Effects of prolonged exposure to low-dose fumonisin B1 in pigs.

From the point of view of human exposure, fumonisins (FB1, FB2, FB1, FB4), a relatively recently (1988) discovered and identified group of mycotoxins, represent one of the five most important mycotoxin groups causing human disease. In an earlier experiment studying the effects of relatively low doses (10, 20 and 40 p.p.m.) of FB1 in weaned piglets, it was established that the 4-week feeding of 10 p.p.m. (mg/kg feed) FB1 produced mild pulmonary oedema. This suggested the importance of studies with even lower doses of the toxin to determine the tolerable limits. The objective of this experiment …

Cancer initiation by fumonisin B(1) in rat liver--role of cell proliferation.

Fumonisin B(1) (FB(1)), a carcinogenic mycotoxin produced by the fungus Fusarium verticillioides in corn, causes cancer initiation in rat liver in a similar manner to genotoxic carcinogens although apparently with different kinetics. The present experiment was designed to evaluate the role of regenerative cell proliferation, effected by partial hepatectomy (PH) and carbontetrachloride (CCl(4)) and direct mitogen-induced hyperplasia, induced by lead nitrate (PbNO(3)), on FB(1)-induced cancer initiation. Initiation was effected over a period of 14 days by gavage administration of FB(1) at differ

Acute toxicity of penicillic acid and rubratoxin B in dogs.

The effect of intraperitoneally administered penicillic acid, a mycotoxin produced by several species of Penicillium and Aspergillus, on female dogs of mixed breeding was determined by serum tests, by observation of clinical signs and survival times, and by evaluation of gross and microscopic lesions. Combination studies employing penicillic acid and a second mycotoxin, rubratoxin B, also were undertaken. Post mortem examination disclosed hemorrhaging of the serosal surfaces of the abdomen of dogs receiving penicillic acid. The most significant histologic change observed in penicillic-acid-tre

Biocontrol of Fusarium Species Utilizing Indigenous Rooibos and Honeybush Extracts.

Mycotoxins produced by several Fusarium species have a significant effect on reducing maize yield and grain quality and have led to food safety concerns. The antifungal activities of rooibos (Aspalathus linearis) and honeybush (Cyclopia species) tea extracts reduced the growth of plant pathogen Botrytis cinerea, but their efficacy against Fusarium spp. is unknown. In this study, we examined the effects of fermented and unfermented rooibos (A. linearis) and honeybush (Cyclopia subternata) aqueous extracts as well as green tea (Camellia sinensis) against 10 Fusarium species. Conidial viability w…

Application of Graphitized Multi-Walled Carbon Nanotubes Combined with Orbitrap High-Resolution Mass Spectrometry for the Rapid Detection of Ten Toxins in Wild Mushrooms.

Wild mushroom poisoning is an emerging global food safety issue, especially in subtropical regions like southwestern China, where incidents are geographically clustered. Current detection methods are often time-consuming and overlook region-specific toxins. We developed a rapid, sensitive, and accurate method for the simultaneous detection of ten characteristic mushroom toxins prevalent in Guizhou, China. The method combines graphite multi-walled carbon nanotubes (G-MWCNTs) for sample preparation with Orbitrap high-resolution mass spectrometry (HRMS). Wild mushroom samples were extracted via u

Assessing the Risk of Exposure to Aflatoxin B1 through the Consumption of Peanuts among Children Aged 6-59 Months in the Lusaka District, Zambia.

Aflatoxins B1 (AFB1) are fungi-produced toxins found in crops like peanuts, maize, and tree nuts. They constitute a public health concern due to their genotoxic and carcinogenic effects. A deterministic exposure risk assessment to AFB1 through the consumption of peanuts was conducted on children using the Margin of Exposure (MOE) and the liver cancer risk approaches. Data on AFB1 concentrations in peanuts, quantities of peanut consumption, and the weights of the children were obtained from the literature. Generally, MOE values were below the safe margin of 10,000, ranging between 3.68 and 0.14

A Novel Multicellular Placental Barrier Model to Investigate the Effect of Maternal Aflatoxin B1 Exposure on Fetal-Side Neural Stem Cells.

Ingestion of food toxins such as aflatoxin B1 (AFB1) during pregnancy may impair fetal neurodevelopment. However, animal model results may not be accurate due to the species' differences, and testing on humans is ethically impermissible. Here, we developed an in vitro human maternal-fetal multicellular model composed of a human hepatic compartment, a bilayer placental barrier, and a human fetal central nervous system compartment using neural stem cells (NSCs) to investigate the effect of AFB1 on fetal-side NSCs. AFB1 passed through the HepG2 hepatocellular carcinoma cells to mimic the maternal

Sensitive and selective alternariol analysis by a newly developed monoclonal antibody-based immunoassay.

Alternariol is a toxic secondary metabolite produced by Alternaria fungi. Nowadays, this mycotoxin can be found in many products of plant origin at concerning concentrations. The aim of the present study was to develop a highly sensitive and selective immunochemical method for the analysis of alternariol. To this end, hapten synthesis was carried out from the scratch and specific high-affinity monoclonal antibodies to alternariol were generated by using, for the first time, alternariol bioconjugates with unambiguous linker tethering sites. A novel indirect enzyme-linked immunosorbent assay tha

Targeted Sphingolipid Analysis in Heart, Gizzard, and Breast Muscle in Chickens Reveals Possible New Target Organs of Fumonisins.

