Intercellular communication between vascular smooth muscle and endothelial cells mediated by heparin-binding epidermal growth factor-like growth factor and vascular endothelial growth factor.

Heparin-binding epidermal growth factor-like growth factor (HB-EGF), a potent mitogen and migration factor for vascular smooth muscle cells (SMC), promoted neovascularization in vivo in the rabbit cornea. MRI demonstrated quantitatively the angiogenic effect of HB-EGF when introduced subcutaneously into nude mice. HB-EGF is not directly mitogenic to endothelial cells but it induced the migration of bovine endothelial cells and release of endothelial cell mitogenic activity from bovine vascular SMC. This mitogenic activity was specifically blocked by neutralizing anti-vascular endothelial growt

Interactions of nonprostanoid trimetoquinol analogs with thromboxane A2/prostaglandin H2 receptors in human platelets, rat vascular endothelial cells and rat vascular smooth muscle cells.

Trimetoquinol (TMQ), a nonprostanoid compound, inhibits thromboxane A2 agonist-induced responses in platelets and vascular smooth muscle. Sixteen TMQ analogs were used to examine the stereochemical requirements of the interaction with thromboxane A2/prostaglandin H2 (TP) receptor sites in human platelets (HP), and cultured rat vascular endothelial (RVEC) and smooth muscle (RVSMC) cells. [3H]SQ 29548 was used as the ligand for TP receptors. The receptor binding affinities of these TMQ analogs for TP receptors in HP, RVEC and RVSMC were highly correlated with each other, and to their reported in

PCSK9 (Proprotein Convertase Subtilisin/Kexin Type 9) Triggers Vascular Smooth Muscle Cell Senescence and Apoptosis: Implication of Its Direct Role in Degenerative Vascular Disease.

OBJECTIVE: PCSK9 (proprotein convertase subtilisin/kexin type 9) plays a critical role in cholesterol metabolism via the PCSK9-LDLR (low-density lipoprotein receptor) axis in the liver; however, evidence indicates that PCSK9 directly contributes to the pathogenesis of various diseases through mechanisms independent of its LDL-cholesterol regulation. The objective of this study was to determine how PCSK9 directly acts on vascular smooth muscle cells (SMCs), contributing to degenerative vascular disease. Approach and Results: We first examined the effects of PCSK9 on cultured human aortic SMCs.

Berberine suppresses mechanical stretch-induced apoptosis and proliferation in vascular smooth muscle cells via ROS/PDI/Drp1 signaling pathway.

BACKGROUND: Vascular smooth muscle cells (VSMCs) exhibit heterogeneous responses to mechanical stretch stress (SS), with some undergoing apoptosis and others proliferating. The mechanisms underlying this differential fate decision remain unclear. OBJECTIVE: This study investigated whether SS-induced heterogeneity in F-actin expression dictates distinct mitochondrial reactive oxygen species (ROS) responses, leading to different VSMC fates, and evaluated the therapeutic potential of the berberine (BBR). METHODS: VSMCs were subjected to SS. F-actin heterogeneity was characterized, and mitochond

Loss of the Coronary Artery Disease Risk Gene LMOD1 in Vascular Smooth Muscle Cells Triggers Rapid-Onset Coronary Atherosclerosis.

BACKGROUND: Atherosclerosis is the primary underlying cause of coronary artery disease. LMOD1 is a coronary artery disease risk gene whose role in coronary artery pathophysiology is unknown. Whole-body loss of Lmod1 causes a lethal neonatal visceral myopathy in mice, necessitating unique approaches for the study of vascular smooth muscle cell (VSMC) phenotypes. METHODS: Control mice (Lmod1WT carrying a Cre recombinase allele) and VSMC-restricted Lmod1 knockout mice (Lmod1SMKO) were subjected to various atherogenic regimens. Atherosclerosis and LMOD1 (leiomodin 1) expression in mouse and human

Cell-Surface Signatures and Targets of Modulated Vascular Smooth Muscle Cells in Atherosclerosis: From State Identification to Precision Intervention.

Atherosclerotic plaques contain vascular smooth muscle cell (VSMC)-derived populations that no longer fit a simple contractile-versus-synthetic model. Lineage tracing, single-cell transcriptomics, spatial profiling and multimodal surface-protein measurements now show that VSMC-derived cells occupy heterogeneous lesional states with pathogenic, reversible or plaque-stabilizing properties. This diversity creates a translational bottleneck. Intracellular markers and transcriptomic clusters can define state transitions, but they do not by themselves provide handles for live-cell isolation, molecul

Heme-NO Dilates Arteries via Mobilization of NO Moieties From an Intracellular NO Store Within Vascular Smooth Muscle Cells.

