High-Risk Factors of In-Hospital Death Following Complex High-risk and Indicated Patients After Percutaneous Coronary Intervention Supported by Extracorporeal Membrane Oxygenation.

BACKGROUND: Complex high-risk and indicated patients (CHIPs) increase the risk of in-hospital death after percutaneous coronary intervention (PCI). Extracorporeal membrane oxygenation (ECMO) support can improve survival. However, there remains a gap in knowledge regarding how to identify and manage these high-risk patients effectively to reduce mortality. This study aimed to determine the independent high-risk factors associated with increased risk of in-hospital mortality among CHIPs after PCI with ECMO support. This research focused on providing clinicians with more accurate risk assessment

Development of nanobody-conjugated LL37 for synergistic therapy against MDR Acinetobacter baumannii.

Multidrug resistance (MDR) of the pathogen Acinetobacter baumannii is a major challenge to global healthcare due to the limited treatment options. The emergence of MDR bacteria necessitates innovative therapeutic approaches, especially given the associated economic burden and the rapid spread of infections. Conventional treatments such as antibiotics and vaccines face significant obstacles. Antimicrobial peptides (AMPs) such as LL37 have potential as an alternative treatment due to their broad-spectrum activity and ability to target specific bacterial structures such as the outer membrane prot

Characterization and comparative genomic analysis of a novel bacteriophage against an extremely drug-resistant Pseudomonas aeruginosa ST1971.

UNLABELLED: Given the rising prevalence of multidrug-resistant strains, Pseudomonas aeruginosa necessitates alternative therapies, and phages are emerging as promising antibiotic alternatives. Our study introduces a novel lytic phage, CRPAP1904, exhibiting potent activity against an extremely drug-resistant P. aeruginosa ST1971 strain CRPA190. Phylogenetic and comparative genomic analyses validated the novelty of phage CRPAP1904, suggesting that it represents a new species of the Kochitakasuvirus genus. The phage CRPAP1904 has a double-stranded DNA genome of 62,430 bp base pairs, a GC content

A genetically based computational drug repurposing framework for rapid identification of candidate compounds: application to COVID-19.

BACKGROUND: The development and approval of novel drugs are typically time-intensive and expensive. Leveraging a computational drug repurposing framework that integrates disease-relevant genetically regulated gene expression (GReX) and large longitudinal electronic medical record (EMR) databases can expedite the repositioning of existing medications. However, validating computational predictions of the drug repurposing framework remains a challenge. METHODS: To benchmark the drug repurposing framework, we first performed a 5-method-rank-based computational drug prioritization pipeline by inte

Dual orexin receptor antagonists for preventing delirium in hospitalized older adults: protocol for a randomized-trial systematic review and meta-analysis.

BACKGROUND: Delirium is a common, serious, and potentially preventable neuropsychiatric syndrome among hospitalized older adults. It is characterized by acute disturbance in attention and awareness with additional cognitive disturbance and a fluctuating course. Dual orexin receptor antagonists (DORAs), including suvorexant, lemborexant, and daridorexant, promote sleep by antagonizing orexin-mediated wakefulness rather than by direct gamma-aminobutyric acid-ergic sedation. Whether DORAs prevent delirium in hospitalized older adults remains uncertain, and prior syntheses have not fully resolved

Targeting microbial β-CAs: bridging in silico screening with in vitro validation.

Antimicrobial resistance (AMR) is one of the world's most pressing health problems and requires immediate action from the scientific community. Pseudomonas aeruginosa is a particularly concerning bacterium due to its high level of resistance, especially among hospitalized and immunocompromised individuals. We recently investigated PsCA3, one of three P. aeruginosa β‑carbonic anhydrases (β-CAs), as a potential new pharmacological target for developing innovative antibacterial drugs. In this study, we applied a consensus structure-based virtual screening approach to select the most promising PsC

Enhancing adherence: Evaluating interventions for Heart Failure management in older adults.

BACKGROUND: Management of heart failure (HF) is challenging, particularly because of the high rates of medication non-adherence among older adults. This leads to increased hospital readmission and healthcare costs. To address these challenges, a combination of non-pharmacological strategies, such as lifestyle changes and targeted pharmacological interventions, is crucial for improving the outcomes and reducing the burden of HF. OBJECTIVE: This scoping review aims to map out existing literature and highlight the interventions used for HF management to enhance adherence in the older adult popul
