-forming units). Inflammation responses also were examined.
METHODS: Cardiac tissues were harvested from Sprague-Dawley rats 4, 8, 12, and 24 h after bacterial challenge (or intratracheal vehicle for

Radiation-induced toxicity of the digestive tract is a major clinical concern as many cancer survivors have received radiotherapy for tumours of the abdominopelvic area. The coordination and orchestration of a tissue's response to stress depend not only on the phenotype of the cells that make up the tissue but also on cell-cell interactions. The digestive system, i.e., the intestine/colon/rectum, is made up of a range of different cell populations: epithelial cells, stromal cells, i.e. endothelial cells and mesenchymal lineages, immune cells and nerve cells. Moreover, each of these populations

Dental implants need to combine mechanical strength with promoted osseointegration. Currently used subtractive manufacturing techniques require a multi-step process to obtain a rough surface topography that stimulates osseointegration. Advantageously, additive manufacturing (AM) enables direct implant shaping with unique geometries and surface topographies. In this study, zirconia implants with integrated lamellar surface topography were additively manufactured by nano-particle ink-jetting. The ISO-14801 fracture load of as-sintered implants (516±39 N) resisted fatigue in 5-55 °C water thermo-

Successful gastrointestinal (GI) colonization by bacterial pathogens requires adaptation to nutrient competition and host-derived stresses in the gut, with adaptation via the bacterial stringent stress response playing a critical role. Epidemiological data suggest that the GI tract serves as a reservoir from which K. pneumoniae can spread and cause invasive disease or transmit to another host. DksA is a conserved stringent response transcriptional regulator that was identified in an in vivo transposon mutagenesis screen as an important K. pneumoniae gut determinant. However, its role in K. pne

Paracoccidioides brasiliensis is a pathogenic fungus capable of thermodimorphic transition, causing systemic mycoses in humans. Fludioxonil, a phenylpyrrole fungicide, inhibits this transition and affects genes related to fungal cell wall composition. This study explored the proteomic response of P. brasiliensis yeast cells exposed to fludioxonil, identifying over 4100 proteins, approximately 50% of the total proteome, through DIA (data-independent acquisition) mass spectrometry. Treatment led to significant proteomic changes, with 28 proteins upregulated and 26 downregulated. Upregulated prot

Plants are susceptible to several biotic and abiotic stressors, including pathogens, extreme salinity, temperature, and drought. Continuous exposure to these conditions activates complex molecular mechanisms in plants. These include stress-signaling pathways mediated by phytohormones such as cytokins, salicylic acid, auxins, abscisic acid, jasmonic acid, and ethylene. These signaling molecules further interact with cellular pathways regulating stress-responsive gene expression through transcription factors such as DREBs, MYBs, bZIPs, and NACs. Key cellular pathways include mitogen-activated pr

The human gut microbiome plays a central role in human health, and has been implicated in the development of a number of chronic gastrointestinal and systemic diseases. For example, microorganisms can serve as microbial endocrine mediators and can respond to stimuli and produce neurochemicals, ultimately influencing the brain-gut-microbiome axis of their host, a bidirectional communication system between the central nervous system and the gastrointestinal tract, especially during developmental stages. To begin to explore potential dynamic changes of the gut microbiome, we characterized gut mic

Mutations in CHCHD10, a mitochondrial intermembrane space (IMS) protein implicated in proteostasis and cristae maintenance, cause mitochondrial disease. Knock-in mice modeling the human CHCHD10S59L variant associated with ALS-FTD develop a mitochondrial cardiomyopathy driven by CHCHD10 aggregation and activation of the mitochondrial integrated stress response (mtISR). We show that cardiac dysfunction is associated with dual defects originating at the onset of disease: (1) bioenergetic failure linked to impaired mitochondrial copper homeostasis and cytochrome c oxidation, and (2) maladaptive mt…

Iron overload contributes to proliferative vascular diseases, yet its interplay with hydrogen sulfide (H2S) in vascular smooth muscle cell (VSMC) proliferation remains poorly understood. This study elucidates H2S's role in mitigating iron-overload-induced oxidative stress and cellular damage. Using aortic VSMCs from wildtype (WT) and cystathionine γ-lyase-knockout (CSE-KO) mice treated with ferric ammonium citrate (FAC) at concentrations equivalent to serum levels of iron and citrate, we demonstrate that FAC triggers the integrated stress response (ISR) in WT cells, upregulating CSE to enhance…

Mutations in CHCHD10, a mitochondrial protein with undefined functions, are associated with autosomal dominant mitochondrial diseases. Chchd10 knock-in mice harboring a heterozygous S55L mutation (equivalent to human pathogenic S59L) develop a fatal mitochondrial cardiomyopathy caused by CHCHD10 aggregation and proteotoxic mitochondrial integrated stress response (mtISR). In mutant hearts, mtISR is accompanied by a metabolic rewiring characterized by increased reliance on glycolysis rather than fatty acid oxidation. To counteract this metabolic rewiring, heterozygous S55L mice were subjected t…

AIMS: Heart failure and associated cachexia is an unresolved and important problem. This study aimed to determine the factors that contribute to cardiac cachexia in a new model of heart failure in mice that lack the integrated stress response (ISR) induced eIF2α phosphatase, PPP1R15A.
METHODS AND RESULTS: Mice were irradiated and reconstituted with bone marrow cells. Mice lacking functional PPP1R15A, exhibited dilated cardiomyopathy and severe weight loss following irradiation, whilst wild-type mice were unaffected. This was associated with increased expression of Gdf15 in the heart and incre…

Oxidative stress is a major consequence of environmental, metabolic, and disease-related challenges in poultry and can compromise cellular integrity and function. Although antioxidant defense mechanisms are broadly conserved across vertebrates, oxidative-stress responses in avian cells remain less well characterized than those in mammalian systems. Superoxide dismutase 1 (SOD1) is a key antioxidant enzyme, but how elevated SOD1 expression influences both oxidative injury and stress-responsive transcription in chicken cells remains incompletely understood. In this study, we established a stable

Plants' immobility renders them highly vulnerable to heat stress, which disrupts water relations, photosynthesis, respiration, and cellular homeostasis, ultimately reducing growth and yield. To survive, plants deploy a multifaceted heat stress response (HSR) that integrates calcium signaling, molecular chaperones, antioxidant enzymes, and phytohormonal networks. This review synthesizes recent advances in understanding the molecular crosstalk between phytohormones and protein synthesis during plant heat stress responses, with a particular focus on two key HSR modules: protein synthesis pathways

The immunoregulatory architecture of human oral tissues remains poorly defined despite their central role as barrier interfaces. We present the first integrated single cell and dual platform spatial proteotranscriptomic atlas of oral tissues, profiling >250,000 single-cell transcriptomes and >4 million spatially-resolved cells across 13 niches. Using our AI enabled AstroSuite (TACIT, Constellation, STARComm, hist2omics), we defined tissue cellular neighborhoods (TCNs) and multicellular interaction modules (MCIMs) in health, revealing peri-epithelial fibroblast-centered hubs enriched for effect
