Production of liposomes by microfluidics: The impact of post-manufacturing dilution on drug encapsulation and lipid loss.

Microfluidic mixing is recognized as a convenient method to produce liposomes for its scalability and reproducibility. Numerous studies have described the effect of process parameters such as flow rate ratios and total flow rate on size and size distribution of vesicles. In this work, we focused our attention on the effect of flow rate ratios on the encapsulation efficiency of liposomes, as we hypothesized that different amount of residual organic solvent could affect the retention of lipophilic drug molecules within the bilayer. In a further step, we investigated how the liposomes integrity a

[Channel-forming action--a general property of venoms of spiders in the family Theridiidae (Aranei)].

The study of venom action of 10 spider species of Theridiidae family has shown that they cause the formation of the cation selective channels on the bilayer lipid membranes. This action does not depend on the species sex and age and it is the important diagnostic characteristic of the family. The channel-forming effect is due to the presence of the peptide with molecular weight about 5000 daltons in venoms of Theridiidae spider family.

The plasma membrane of bloodstream-form African trypanosomes confers susceptibility and specificity to killing by hydrophobic peptides.

Trypanosoma brucei is the causative agent of both a veterinary wasting disease and human African trypanosomiasis, or sleeping sickness. The cell membrane of the developmental stage found within the mammalian host, the bloodstream form (BSF), is highly dynamic, exhibiting rapid rates of endocytosis and lateral flow of glycosylphosphatidylinositol-anchored proteins. Here, we show that the cell membrane of these organisms is a target for killing by small hydrophobic peptides that increase the rigidity of lipid bilayers. Specifically, we have derived trypanocidal peptides that are based upon the h…

Calcium Reshapes Aβ Aggregation at Anionic Lipid Membranes.

Dysregulated calcium homeostasis is a hallmark of neurodegenerative disorders, such as Alzheimer's, Parkinson's, and Huntington's diseases. In Alzheimer's disease (AD), the aggregation of amyloid beta (Aβ) peptides at neuronal membranes is shown to be modulated by the presence of Ca2+ ions, yet the molecular mechanism by which Ca2+ reshapes Aβ aggregation at anionic membrane surfaces remains poorly understood. To address this knowledge gap, we employed coarse-grained molecular dynamics simulations to investigate the aggregation of the model amyloidogenic K16LVFFAE22 fragment of Aβ (Aβ16-22), o

Acetylsalicylic acid (aspirin) and salicylic acid interaction with the human erythrocyte membrane bilayer induce in vitro changes in the morphology of erythrocytes.

Despite the well-documented information, there are insufficient reports concerning the effects of salicylate compounds on the structure and … differential scanning calorimetry (DSC) to detect the changes in the thermotropic phase behavior of lipid bilayers resulting from ASA and SA interaction with PC and PE molecules. In an attempt to further elucidate their effects on cell membranes, the present work also examined their influence on the morphology of intact human erythrocytes by means of defocusing …

Volume sensitive efflux of taurine in HEK293 cells overexpressing phospholemman.

The role of the phospholemman (PLM) on the efflux of taurine and chloride induced by swelling was studied in HEK293 cells overexpressing stable transfected PLM. PLM, a substrate for protein kinases C and A, is a protein that induces an anion current in Xenopus oocytes and forms taurine-selective channels in lipid bilayers. Taurine contributes as an osmolyte to regulatory volume decrease (RVD) and is highly permeable through PLM channels in bilayers. In PLM-overexpressing cells the process of RVD was more rapid and efficient (75%) than in control cells (44%). Also, [(3)H]taurine and (125)I effl…

Tarantula toxins use common surfaces for interacting with Kv and ASIC ion channels.

Tarantula toxins that bind to voltage-sensing domains of voltage-activated ion channels are thought to partition into the membrane and bind to the channel within the bilayer. While no structures of a voltage-sensor toxin bound to a channel have been solved, a structural homolog, psalmotoxin (PcTx1), was recently crystalized in complex with the extracellular domain of an acid sensing ion channel (ASIC). In the present study we use spectroscopic, biophysical and computational approaches to compare membrane interaction properties and channel binding surfaces of PcTx1 with the voltage-sensor toxin

Mechanism of lipid transfer by bridge-like protein VPS13A and the scramblase XK.

In eukaryotes, bridge-like lipid-transfer proteins (BLTPs) are central in mediating vesicle-independent lipid transfer between organelles. BLTPs span the cytosolic space between organelles at contact sites, featuring hydrophobic channels for lipids to travel between membranes. How BLTPs cooperate with partner proteins to orchestrate lipid delivery remains a mystery. Here, we used cryo-electron microscopy to visualize a complex comprising the prototypical BLTP VPS13A and the plasma membrane-localized scramblase XK at near-atomic resolution. VPS13A interacts with XK via its pleckstrin homology d

TMC1 and TMC2 are cholesterol-dependent scramblases that regulate membrane homeostasis in auditory hair cells.

TMC1 and TMC2, the pore-forming subunits of the mechanoelectrical transduction (MET) complex in inner ear sensory hair cells, are essential for auditory and vestibular function. Pathogenic mutations in TMC1 are a leading cause of genetic hearing loss, but their underlying cellular mechanisms remain poorly understood. Here, we reveal that TMC1 and TMC2 are cholesterol-regulated lipid scramblases whose activity modulates plasma membrane asymmetry. Using reconstituted proteoliposomes and molecular dynamics simulations, we demonstrate that both proteins facilitate phospholipid translocation across

Structures, functions, and syntheses of glycero-glycophospholipids.

