RNA Interference Restricts Rift Valley Fever Virus in Multiple Insect Systems.

The emerging bunyavirus Rift Valley fever virus (RVFV) is transmitted to humans and livestock by a large number of mosquito species. RNA interference (RNAi) has been characterized as an important innate immune defense mechanism used by mosquitoes to limit replication of positive-sense RNA flaviviruses and togaviruses; however, little is known about its role against negative-strand RNA viruses such as RVFV. We show that virus-specific small RNAs are produced in infected mosquito cells, in Drosophila melanogaster cells, and, most importantly, also in RVFV vector mosquitoes. By addressing the pro

Immune activation by life-shortening Wolbachia and reduced filarial competence in mosquitoes.

Wolbachia strain wMelPop reduces the longevity of its Drosophila melanogaster host and, when introduced into the mosquito Aedes aegypti, halves its life span. We show that wMelPop induces up-regulation of the mosquito's innate immune system and that its presence inhibits the development of filarial nematodes in the mosquito. These data suggest that wMelPop could be used in the global effort to eliminate lymphatic filariasis and possibly for the control of other mosquito-borne parasites where immune preactivation inhibits their development. The cost of constitutive immune up-regulation may cont

Chromobacterium Csp_P reduces malaria and dengue infection in vector mosquitoes and has entomopathogenic and in vitro anti-pathogen activities.

Plasmodium and dengue virus, the causative agents of the two most devastating vector-borne diseases, malaria and dengue, are transmitted by the two most important mosquito vectors, Anopheles gambiae and Aedes aegypti, respectively. Insect-bacteria associations have been shown to influence vector competence for human pathogens through multi-faceted actions that include the elicitation of the insect immune system, pathogen sequestration by microbes, and bacteria-produced anti-pathogenic factors. These influences make the mosquito microbiota highly interesting from a disease control perspective.

Genetic selection of a Plasmodium-refractory strain of the malaria vector Anopheles gambiae.

The anopheline mosquito is the target in most malaria control programs, primarily through the use of residual insecticides. A mosquito was studied that is refractory to most species of malaria through a genetically controlled mechanism. A strain of Anopheles gambiae, which was selected for complete refractoriness to the simian malaria parasite Plasmodium cynomolgi, also has varying degrees of refractoriness to most other malaria species examined, including the human parasites P. falciparum, P. ovale, and P. vivax for which this mosquito is the principal African vector. Furthermore, the refract

Ecological niche modeling of mosquito vectors of West Nile virus in St. John's County, Florida, USA.

BACKGROUND: The lack of available vaccines and consistent sporadic transmission of WNV justify the need for mosquito vector control and prediction of their geographic distribution. However, the distribution of WNV transmission is dependent on the mosquito vector and the ecological requirements, which vary from one place to another. METHODS: Presence/density data of two WNV mosquito vectors, Culex nigripalpus and Cx. quinquefasciatus, was extracted within 5 km buffer zones around seropositive records of sentinel chickens in order to delineate their predicting variables and model the habitat su

Backward bifurcation and optimal control in transmission dynamics of west nile virus.

The paper considers a deterministic model for the transmission dynamics of West Nile virus (WNV) in the mosquito-bird-human zoonotic cycle. The model, which incorporates density-dependent contact rates between the mosquito population and the hosts (birds and humans), is rigorously analyzed using dynamical systems techniques and theories. These analyses reveal the existence of the phenomenon of backward bifurcation (where the stable disease-free equilibrium of the model co-exists with a stable endemic equilibrium when the reproduction number of the disease is less than unity) in WNV transmissio

Anthropogenic changes and associated impacts on vector-borne diseases.

Urbanization impacts the community composition, abundance, and richness of mosquitoes. As urbanization processes increase globally, it is important to better understand the biodiversity loss caused by anthropogenic changes and associated impacts on vector-borne diseases. Mosquito surveillance and control are key for reducing the risk of mosquito-borne pathogen transmission.

Novel Sterile Insect Technology Program Results in Suppression of a Field Mosquito Population and Subsequently to Reduced Incidence of Dengue.

BACKGROUND: There is a steady rise in the global incidence of Aedes-borne arbovirus disease. It has become urgent to develop alternative solutions for mosquito vector control. We developed a new method of sterilization of male mosquitoes with the goal to suppress a local Aedes aegypti population and to prevent the spread of dengue. METHODS: Sterile male mosquitoes were produced from a locally acquired Ae. aegypti colony by using a treatment that includes double-stranded RNA and thiotepa. A field study was conducted with sterile mosquito releases being performed on a weekly basis in predefined

Use of the VectorTest for Advanced Warning of Human West Nile Virus Cases in Mississippi.

West Nile virus (WNV) continues to persist in Mississippi; 2012 was the worse year for human infections, with a total of 247 reported human cases and five deaths. Public health officials are keenly interested in ways to detect WNV in advance in their jurisdictions, so they can implement appropriate and timely mosquito control in affected areas. A total of 40,312 female Culex quinquefasciatus mosquitoes were collected by gravid traps in Mississippi in 2013 and 2014 and tested by VectorTest, a rapid immunochromatographic assay (“dip-stick” test) that is a highly specific and effective rapid thre…

Brugia malayi and Brugia pahangi: transmission blocking activity of ivermectin and brugian filarial infections in Aedes aegypti.

Brugia malayi- or Brugia pahangi-infected, microfilaremic jirds (Meriones unguiculatus) were treated with ivermectin at a single dose of 200 micrograms/kg body weight, administered subcutaneously. After different time intervals, Aedes aegypti mosquitoes were fed on treated or untreated jirds. Sausage stage, L2, and L3 larvae failed to develop in mosquitoes that fed on jirds from 15 to 30 days post-treatment. After 1 month, the numbers of L3 larvae recovered from mosquitoes fed on treated B. pahangi jirds were comparable to controls. However, the number of L3's recovered from mosquitoes fed on

A scoping review: Community health workers engagement in mosquito borne disease prevention activities.

