Spatiotemporal dynamics of HIV in the Brain: perspective of mathematical modeling for its control via antiretroviral therapy.

The brain, as a reservoir of human immunodeficiency virus (HIV), has received tremendous attention due to the association with the brain's infection with HIV-associated neurocognitive disorders (HAND). Along with the blood-brain barrier (BBB), heterogeneity across the various regions inside the brain makes HIV infection particularly complex to identify the ideal treatment for controlling HIV in the brain. In this study, we developed a mathematical model to describe the spatiotemporal dynamics of HIV infection in three essential regions of the brain: the prefrontal cortex (PF), the choroid plex

Dynamical analysis of a general delayed HBV infection model with capsids and adaptive immune response in presence of exposed infected hepatocytes.

The aim of this paper is to develop and investigate a novel mathematical model of the dynamical behaviors of chronic hepatitis B virus infection. The model includes exposed infected hepatocytes, intracellular HBV DNA-containing capsids, uses a general incidence function for viral infection covering a variety of special cases available in the literature, and describes the interaction of cytotoxic T lymphocytes that kill the infected hepatocytes and the magnitude of B-cells that send antibody immune defense to neutralize free virions. Further, one time delay is incorporated to account for actual

Vector-borne disease outbreak control via instant releases.

This paper is devoted to the study of optimal release strategies to control vector-borne diseases, such as dengue, Zika, chikungunya and malaria. Two techniques are considered: the sterile insect one (SIT), which consists in releasing sterilized males among wild vectors in order to perturb their reproduction, and the Wolbachia one (presently used mainly for mosquitoes), which consists in releasing vectors, that are infected with a bacterium limiting their vectorial capacity, in order to replace the wild population by one with reduced vectorial capacity. In each case, the time dynamics of the v

Game-Theoretical Model of Retroactive Hepatitis B Vaccination in China.

Hepatitis B (HepB) is one of the most common infectious diseases affecting over two billion people worldwide. About one third of all HepB cases are in China. In recent years, China made significant efforts to implement a nationwide HepB vaccination program and reduced the number of unvaccinated infants from 30 to 10%. However, many individuals still remain unprotected, particularly those born before 2003. Consequently, a catch-up retroactive vaccination is an important and potentially cost-effective way to reduce HepB prevalence. In this paper, we analyze a game theoretical model of HepB dynam

The effect of demographic and environmental variability on disease outbreak for a dengue model with a seasonally varying vector population.

Seasonal changes in temperature, humidity, and rainfall affect vector survival and emergence of mosquitoes and thus impact the dynamics of vector-borne disease outbreaks. Recent studies of deterministic and stochastic epidemic models with periodic environments have shown that the average basic reproduction number is not sufficient to predict an outbreak. We extend these studies to time-nonhomogeneous stochastic dengue models with demographic variability wherein the adult vectors emerge from the larval stage vary periodically. The combined effects of variability and periodicity provide a better

Developing, mature, and unique functions of the child's brain in reading and mathematics.

Cognitive development research shows that children use basic "child-unique" strategies for reading and mathematics. This suggests that children's neural processes will differ qualitatively from those of adults during this developmental period. The goals of the current study were to 1) establish whether a within-subjects neural dissociation between reading and mathematics exists in early childhood as it does in adulthood, and 2) use a novel, developmental intersubject correlation method to test for "child-unique", developing, and adult-like patterns of neural activation within those networks. A

Vector Preference Annihilates Backward Bifurcation and Reduces Endemicity.

We propose and analyze a mathematical model of a vector-borne disease that includes vector feeding preference for carrier hosts and intrinsic incubation in hosts. Analysis of the model reveals the following novel results. We show theoretically and numerically that vector feeding preference for carrier hosts plays an important role for the existence of both the endemic equilibria and backward bifurcation when the basic reproduction number [Formula: see text] is less than one. Moreover, by increasing the vector feeding preference value, backward bifurcation is eliminated and endemic equilibria f

Are self-reported telemonitored blood pressure readings affected by end-digit preference: a prospective cohort study in Scotland.

OBJECTIVE: Simple forms of blood pressure (BP) telemonitoring require patients to text readings to central servers creating an opportunity for both entry error and manipulation. We wished to determine if there was an apparent preference for particular end digits and entries which were just below target BPs which might suggest evidence of data manipulation. DESIGN: Prospective cohort study SETTING: 37 socioeconomically diverse primary care practices from South East Scotland. PARTICIPANTS: Patients were recruited with hypertension to a telemonitoring service in which patients submitted home BP

A clinically parameterized mathematical model of Shigella immunity to inform vaccine design.

We refine and clinically parameterize a mathematical model of the humoral immune response against Shigella, a diarrheal bacteria that infects 80-165 million people and kills an estimated 600,000 people worldwide each year. Using Latin hypercube sampling and Monte Carlo simulations for parameter estimation, we fit our model to human immune data from two Shigella EcSf2a-2 vaccine trials and a rechallenge study in which antibody and B-cell responses against Shigella's lipopolysaccharide (LPS) and O-membrane proteins (OMP) were recorded. The clinically grounded model is used to mathematically inve

Assessment of Bone Mineral Density at the Distal Femur and the Proximal Tibia by Dual-Energy X-ray Absorptiometry in Individuals With Spinal Cord Injury: Precision of Protocol and Relation to Injury Duration.

