AlphaFold 3 prediction of TCR mimic Fv - pMHC complex structures and their validation and optimization.

Monoclonal antibody-derived chimeric antigen receptors (CAR) that mimic T‑cell receptors (TCR) on binding to peptide-major histocompatibility complex (pMHC) and activating T‑cell functions hold great promise for the development of effective immunotherapy. This study applied AlphaFold 3 (AF3) to model the quaternary structures of TCR mimic antibody variable fragment (TCRm Fv) complexed with NY-ESO-1157-165/HLA-A*02:01. Benchmark study suggested that reliable TCRm Fv-pMHC structures were achieved by AF3 with high confidence. Toward NY-ESO-1/A2-specific CAR clones, AF3 prediction revealed their i…

The partitioning of TCR repertoires by thymic selection.

αβ T cells are critical components of the adaptive immune system; they maintain tissue and immune homeostasis during health, provide sterili … operties are initially established during T cell development when thymocytes use TCR recognition of major histocompatibility complex (MHC) and MHC-like ligands to instruct survival versus death and to differentiate into a plethora of inflammatory and regulatory T cell lineages. Here, we review the ligand specificity of the preselection thymocyte repertoire and argue that developm …

MHC-DRB alleles with amino acids Val11, Phe13, and the shared epitopes promote collagen-induced arthritis and a rapid IgG1 response in Filipino cynomolgus macaques.

Macaques are useful animal models for studying the pathogenesis of rheumatoid arthritis (RA) and the development of anti-rheumatic drugs. The purpose of this study was to identify the major histocompatibility complex (MHC) polymorphisms associated with the pathology of collagen-induced arthritis (CIA) and anti-collagen IgG induction in a cynomolgus macaque model, as MHC polymorphisms affect the onset of CIA in other animal models. Nine female Filipino cynomolgus macaques were immunized with bovine type II collagen (b-CII) to induce CIA, which was diagnosed clinically by scoring the symptoms of…

Specific TaqMan probed real-time quantitative RT-PCR methods and their application to differentiate the transcripts of duplicated BF or BLB genes in chicken MHC.

BF and BLB genes of chicken major histocompatibility complex (MHC) are responsible for classical antigen processing and presentation; therefore they play a central role in determining the genetic resistance or susceptibility of different MHC-B haplotypes to some infectious diseases. In this study, we developed specific TaqMan probed real-time quantitative reverse transcription PCR (TaqMan qRT-PCR) methods based on the diagnostic nucleotide polymorphisms present in duplicated BF or BLB genes in B2 and B19 haplotypes. The results showed very similar amplification efficiency but no cross-reaction…

Glioblastoma-derived mechanisms of systemic immunosuppression.

Abnormalities of cellular immunity are commonly seen in patients with glioblastoma (GBM), and the subsequent relative immunosuppression likely contributes to poor tumor-specific responses in affected individuals. Endogenous immune regulation is likely to limit the efficacy of a wide array of immunotherapeutic strategies, therefore mandating consideration in the continued development of novel treatments for GBM. Various tumor-associated factors have been implicated as potential generators of the immunosuppressive effect. This article outlines relevant experimentation exploring the nature of imm

Genetics research in systemic lupus erythematosus for clinicians: methodology, progress, and controversies.

PURPOSE OF REVIEW: Clinical journals are reporting genetic associations with systemic lupus erythematosus (SLE) with increasing frequency. I … ide association studies in SLE. Recent work also indicates that several regions within the extended major histocompatibility complex contribute independently to SLE risk. Evidence of additive statistical interaction has been found between IRF5 and TYK2, IRF5, and STAT4, and between NAT2 and exposure to tobacco smoke. SUMMARY: Many new genes have been associated with SLE suscepti …

Genetics and geoepidemiology of primary biliary cirrhosis: following the footprints to disease etiology.

The etiopathogenesis of primary biliary cirrhosis (PBC), an autoimmune disease characterized by the progressive destruction of intrahepatic bile ducts, is still enigmatic. There is growing evidence that suggests that the disease results from a combination of genetic and environmental factors. PBC should be regarded as a peculiar, yet representative autoimmune disease. For example, the relative risk for siblings and the concordance rate in monozygotic twins in PBC are among the highest reported in autoimmune conditions. Association studies for specific genes have provided weak associations, whi

Neuroinflammation in the rat--CNS cells and their role in the regulation of immune reactions.