BACKGROUND: Community health workers (CHWs) play increasingly important roles in vector-borne disease prevention globally, yet their contributions remain underutilized and poorly documented in the literature. Understanding how CHWs are deployed across the prevention continuum and identifying barriers to their effectiveness is essential for strengthening mosquito-borne disease control programs. OBJECTIVES: (1) describe the scope and characteristics of studies engaging Community Health Workers (CHWs) in vector-borne disease prevention through community-based strategies; (2) organize the finding

Climatic, Socioecological and Environmental Determinants of Aedes spp. Dynamics at the Community Interface: A Systematic Review With Reflections From a One Health Perspective.

BACKGROUND AND OBJECTIVES: Mosquitoes of the genus Aedes, including but not limited to Aedes aegypti , are major vectors of arboviruses such as dengue, Zika and other related diseases. Their global expansion is driven by climate change, globalisation, urbanisation and human mobility. Understanding how these factors shape their reproduction, survival and persistence is crucial for strengthening integrated vector control within a One Health framework. This review synthesises current evidence on these determinants to support more sustainable control strategies. METHODS: A systematic review was c

Chlorfenapyr bednets effectively overcome pyrethroid resistance escalation in highly resistant Anopheles malaria vectors in Uganda.

Escalating insecticide resistance threatens the efficacy of LLINs, undermining malaria control in Africa. We conducted the first experimental hut trials in Uganda using highly resistant free-flying wild Anopheles mosquitoes and F2 hybrids of FANG and Uganda An. funestus to evaluate the performance of bednets. The interceptor G2 (chlorfenapyr) bednet demonstrated superior efficacy compared to Interceptor (pyrethroid-only) net [mortality odds ratio (OR): 18.7 (8.05-48.6) P < 0.0001], achieving an overall mortality rate of 70.6% and 63.2% against An. funestus and An. gambiae respectively. Modelling the temporal dynamics of Microsporidia MB prevalence in Anopheles mosquitoes under environmental variability.

Releasing Microsporidia MB-positive mosquitoes is being investigated as a promising component of Integrated Vector Management (IVM) strategies for malaria control. In this study, we developed a System Dynamics model to replicate the observed interactions between humans, mosquitoes, the Plasmodium parasite, and the symbiont MB. The model accounts for the impact of environmental variables - such as humidity, rainfall, and temperature - on mosquitoes, Plasmodium, and MB, using exogenous inputs derived from daily climatic observations (rainfall, relative humidity, and temperature) recorded during

Surveillance and control efficacy of the Bergerac, France, 2025 chikungunya outbreak.

The spread of the highly invasive mosquito, Aedes albopictus, across Europe, combined with climate change and human travel and trade, has led to new epidemic threats from mosquito-borne viruses, most significantly dengue and chikungunya, which are increasing in frequency and magnitude. In 2025, mainland France has seen a record number of autochthonous cases and outbreaks of chikungunya, spread across multiple locations, primarily introduced by travellers from the French Overseas Territory of La Réunion which is experiencing severe chikungunya outbreaks. Here, we describe one of the largest Fre

The BiteBarrier transfluthrin emanator demonstrates significant protection against susceptible and resistant malaria and arbovirus vectors in semi-field trials in Tanzania.

Controlling mosquito-borne diseases is becoming increasingly challenging due to factors such as insecticide resistance and shifts in mosquito behavior. The increasing proportion of early evening, morning, and outdoor biting reduces the effectiveness of core interventions like bed nets, which mainly protect people while sleeping indoors. In response, spatial emanators that release volatile active ingredients into the surrounding air to reduce human-vector contact offer a scalable, complementary strategy. This study evaluated the impact of BiteBarrier, a transfluthrin-based spatial emanator, ove

Mobile app-based malaria surveillance and vector control integration: a scoping review of evidence from endemic settings.

BACKGROUND: Effective malaria surveillance is critical for timely and targeted vector control, especially in endemic settings approaching elimination. In recent years, mHealth technologies have emerged as promising tools to improve case detection, reporting efficiency, and spatial targeting of interventions. However, the extent to which mobile-app-based surveillance systems are integrated with vector control strategies remains unclear. This scoping review aimed to synthesize existing evidence on the implementation of mobile-based malaria surveillance platforms and assess their role in supporti

[Actions in the Valencian Autonomous Region against the tiger mosquito and arboviruses in the period 2016-2021].

OBJECTIVE: In recent years, arboviruses have become increasingly important in Europe due to autochthonous outbreaks of emerging and re-emerging diseases such as chikungunya fever, dengue and Zika. These viruses represent a threat in temperate areas such as the Valencian Community (CV), where the presence of their vector, the Asian tiger mosquito, is widely distributed. The objective of this work was to report on the actions carried out within the CV Programme, in the management of declared cases of imported arboviruses from 2016 to 2021. METHODS: To this end, coordinated work procedures had b

Architectural Design Features in High Density Urban Residential Estates and Their Role in Mosquito Incidence in Singapore.

Existing research has established building age as a dominant urban factor influencing mosquito incidence in Singapore, which presents a problem for vector-borne diseases like Dengue. However, in-depth research understanding this relationship has yet to be conducted. This paper explores the correlation between detailed building design features and female Aedes Aegypti mosquito counts, applied to a novel dataset of 400 high-rise residential buildings representing a nationally representative sample. Based on linear regression results, a range of specific building features was found to be statisti