Nitrosyl heme (heme-NO) has recently emerged as a surrogate signaling entity for nitric oxide (NO). However, questions remain about how heme-NO signals across the cell membrane. Herein, we test the hypothesis that extracellular heme-NO signals as a nitrodilator that vasodilates by mobilizing NO moiety from a nitrodilator-activated intracellular NO store (NANOS) in the vasculature. We identify a novel mechanism for glutathione-catalyzed formation of a model compound, alb-heme-NO, and determine glutathione (GSH) as a ligand in its structure. Heme-NO complexes with plasma proteins in blood and, a

Loss of CASQ2 promotes vascular smooth muscle cell phenotypic switching in aortic dissection uncovered by integrated single-cell transcriptomics.

BACKGROUND: Aortic dissection (AD) is a life-threatening cardiovascular disease with limited effective therapeutic options. Phenotypic switching of aortic vascular smooth muscle cells (VSMCs) from a contractile to a pathological state is a critical driver of AD progression. However, the molecular regulators governing this transition remain incompletely understood. This study aimed to identify a critical regulator of VSMC phenotypic switching in AD and to investigate its underlying mechanisms and translational relevance. METHODS: Single-cell RNA sequencing (scRNA-seq) and bulk RNA sequencing d

Multi-omics integration study of vascular smooth muscle cell phenotypic conversion identified novel biomarkers in idiopathic pulmonary arterial hypertension.

BACKGROUND: Idiopathic pulmonary arterial hypertension (IPAH) is a progressive, high‑mortality disease driven in part by maladaptive vascular remodeling. Phenotypic conversion of vascular smooth muscle cells (VSMCs) is implicated in this process, but the cellular heterogeneity and regulatory drivers in IPAH remain incompletely defined. METHODS: We integrated scRNA‑seq datasets from IPAH and control lungs samples with bulk RNA‑seq from IPAH patients, MCT‑induced rat IPAH models, applying diffusion‑map dimensionality reduction, pseudotime inference, CellChat, and machine learning to nominate phe

ALKBH5 as a therapeutic target for hypertension: Inhibiting vascular smooth muscle cell phenotypic switch via the TEAD3/PDZK1 axis.

Hypertension is a chronic disease accompanied by vascular remodeling. As an m6A demethylase, ALKBH5 is implicated in cardiovascular diseases. Herein, we demonstrated that ALKBH5 expression was upregulated in the aortic tissues of spontaneously hypertensive rats (SHR), with predominant localization in vascular smooth muscle cells (VSMCs). To define ALKBH5 function, we generated an SM22α-driven, VSMC-specific Alkbh5-knockdown AAV9 vector and administered a single tail-vein injection (11 × 10¹² v.g./rat) to 4-week-old male SHRs. Alkbh5 knockdown markedly lowered arterial pressure in SHRs, amelior

CircUBE4B attenuates AngII-induced vascular smooth muscle cell proliferation and migration through upregulating GSK3B by sponging miR-199a-5p.

Aortic dissection (AD) is a life-threatening medical condition generally affecting those 65-75 years of age with poor outcome and various complications. It is urgent to investigate the pathogenesis of AD to explore novel diagnostic markers and therapeutic targets for AD. Pathological vascular remodeling due to dysregulated proliferation and migration of vascular smooth muscle cells (VSMCs) would promote AD development. Glycogen synthase kinase 3 beta (GSK3B) could suppress VSMC proliferation and migration. However, the effect of GSK3B on VSMCs and the mechanism regulating GSK3B in AD remains u

Adenylate kinase 4 (AK4) deficiency prevents vascular smooth muscle cell phenotypic switching by regulating mitochondrial dysfunction through AMPKα inactivation.

BACKGROUND AND AIMS: Mitochondrial dynamics are key mechanism regulating the conversion of vascular smooth muscle cells (VSMCs) from a contractile to a synthetic phenotype, which is involved in neointima formation and restenosis. However, the underlying mechanisms leading to mitochondrial dysfunction are not fully understood. METHODS: Western blot was used to detect the expression of relevant molecules at the protein level. CCK-8, EdU assays, and transwell were used to test cell proliferation and migration capacity. Flow cytometry was used to assess cell cycle and ROS. RESULTS: AK4 was upreg

Delineation of a thrombin receptor-stimulated vascular smooth muscle cell transition generating cells in the plaque-stabilizing fibrous cap.