Spinal cord injury (SCI) is characterized by marked bone loss at the knee, and there is a need for established dual-energy X-ray absorptiometry (DXA) protocols to examine bone mineral density (BMD) at this location to track therapeutic progress and to monitor fracture risk. The purpose of this study was to quantify the precision and reliability of a DXA protocol for BMD assessment at the distal femur and the proximal tibia in individuals with SCI. The protocol was subsequently used to investigate the relationship between BMD and duration of SCI. Nine individuals with complete SCI and 9 able-bo

KIAA0319 and ROBO1: evidence on association with reading and pleiotropic effects on language and mathematics abilities in developmental dyslexia.

Substantial heritability has been reported for developmental dyslexia (DD), and KIAA0319 and ROBO1 appear as more than plausible candidate susceptibility genes for this developmental disorder. Converging evidence indicates that developmental difficulties in oral language and mathematics can predate or co-occur with DD, and substantial genetic correlations have been found between these abilities and reading traits. In this study, we explored the role of eight single-nucleotide polymorphisms spanning within KIAA0319 and ROBO1 genes, and DD as a dichotomic trait, related neuropsychological phenot

Partial Residual Plots as an Integrated Model Diagnostic Tool in Model-Based Meta-Analysis.

The use of partial residual plots (PRPs) was explored as a model diagnostic tool in Model-based Meta-Analysis (MBMA). Mathematical derivations illustrating the concepts were followed by an MBMA example using publicly available literature data of anti-depressive treatments with fluoxetine and venlafaxine. An Emax dose-response model was identified for venlafaxine while a constant drug effect combining all dose levels vs. placebo was identified for fluoxetine. The larger the mean baseline Hamilton Depression Rating (HAMD) score, the larger the expected drug effect (P = 0.0122), based on the like

A paradigm shift in retinal detachment repair: The concept of integrity.

The management of rhegmatogenous retinal detachment has rapidly evolved over recent decades. A range of surgical techniques exist, all of which can achieve retinal reattachment in most cases. In recent years there have also been vast technical advances in retinal imaging that have introduced novel ways of visualizing and studying the retinal macro and microstructural anatomy following retinal detachment repair. Recent clinical trial data demonstrates that functional and patient-reported outcomes of retinal reattachment differ with surgical technique, accompanied by differences in anatomic biom

Venous return and the physical connection between distribution of segmental pressures and volumes.

More than sixty years ago, Guyton and coworkers related their observations of venous return to a mathematical model. Showing steady-state flow (F) as proportional to the difference between mean systemic pressure (Pms) and right atrial pressure (Pra), the model fit their data. The parameter defined by the ratio (Pms - Pra)/F, first called an "impedance," came to be called the "resistance to venous return." The interpretation that Pra opposes Pms and that, to increase output, the heart must act to reduce back pressure at the right atrium was widely accepted. Today, the perceived importance of Pm

Cardinal points and generalizations.

In the presence of astigmatism a focal point typically becomes the well-known interval of Sturm with its pair of axially-separated orthogonal line singularities. The same is true of nodal points except that the issues are more complicated: a nodal point may become a nodal interval with a pair of nodal line singularities, but they are not generally orthogonal, and it is possible for there to be only one line singularity or even none at all. The effect of astigmatism on principal points is the motivation behind this paper. The three classes of cardinal points are defined in the literature in a d

Exploring the effective strategies for allocating limited resources to minimize cholera outbreaks.

Despite the near-complete elimination of cholera in many developing countries, numerous lower-income countries continue to face recurring epidemics. Although extensive research has been conducted on cholera transmission dynamics and control, a comprehensive approach to managing outbreaks in resource-limited settings remains elusive. This study introduces a cholera epidemic model incorporating a resource allocation strategy to balance efforts between reducing transmission and enhancing recovery rates. The model is validated using weekly cholera data from the resurgence in Haiti (October 2022-Ma

Modeling the effects of a Shock-and-Kill Treatment for HIV: Latency-Reversing Agents and Natural Killer Cells.

Despite the substantial success of combination antiretroviral therapy (ART) in suppressing HIV replication, achieving a complete cure remains challenging due to the persistence of viral reservoirs. The use of latency-reversing agents (LRAs) combined with natural killer (NK) cells in a "shock-and-kill" strategy has been experimentally confirmed as an effective approach to reducing reservoirs. Here, we utilized an HIV infection mathematical model that incorporates both 'virus-cell' and 'cell-cell' infection modes to assess the dynamic synergy of ART, LRAs, and NK cells. Model calibration was per

Mathematical Assessment of the Roles of Vaccination and Pap Screening on the Burden of HPV and Related Cancers in Korea.

This study is based on using a novel sex-structured mathematical model to assess the effectiveness of vaccination and Pap screening against HPV and related cancers in South Korea. In addition to its disease-free equilibrium (DFE) being locally-asymptotically stable when the associated control reproduction number is less than one, the model could have one or three endemic equilibria, for a special case with negligible disease-induced mortality, if the reproduction number exceeds one. It's shown, using a Krasnoselskii sublinearity argument, that this special case has a unique and locally-asympto

Hemodynamics-driven mathematical model of first and second heart sound generation.

To the best of our knowledge, this is the first hemodynamic-based heart sound generation model embedded in a complete real-time computational model of the cardiovascular system. Simulated heart sounds are similar to experimental and clinical measurements, both quantitatively and qualitatively. Our model can be used to investigate the relationships between heart sound acoustic features and hemodynamic factors/anatomical parameters.
