Amine modulation of glutamate responses from pyloric motor neurons in lobster stomatogastric ganglion.

The amines dopamine (DA), serotonin (5-HT), and octopamine (Oct) each elicit a distinctive motor pattern from a quiescent pyloric network in the lobster stomatogastric ganglion (STG). We previously have demonstrated that these amines alter the synaptic strength at multiple, distributed sites within the pyloric network that could contribute to the amine-induced motor patterns. Here, we examined the postsynaptic contribution to these changes in synaptic strength by determining how the amines modify responses of pyloric motor neurons to glutamate (Glu), one of the network transmitters, applied io…

Amyotrophic lateral sclerosis cerebrospinal fluid is not toxic to cultured spinal motor neurons.

We have studied an effect of cerebrospinal fluid (CSF) from patients with amyotrophic lateral sclerosis (ALS) on explanted and dissociated ventral spinal cord cultures from 13-day-old rat embryos. CSF samples were obtained from 10 ALS patients, 10 other neurological patients, and 10 non-neurological patients. CSF was added at dilution of 10, 25, and 50%. Neurite length and neuronal survival ratio were not significantly different among these three groups. ALS CSF does not contain a neurotoxic factor on the cultured spinal motor neurons.

Changes in sizes of cortical and lower motor neurons in amyotrophic lateral sclerosis.

It has been suggested that the degeneration of lower motor neurons in amyotrophic lateral sclerosis (ALS) is a transneuronal event, secondary to the loss of corticospinal and corticobulbar neurons. In an attempt to test this hypothesis, the cross-sectional areas of pyramidal cells in layer 5 of the foot and tongue areas of the precentral gyri were measured in 12 cases of the classical sporadic form of ALS, and in 10 control subjects. The areas of motor neurons in the hypoglossal nuclei and in the ventral horns of segment L4 of the spinal cord were also measured. The number of neurons per 20 mi…

Targeting Motor End Plates for Delivery of Adenoviruses: An Approach to Maximize Uptake and Transduction of Spinal Cord Motor Neurons.

Gene therapy can take advantage of the skeletal muscles/motor neurons anatomical relationship to restrict gene expression to the spinal cord ventral horn. Furthermore, recombinant adenoviruses are attractive viral-vectors as they permit spatial and temporal modulation of transgene expression. In the literature, however, several inconsistencies exist with regard to the intramuscular delivery parameters of adenoviruses. The present study is an evaluation of the optimal injection sites on skeletal muscle, time course of expression and mice's age for maximum transgene expression in motor neurons. …

Abnormal glycosylation of motor neurons with N-acetyl-D-galactosamine in a case of subacute motor neuronopathy associated with lymphoma.

Lectins, glycine max (SBA) and Vicia villosa (VVA), which identify terminal N-acetyl-D-galactosamine (GalNAc), stained the motor neurons of a patient with subacute motor neuronopathy associated with neoplastic angioendotheliosis. In normal controls and in patients with other neurological disorders including amyotrophic lateral sclerosis, neither lectin stained motor neurons. Abnormal glycosylation of motor neurons with terminal galNAc may be associated with the pathogenesis of subacute motor neuronopathy in this patient.

Molecular signatures of selective motor neuron vulnerability in old age.

Motor neuron (MN) death and impairment of fast-fatigable motor units is widely recognized in elderly humans and rats. However, the molecular signatures identifying vulnerable MNs remains underexplored. In murine models of amyotrophic lateral sclerosis, matrix metalloproteinase 9 (MMP-9) and chondrolectin (Chodl) decline with disease progression and have been considered molecular markers of fast-fatigable MNs. However, their expression in brainstem and lumbar MNs, in accordance with the 'size principle' (in which motor units are recruited in a size-dependent manner, with MNs of lower capacitanc

Assisted ventilation withdrawal in motor neuron disease: updated results.