Alteration of sphingolipid synthesis is a key event in fumonisins toxicity, but only limited data have been reported regarding the effects of fumonisins on the sphingolipidome. Recent studies in chickens found that the changes in sphingolipids in liver, kidney, lung, and brain differed greatly. This study aimed to determine the effects of fumonisins on sphingolipids in heart, gizzard, and breast muscle in chickens fed 20.8 mg FB1 + FB2/kg for 9 days. A significant increase in the sphinganine:sphingosine ratio due to an increase in sphinganine was observed in heart and gizzard. Dihydroceramides

Discovery of the Biosynthetic Pathway of Beticolin 1 Reveals a Novel Non-Heme Iron-Dependent Oxygenase for Anthraquinone Ring Cleavage.

This study used light-mediated comparative transcriptomics to identify the biosynthetic gene cluster of beticolin 1 in Cercospora. It contains an anthraquinone moiety and an unusual halogenated xanthone moiety connected by a bicyclo[3.2.2]nonane. During elucidation of the biosynthetic pathway of beticolin 1, a novel non-heme iron oxygenase BTG13 responsible for anthraquinone ring cleavage was discovered. More importantly, the discovery of non-heme iron oxygenase BTG13 is well supported by experimental evidence: (i) crystal structure and the inductively coupled plasma mass spectrometry revealed

Metarhizium anisopliae enzymes and toxins.

Entomopathogenic fungi are both a feasible system for the control of insect pests in agriculture with a growing market and an important model for studies of host-pathogen interaction. In the last ten years the actual use of fungi, mainly Metarhizium anisopliae and Beauveria bassiana, is increasing reaching commercial scale in Countries like Brazil, China and Mexico among others. At the same time important progress has occurred in the understanding of the molecular aspects of the pathogenesis and in the development of tools to validate putative virulence factors by the construction of over-expr

[Putative virulence factors of Aspergillus species].

Members of the genus Aspergillus are filamentous, ubiquitous fungi found in nature. Aspergillus species can cause clinical settings in man, such as opportunistic infections, allergic states and toxicoses. Immunosuppression is the major factor predisposing to development of opportunistic mycoses. To invade the host, Aspergillus species must be able to adhere to and penetrate the host tissues. Much work has focused on the search for virulence determinants that allow the establishment of the fungus within the host. Adhesins, pigment production, toxic metabolites and extracellular enzymes are thou

The role of ochratoxin A as a possible cause of Balkan endemic nephropathy and its risk evaluation.

This article presents evidences which support the mycotoxic hypothesis of Balkan endemic nephropathy (BEN), examines the significance of human exposure to ochratoxin A (OTA) in BEN-endemic areas, and gives some ideas for future investigations. The morphological and ultrastructural similarity between OTA-induced mycotoxic porcine nephropathy (MPN) and BEN, epidemiological studies, and relationship between the incidence of BEN and the presence of OTA in human blood in contamination levels similar to those in pigs suffering from MPN suggest that OTA could be an etiological factor in BEN.

Immune and bone properties of chicks consuming corn contaminated with a Fusarium that induces dyschondroplasia.

A study was conducted to contrast the dyschondroplasia-inducing capability of several species of Fusarium with that of the natural fungi found in poultry diets and litter. Day-old broiler chicks were fed pure corn cultures of specific fungal isolates for 3 weeks. Humoral immunity to sheep red blood cells (SRBC), body weight, valgus and varus leg deformities, incidence of dyschondroplasia, and mechanical properties of the tibiotarsi were examined. F. equiseti #15 was the only fungal isolate to induce dyschondroplasia to any significant degree, which confirmed previous work in Minnesota. This is

Aflatoxin contamination and detoxification in traditional Chinese medicines.

Aflatoxins are toxic compounds produced by certain molds, primarily Aspergillus flavus and Aspergillus parasiticus. These mycotoxins can contaminate various agricultural products, including grains, nuts, and seeds, posing serious health risks. Similarly, a significant portion of Traditional Chinese Medicines (TCMs) is either cultivated through artificial means or harvested from natural environments, making it susceptible to contamination as well. By addressing these critical aspects, the review aims to enhance the safety and quality of TCMs. Despite extensive research on aflatoxins in food cro…

In Vitro Antimalarial Activity of Trichothecenes against Liver and Blood Stages of Plasmodium Species.

Trichothecenes (TCNs) are a large group of tricyclic sesquiterpenoid mycotoxins that have intriguing structural features and remarkable biological activities. Herein, we focused on three TCNs (anguidine, verrucarin A, and verrucarol) and their ability to target both the blood and liver stages of Plasmodium species, the parasite responsible for malaria. Anguidine and verrucarin A were found to be highly effective against the blood and liver stages of malaria, while verrucarol had no effect at the highest concentration tested. However, these compounds were also found to be cytotoxic and, thus, n…
