Neural mechanisms of the V2M-PrL pathway mediate blue light-induced spatial memory impairment.

Blue light has been shown to impair spatial memory in mice. However, the underlying neural circuit mechanisms of this impairment remain elusive. Our study revealed that 600-lux blue light disrupted spatial memory retrieval in male mice through the secondary visual cortex (V2M) → the prelimbic cortex (PrL) neural pathway. Specifically, viral tracing and multichannel fiber optic recordings demonstrated that glutamatergic V2M neurons (V2MGlu) projected to GABAergic neurons in the PrL (PrLGABA), thereby inhibiting the excitability of glutamatergic PrL neurons (V2MGlu→PrLGABA→Glu). The selective ab

Altered functional connectivity related to prepulse inhibition in functional movement disorder.

BACKGROUND: Functional neurological disorder (FND) is increasingly understood as a multinetwork disorder. Deficits in subcortical sensorimotor gating, indexed by reduced prepulse inhibition (PPI) of the startle reflex, are reported in FND and may reflect impaired filtering and contextual weighting of sensory input. However, the neural correlates linking impaired gating to intrinsic network dysfunction remain unclear. METHODS: We examined the relationship between PPI and resting-state functional connectivity in 38 patients with functional movement disorder (FMD) and 39 healthy controls (HC). P

Pharmacological and Physiological Characteristics of Synaptic Transmissions From the Medial Prefrontal Cortex Onto Noradrenergic Neurons and Inhibitory Interneurons in the Mouse Locus Coeruleus.

The locus coeruleus (LC) is the brain's primary source of norepinephrine. It modulates the brain's overall state of arousal. Recent evidence suggests that the LC also regulates immediate attentional responses by resetting related cortical networks to optimize behavioral outcomes. Cortical regions associated with high cognitive function, such as the medial prefrontal cortex (mPFC), are theorized to influence LC output directly to regulate behavior. However, the available evidence is insufficient for a comprehensive understanding of the underlying mechanisms and properties. To address this gap i

Evidence of widespread brain inhibition following conventional but not individualised non-motor repetitive transcranial magnetic stimulation: A multimodal study.

BACKGROUND: Studies using repetitive transcranial magnetic stimulation (rTMS) frequently target non-motor brain areas to treat various conditions, including pain. However, whether and how non-motor rTMS affects neural activity is still debated in the scientific community. OBJECTIVE: Characterize the local and global inhibitory effects of 2 non-motor rTMS protocols on brain activity. METHODS: Participant's (n = 13) functional magnetic resonance imaging (fMRI) signal associated with a painful electrical paradigm was measured before and after 2 distinct low-frequency rTMS protocols (target: lef

Parkinsonism disrupts the balance between excitatory and inhibitory activity within the primary motor cortex during movement.

Parkinson's disease (PD) has been associated with alterations in neuronal activity in the basal ganglia-thalamocortical (BGTC) network. Previous studies have suggested that cortical disinhibition is a feature of PD, but there has been little direct evidence of the changes in cortical neuronal spiking activity to support this hypothesis. To test the hypothesis that activity in the motor cortex is enhanced in PD, we investigated the effects of parkinsonism on movement-related neuronal activity in the primary motor cortex (M1). Microelectrode arrays were chronically implanted in M1 of two nonhuma

Characterization of optogenetically activated inhibitory inputs onto cholinergic motor neurons in the spinal dorsolateral nucleus.

Inhibitory control of external urethral sphincter motor neurons (EUS-MNs) in the spinal dorsolateral nucleus (DLN), which corresponds to a portion of Onuf's nucleus in humans, is essential for normal micturition by inducing EUS relaxation during voiding; yet synaptic mechanisms remain poorly characterized. Using neonatal mice P8-P12, we developed a slicing technique-cutting spinal cords at 150° from the coronal plane (30° from the horizontal plane in the agarose block), for maximizing EUS-MNs captured per slice. Using transgenic mice co-expressing channelrhodopsin-2 in inhibitory interneurons

Functional networks of inhibitory neurons orchestrate synchrony in the hippocampus.

Inhibitory interneurons are pivotal components of cortical circuits. Beyond providing inhibition, they have been proposed to coordinate the firing of excitatory neurons within cell assemblies. While the roles of specific interneuron subtypes have been extensively studied, their influence on pyramidal cell synchrony in vivo remains elusive. Employing an all-optical approach in mice, we simultaneously recorded hippocampal interneurons and pyramidal cells and probed the network influence of individual interneurons using optogenetics. We demonstrate that CA1 interneurons form a functionally interc

Moderate to severe acute pain disturbs motor cortex intracortical inhibition and facilitation in orthopedic trauma patients: A TMS study.

OBJECTIVE: Primary motor (M1) cortical excitability alterations are involved in the development and maintenance of chronic pain. Less is known about M1-cortical excitability implications in the acute phase of an orthopedic trauma. This study aims to assess acute M1-cortical excitability in patients with an isolated upper limb fracture (IULF) in relation to pain intensity. METHODS: Eighty-four (56 IULF patients <14 days post-trauma and 28 healthy controls). IULF patients were divided into two subgroups according to pain intensity (mild versus moderate to severe pain). Short-term plasticity in glomerular inhibitory circuits shapes olfactory bulb output.