Biological membranes consist of integral and peripheral protein-associated lipid bilayers. Although constituent lipids vary among cells, membrane lipids are mainly classified as phospholipids, glycolipids, and sterols. Phospholipids are further divided into glycerophospholipids and sphingophospholipids, whereas glycolipids are further classified as glyceroglycolipids and sphingoglycolipids. Both glycerophospholipids and glyceroglycolipids contain diacylglycerol as the common backbone, but their head groups differ. Most glycerolipids have polar head groups containing phosphate esters or sugar m…

Ultrasensitive Plasmonic Platform for Label-Free Detection of Membrane-Associated Species.

Lipid membranes and membrane proteins are important biosensing targets, motivating the development of label-free methods with improved sensitivity. Silica-coated metal nanoparticles allow these systems to be combined with supported lipid bilayers for sensing membrane proteins through localized surface plasmon resonance (LSPR). However, the small sensing volume of LSPR makes the thickness of the silica layer critical for performance. Here, we develop a simple, inexpensive, and rapid sol-gel method for preparing thin conformal, continuous silica films and demonstrate its applicability using gold…

BAR domain proteins regulate Rho GTPase signaling.

BAR proteins comprise a heterogeneous group of multi-domain proteins with diverse biological functions. The common denominator is the Bin-Amphiphysin-Rvs (BAR) domain that not only confers targeting to lipid bilayers, but also provides scaffolding to mold lipid membranes into concave or convex surfaces. This function of BAR proteins is an important determinant in the dynamic reconstruction of membrane vesicles, as well as of the plasma membrane. Several BAR proteins function as linkers between cytoskeletal regulation and membrane dynamics. These links are provided by direct interactions betwee…

A layered target-release nanoparticle system for normalizing the lung cancer microenvironment and enhancing antitumor effect.

BACKGROUND: Abnormal tumor vasculature and dense collagen fiber networks contribute to elevated interstitial pressure in solid tumors, compressing blood vessels and impairing the delivery of nanoparticle (NP)-based therapeutics. Promoting normalization of the tumor microenvironment is a promising strategy for enhancing drug penetration. This study aimed to develop a pH-responsive bilayer nanoplatform capable of sequentially delivering a microenvironment modulator and a targeted chemotherapeutic agent for improved efficacy against lung cancer. METHODS: We designed a bilayer lipid NP system wit

An artificial model of ATP-binding cassette mediated transport.

Active drug efflux is a critical bottleneck in multidrug-resistance (MDR) in cancers, bacterial infections and in neurological disease. Preclinical acellular permeation assays do not predict efflux (active) transport, and cell-based assays may be confounded by the complexity and heterogeneity of cells which can make efflux data analysis challenging. We report here a cell-free platform as a model for functional multidrug resistance (MDR) efflux pump that mimics the inner membrane of Escherichia coli (E. coli). The system comprises a PE:PG:CL (67:23.2:9.8 mol%) lipid bilayer reconstituted with A

Molecular details of α-synuclein membrane association revealed by neutrons and photons.

α-Synuclein (α-syn) is an abundant neuronal protein associated with Parkinson's disease that is disordered in solution, but it exists in equilibrium between a bent-helix and an elongated-helix on negatively charged membranes. Here, neutron reflectometry (NR) and fluorescence spectroscopy were employed to uncover molecular details of the interaction between α-syn and two anionic lipids, phosphatidic acid (PA) and phosphatidylserine (PS). Both NR and site-specific Trp measurements indicate that penetration depth of α-syn is similar for either PA- or PS-containing membranes (∼9-11 Å from bilayer

Topological Changes in Lipid Membrane Surfaces Induced by Lowering of pH in Isotonic Conditions.

Through the use of fluorescence probes, FTIR spectroscopy, zeta potential measurements, and surface pressure-area isotherms, it is demonstrated that lowering the pH increases the hydrophobicity of DPPC lipid vesicles in the gel state. This effect is noticeable at pH 2. The increase in hydrophobicity is comparable to that produced by hypertonic stress, in which vesicles shrink due to a decrease in volume and an increase in curvature. However, in the case of pH, no effect on vesicle size, area per lipid, and hydrocarbon chain packing is observed. It is concluded that the protonation of phosphate

The electrical interplay between proteins and lipids in membranes.

All molecular interactions that are relevant to cellular and molecular structures are electrical in nature but manifest in a rich variety of forms that each has its own range and influences on the net effect of how molecular species interact. This article outlines how electrical interactions between the protein and lipid membrane components underlie many of the activities of membrane function. Particular emphasis is placed on spatially localised behaviour in membranes involving modulation of protein activity and microdomain structure. The interactions between membrane lipids and membrane prote

Fluctuation spectra of free and supported membrane pairs.

Fluctuation spectra of fluid compound membrane systems are calculated. The systems addressed contain two (or more) almost parallel membranes that are connected by harmonic tethers or by a continuous, harmonic confining potential. Additionally, such a compound system can be attached to a supporting substrate. We compare quasi-analytical results for tethers with analytical results for corresponding continuous models and investigate under what circumstances the discrete nature of the tethers actually influences the fluctuations. A tethered, supported membrane pair with similar bending rigidities

Stochastic process description of lipid flip-flop.

Since lipid bilayers are self-assembled macroscopic aggregates, their constituent lipid molecules can spontaneously transition between the two leaflets. This so-called "flip-flop" is almost universally described via first-order chemical kinetics: the net "flux" leaving a given leaflet is proportional to the number of lipids it contains. However, this model ignores interactions, such as those arising from packing or non-ideal mixing, and restricting the analysis to macroscopic rate equations misses fluctuations. Here, we employ tools from the field of stochastic processes to examine the impact …