Recent discoveries suggest that the resident cells of the central nervous system (CNS) the nerve cells and glia, play a more immunologically active role than was previously assumed. Neuroglial communication is of central interest in virtually all types of pathological conditions that affect the brain and several features of the activation that results from nerve cell damage resemble the type of innate immune reactions that occur in other parts of the body In particular, the characteristics of the activation of these CNS cells will affect both the interaction with cells of the immune system as

Innate resistance to Listeria monocytogenes in tumor-bearing mice.

We have previously described the isolation, cloning, and characterization of a tumorigenic murine fibrosarcoma, designated JBS. Growth of JBS tumors in syngeneic mice initiates an anti-tumor immune response that initially manifests as progressive splenic hyperplasia and an increased proliferative ability in cultured splenocytes. In animals with tumors progressing beyond the 2 cm stage there is a reduction in spleen size and a gradual decrease in splenocyte proliferative abilities, leading to anergy at heavy tumor burdens (>3.5 cm). During the phase of immune hyperresponsiveness in tumor-bearin

Rationale for immunotherapy in multiple sclerosis.

The presumed but unspecified immune-mediated basis for the pathogenesis of multiple sclerosis (MS) has led to therapeutic attempts to modify the immune system in general and in selective ways in patients with MS. In general, antiviral, anti-inflammatory, immunosuppressant, and immunomodulatory therapies have been considered. More specifically, these treatments have involved the use of glucocorticoids; immunosuppressant drugs and physical agents such as irradiation; modifications of the immune environment with therapeutic plasma exchange and intravenous immunoglobulin; and more recently, altera

The liver as a cytokine factory working on mRNA blueprints for cancer immunotherapy.

mRNA has the potential to encode both vaccines and immunomodulatory proteins for cancer immunotherapy. In this issue, Beck et al. report on lipopolyplexed mRNAs encoding albumin-stabilized interleukin-2 to transduce liver cells. These mRNAs attain antitumor efficacy on subcutaneous mouse tumors even if malignant cells lack major histocompatibility complex class I (MHC class I) expression.

T- and B-lymphocyte-independent formation of alveolar macrophage-derived multinucleated giant cells in murine Pneumocystis carinii pneumonia.

Multinucleated giant cells developed in Pneumocystis carinii-diseased gene disruption mutant mice deficient in major histocompatibility complex class II molecules, T-cell receptor alpha beta cells, or all mature T and B lymphocytes. These findings demonstrate lymphocyte-independent fusion of alveolar macrophages under morbid conditions. Pulmonary parasite burden seems to be a decisive factor in multinucleation.

Regulation of cathepsin E expression during human B cell differentiation in vitro.

Cathepsin E is an aspartic proteinase which has been implicated in antigen processing in the class II major histocompatibility complex pathway. In this study we show that cathepsin E, measured at both the protein and message level, is up-regulated late in human B cell activation. The implications of this observation in terms of cathepsin E function are discussed.

Autoimmunity--towards the year 2001.

The interactive roles of T cells, major histocompatibility complex (MHC) class I and II molecules and antigen-presenting cells in the generation of autoimmunity is the subject of much discussion. A recent symposium contributed to the debate by inviting experts in several fields of immunology to answer specific questions relating to the mechanisms that trigger autoimmunity.

Antibodies against MICA antigens and kidney-transplant rejection.

BACKGROUND: Good HLA-A, HLA-B, and HLA-DR matches do not guarantee rejection-free renal transplantation. Some kidney transplants fail despite such matches, suggesting that other antigens might be targets for rejection. Major-histocompatibility-complex (MHC) class I-related chain A (MICA) antigens are polymorphic and can elicit antibody production. We sought to determine whether an immune response to MICA antigens might play a role in the failure of kidney allografts. METHODS: Pretransplantation serum samples from 1910 recipients of kidney transplants from deceased donors were tested for anti-

The β-Carboline Harmine Has a Protective Immunomodulatory Role in Nonhealing Cutaneous Leishmaniasis.

Cutaneous leishmaniasis affects 1 million people worldwide annually. Although conventional treatments primarily target the parasite, there is growing interest in host immune modulation. In this study, we investigated the impact of synthetic β-carboline harmine (ACB1801), previously shown to be immunoregulatory in cancer, on the pathology caused by a drug-resistant Leishmania major strain causing persistent cutaneous lesions. Exposure to ACB1801 in vitro had a modest impact on parasite burden within host macrophages. Moreover, it significantly increased major histocompatibility complex II and c…