Short-term plasticity is a fundamental synaptic property thought to underlie memory and neural processing. The glomerular microcircuit comprises complex excitatory and inhibitory interactions and transmits olfactory nerve signals to the excitatory output neurons, mitral/tufted cells (M/TCs). The major glomerular inhibitory interneurons, short axon cells (SACs) and periglomerular cells (PGCs), both provide feedforward and feedback inhibition to M/TCs and have reciprocal inhibitory synapses between each other. Olfactory input is episodically driven by sniffing. We hypothesized that frequency-dep

Our results provide direct evidence implicating that the balance between neural inhibition and excitation within sensory regions resolves perceptual competition. tDCS to premotor cortex changes action verb understanding: Complementary effects of inhibitory and excitatory stimulation.

Processing the meaning of action language correlates with somatotopic activity in premotor cortex (PMC). A previous neurostimulation study supported a causal contribution of PMC activity to action verb understanding, but the direction of the effect was unexpected: inhibiting PMC made participants respond faster to action verbs. Here we investigated the effects of PMC excitation and inhibition on action verb understanding using tDCS. Right-handed participants received tDCS stimulation with the anodal electrode (presumed to be excitatory) and cathodal electrode (presumed to be inhibitory) placed

Error Processing and Inhibitory Control in Obsessive-Compulsive Disorder: A Meta-analysis Using Statistical Parametric Maps.

BACKGROUND: Error processing and inhibitory control enable the adjustment of behaviors to meet task demands. Functional magnetic resonance imaging studies report brain activation abnormalities in patients with obsessive-compulsive disorder (OCD) during both processes. However, conclusions are limited by inconsistencies in the literature and small sample sizes. Therefore, the aim here was to perform a meta-analysis of the existing literature using unthresholded statistical maps from previous studies. METHODS: A voxelwise seed-based d mapping meta-analysis was performed using t-maps from studie

Inhibition in the auditory brainstem enhances signal representation and regulates gain in complex acoustic environments.

Inhibition plays a crucial role in neural signal processing, shaping and limiting responses. In the auditory system, inhibition already modulates second order neurons in the cochlear nucleus, e.g. spherical bushy cells (SBCs). While the physiological basis of inhibition and excitation is well described, their functional interaction in signal processing remains elusive. Using a combination of in vivo loose-patch recordings, iontophoretic drug application, and detailed signal analysis in the Mongolian Gerbil, we demonstrate that inhibition is widely co-tuned with excitation, and leads only to mi

Cerebellar brain inhibition in the target and surround muscles during voluntary tonic activation.

Motor surround inhibition is the neural mechanism that selectively favours the contraction of target muscles and inhibits nearby muscles to prevent unwanted movements. This inhibition was previously reported at the onset of a movement, but not during a tonic contraction. Cerebellar brain inhibition (CBI) is reduced in active muscles during tonic activation; however, it has not been studied in the surround muscles. CBI was evaluated in the first dorsal interosseus (FDI) muscle as the target muscle, and the abductor digiti minimi, flexor carpi radialis and extensor carpi radialis muscles as surr

High doses of salicylate reduces glycinergic inhibition in the dorsal cochlear nucleus of the rat.

High doses of salicylate induce reversible tinnitus in experimental animals and humans, and is a common tinnitus model. Salicylate probably acts centrally and induces hyperactivity in specific auditory brainstem areas like the dorsal cochlear nucleus (DCN). However, little is known about the effect of high doses of salicylate in synapses and neurons of the DCN. Here we investigated the effects of salicylate on the excitability and evoked and spontaneous neurotransmission in the main neurons (fusiform, cartwheel and tuberculoventral) and synapses of the DCN using whole cell recordings in slices

Genetic evidence for the adhesion protein IgSF9/Dasm1 to regulate inhibitory synapse development independent of its intracellular domain.

Normal brain function requires balanced development of excitatory and inhibitory synapses. An imbalance in synaptic transmission underlies many brain disorders such as epilepsy, schizophrenia, and autism. Compared with excitatory synapses, relatively little is known about the molecular control of inhibitory synapse development. We used a genetic approach in mice to identify the Ig superfamily member IgSF9/Dasm1 as a candidate homophilic synaptic adhesion protein that regulates inhibitory synapse development. IgSF9 is expressed in pyramidal cells and subsets of interneurons in the CA1 region of

Influence of intramuscular heat stimulation on modulation of nociception: complex role of central opioid receptors in descending facilitation and inhibition.

It has been reported that the threshold to activate 'silent' or inactive descending facilitation of nociception is lower than that of descending inhibition. Thus, the development of pain therapy to effectively drive descending inhibition alone, without the confounding influences of facilitation is a challenge. To address this issue we investigated the effects of intramuscular stimulation with a heating-needle on spinal nociception, assessed by measuring nociceptive paw withdrawal reflex in rats. Additionally, involvement of the thalamic 'nociceptive discriminators' (thalamic mediodorsal (MD) a