INTRODUCTION: Patients with ventilator-dependent motor neuron disease (MND) may request withdrawal of their assisted ventilation. Facilitating this process as a healthcare professional (HCP) can be emotionally and practically challenging. The Association for Palliative Medicine (APM) issued guidance to support HCPs and invited anonymised accounts of the withdrawal process to provide an update on the guidance. METHODS: HCPs submitted anonymised accounts via email. Quantitative data was analysed descriptively in Excel. Free-text comments were analysed thematically using an inductive, iterative

Protocol for the Development of The Australian Motor Neurone Disease (MND) Guideline.

INTRODUCTION: Motor neurone disease (MND) is a rare but devastating neurodegenerative condition that results in progressive muscle weakness, disability, and eventual respiratory failure. In Australia, the impact of MND is significant, and there is a pressing need for a high-quality, locally relevant guideline to support equitable, person-centred care. QUESTIONS: This guideline aims to address key clinical and care-related questions that arise across the MND trajectory. It will identify the most pressing topics and uncertainties in MND management through interest holder consultation and struct

Predictors of mortality post-gastrostomy in motor neuron disease patients.

INTRODUCTION/AIMS: Motor neuron disease (MND) is a progressive neurodegenerative condition with a limited life expectancy. There is very little data on mortality and its associated factors beyond 30 days following gastrostomy. We explored the demographic, clinical, and nutritional predictors for early mortality at 30, 90, and 180 days following gastrostomy in these patients. METHODS: This was a retrospective study involving 94 MND patients in Western Australia who underwent gastrostomy between 2015 and 2021. Patients were divided into two groups based on mortality at 30, 90, and 180 days post

Motor Cortical Neuronal Hyperexcitability Associated with α-Synuclein Aggregation.

Dysfunction of the cerebral cortex is thought to underlie motor and cognitive impairments in Parkinson disease (PD). While cortical function is known to be suppressed by abnormal basal ganglia output following dopaminergic degeneration, it remains to be determined how the deposition of Lewy pathology disrupts cortical circuit integrity and function. Moreover, it is also unknown whether cortical Lewy pathology and midbrain dopaminergic degeneration interact to disrupt cortical function in late-stage. To begin to address these questions, we injected α-synuclein (αSyn) preformed fibrils (PFFs) in

The various forms of hereditary motor neuron disorders and their historical descriptions.

Motor neuron disorders comprise a clinically and pathologically heterogeneous group of neurologic diseases characterized by progressive degeneration of motor neurons (including both sporadic and hereditary diseases), affecting the upper motor neurons, lower motor neurons, or both. Hereditary motor neuron disorders themselves represent a vast and heterogeneous group, with numerous clinical and genetic overlaps that can be a source of error. This narrative review aims at providing an overview of the main types of inherited motor neuron disorders by recounting the stages in their historical descr…

Diving deep: zebrafish models in motor neuron degeneration research.

In the dynamic landscape of biomedical science, the pursuit of effective treatments for motor neuron disorders like hereditary spastic paraplegia (HSP), amyotrophic lateral sclerosis (ALS), and spinal muscular atrophy (SMA) remains a key priority. Central to this endeavor is the development of robust animal models, with the zebrafish emerging as a prime candidate. Exhibiting embryonic transparency, a swift life cycle, and significant genetic and neuroanatomical congruencies with humans, zebrafish offer substantial potential for research. Despite the difference in locomotion-zebrafish undulate

[Electromyography in atypical variants of motor neuron disease: a case series].

BACKGROUND: Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disease that affects both the upper and lower motor neurons, it has a heterogeneous clinical presentation, there are atypical variants that differ from the classic form of the disease. The criteria for diagnosis have evolved over time, with the support of electromyography (EMG), we present a patient series with these variants in which EMG was crucial to make the diagnosis. CLINICAL CASES: Six cases are described with atypical presentation of motor neuron disease, for the isolated bulbar ALS phenotype, three cases are repor…