AIMS: Vascular smooth muscle cells (VSMCs) accumulate in atherosclerotic plaques and exhibit remarkable phenotypic plasticity, contributing to both plaque growth and stability. The plaque-stabilizing fibrous cap is rich in VSMC-derived cells, yet the cellular transitions and regulatory mechanisms governing fibrous cap formation remain unclear. Here, we aimed to identify the VSMC phenotypic transitions associated with this critical process. METHODS AND RESULTS: Mapping of lineage-traced VSMCs during plaque development revealed investment of VSMCs prior to fibrous cap formation. Using single-ce

Chlamydia pneumoniae invades into vascular smooth muscle cells through the CXCR4-β-arrestin 2 pathway via TLR2/CXCR4 crosstalk.

Vascular smooth muscle cell (VSMC) dysfunction plays an important role in the pathogenesis of atherosclerosis. Chlamydia pneumoniae (C. pneumoniae) has been shown to infect and grow in VSMCs, and causes VSMC dysfunction, thereby promoting atherosclerosis development. However, it is an enigma how C. pneumoniae invades into VSMCs. In this study, we explored the specific mechanism of C. pneumoniae invading into VSMCs, and found that TLR2 expression and CXCR4 phosphorylation level were elevated after C. pneumoniae infection, and the translocation of TLR2 and CXCR4 to the membrane were increased at

Vascular smooth muscle cells in response to cholesterol crystals modulates inflammatory cytokines release and promotes neutrophil extracellular trap formation.

BACKGROUND: The formation and accumulation of cholesterol crystals (CC) at the lesion site is a hallmark of atherosclerosis. Although studies have shown the importance of vascular smooth muscle cells (VSMCs) in the disease atherosclerosis, little is known about the molecular mechanism behind the uptake of CC in VSMCs and their role in modulating immune response. METHODS: Human aortic smooth muscle cells were cultured and treated with CC. CC uptake and CC mediated signaling pathway and protein induction were studied using flow cytometry, confocal microscopy, western blot and Olink proteomics.

Coptisine inhibits neointimal hyperplasia through attenuating Pak1/Pak2 signaling in vascular smooth muscle cells without retardation of re-endothelialization.

BACKGROUND AND AIMS: Vascular injury-induced endothelium-denudation and profound vascular smooth muscle cells (VSMCs) proliferation and dis-regulated apoptosis lead to post-angioplasty restenosis. Coptisine (CTS), an isoquinoline alkaloid, has multiple beneficial effects on the cardiovascular system. Recent studies identified it selectively inhibits VSMCs proliferation. However, its effects on neointimal hyperplasia, re-endothelialization, and the underlying mechanisms are still unclear. METHODS: Cell viability was assayed by 3-[4,5-dimethylthiazole-2-yl]-2,5-diphenyltetrazolium bromide (MTT)

S-adenosylmethionine attenuates angiotensin II-induced aortic dissection formation by inhibiting vascular smooth muscle cell phenotypic switch and autophagy.

It is well known that aortic dissection (AD) is a very aggressive class of vascular diseases. S-adenosylmethionine (SAM) is an autophagy inhibitor with anti-inflammatory and anti-oxidative stress effects; however, the role of SAM in AD is unknown. In this study, we constructed an animal model of AD using subcutaneous minipump continuous infusion of AngII-induced ApoE-/-mice and a cytopathic model using AngII-induced primary vascular smooth muscle cells (VSMCs) to investigate the possible role of SAM in AD. The results showed that mice in the AngII + SAM group had significantly lower AD inciden

Identification of Novel Gene Regulatory Networks for Dystrophin Protein in Vascular Smooth Muscle Cells by Single-Nuclear Transcriptome Analysis.

Duchenne muscular dystrophy is an X-linked recessive disease caused by mutations in dystrophin proteins that lead to heart failure and respiratory failure. Dystrophin (DMD) is not only expressed in cardiomyocytes and skeletal muscle cells, but also in vascular smooth muscle cells (VSMCs). Patients with DMD have been reported to have hypotension. Single nuclear RNA sequencing (snRNA-seq) is a state-of-the-art technology capable of identifying niche-specific gene programs of tissue-specific cell subpopulations. To determine whether DMD mutation alters blood pressure, we compared systolic, diasto
