Anorectic effect of metformin in obese Zucker rats: lack of evidence for the involvement of neuropeptide Y.

The hypothalamic neuropeptide Y content and preproneuropeptide Y mRNA expression were studied in metformin-treated (300 mg/kg orally for 12 days), in pair-fed and in ad libitum-fed obese Zucker rats in order to elucidate possible mechanisms involved in the anorectic and body weight reducing effect of chronic metformin treatment in genetically obese Zucker rats. In addition the acute influence of metformin on food intake was studied by comparing its effects after oral and parenteral administration. The concentration of neuropeptide Y in the hypothalamic paraventricular nucleus was significantly

Anxiolytic-like effect of neuropeptide Y (NPY), but not other peptides in an operant conflict test.

The peptide messengers neuropeptide Y (NPY), growth hormone-releasing hormone (GHRH), atrial natriuretic peptide (ANP) and beta-endorphin (BEND) were tested in an animal model of anxiety, the Geller-Seifter conflict test. Rats were subjected to a multiple schedule consisting of three components: in the first component, lever-pressing produced food-reward ('unpunished responding'). The second component was a time-out period, during which lever-pressing had no consequences. During the third component, lever-pressing produced food-reward, but was also punished by an incremental foot-shock ('punis

Tyrosine hydroxylase and neuropeptide-Y immunoreactivity in pineal glands developing in situ and in pineal grafts.

Postnatal development of the innervation of the pineal gland in situ as well as the reinnervation of pineal grafts by tyrosine hydroxylase (TH)- and neuropeptide Y (NPY)-immunoreactive nerve fibers were examined using the avidin-biotin-peroxidase immunohistochemical technique. TH-immunoreactive nerve fibers appeared in the pineal gland on the second postnatal day (P2) in both hamsters and gerbils. NPY-immunoreactive nerve fibers first appeared in the pineal gland of gerbils on P2 and in the hamsters on P3. By the seventh postnatal day (P7), the pineal glands of both hamsters and gerbils were r

Neuropeptide Y and vasoactive intestinal peptide in experimental subarachnoid hemorrhage: immunocytochemistry, radioimmunoassay and pharmacology.

The involvement of noradrenaline (NA), neuropeptide Y, (NPY), 5-hydroxytryptamine (5-HT), acetylcholine (ACh) and vasoactive intestinal polypeptide (VIP) has been examined in the late phase of spasm after an experimental subarachnoid hemorrhage (SAH) in a rat model. Immunocytochemistry and radioimmunoassay of blood vessels from the circle of Willis did not show significant differences in NPY- and VIP-like immunoreactivity 2 days post SAH as compared to control vessels. The postjunctional effects of NA, NPY, 5-HT, ACh and VIP were studied two days after SAH using a sensitive in vitro system. NP

Sprouting of aberrant neuropeptide Y-immunoreactive sympathetic nerves into neonatally denervated smooth muscle.

Sympathetic nerves normally project ipsilaterally to lateral cranial targets. Following unilateral superior cervical ganglionectomy in neonatal rats, however, neurons from the contralateral superior cervical ganglion sprout into the denervated region. In the present study we examined neuropeptide Y immunoreactivity (NPY-ir) of neurons comprising ipsilateral (control) and denervation-induced contralateral pathways to the superior tarsal smooth muscle of the eyelid. Fluoro-Gold injection of the control muscle retrogradely labelled 133 +/- 18 neurons in the ipsilateral superior cervical ganglion;

Neuropeptide Y and somatostatin inhibit stimulated cyclic AMP production in rabbit ciliary processes.

The interaction of adrenergic and peptide receptors linked to adenylate cyclase and the inhibition by bioactive peptides of stimulated cyclic AMP production has been investigated in intact, excised rabbit ciliary processes. Cyclic AMP production stimulated by isoproterenol, vasoactive intestinal peptide, or forskolin was inhibited by the biologically active peptides neuropeptide Y, somatostatin, and the synthetic somatostatin analogue SMS 201-995. IC50s determined from dose-response curves of inhibition are consistent with the known abilities of these ligands to modulate cyclic AMP and physiol

Physiological role of neuropeptide Y (NPY) in control of anterior pituitary hormone release in the rat.

Neuropeptide Y (NPY) is a peptide originally isolated from porcine brain and subsequently shown to be widely distributed in the body of several species, including man. Neuropeptide Y is a circulating peptide; however, blood levels were higher in portal than peripheral blood of anesthesized rats. Earlier studies in ovariectomized and intact male rats have shown that intraventricular injection of NPY inhibits release of growth hormone (GH) and luteinizing hormone (LH) without producing significant modification of plasma follicle stimulating hormone (FSH) and thyrotropin stimulating hormone (TSH)

Elevation of plasma neuropeptide Y-like immunoreactivity and noradrenaline during myocardial ischaemia in man.

Plasma levels of neuropeptide Y-like immunoreactivity (NPY-LI) and noradrenaline were studied for 25 h in 22 patients with acute ischaemic heart disease. On admission, NPY-LI levels were above normal in 16 patients, and 20 patients had increased noradrenaline levels. The initial plasma NPY-LI did not differ between patients with acute myocardial infarction (AMI) and angina pectoris. Initial plasma noradrenaline levels were higher in patients with AMI than in those with angina pectoris. Plasma levels of noradrenaline remained elevated in AMI patients, but decreased towards normal values in pati

Neuropeptide Y-containing neurons in the rat superior cervical ganglion: projections to the pineal gland.

The aim of this study was to investigate the localization in the superior cervical ganglia (SCG) of neuropeptide Y-containing neurons innervating the pineal gland. Following injection of the fluorescent tracer Fluoro-Gold (FG) into the superficial part of the pineal gland and retrograde axonal transport, labeled cells were observed predominantly in the rostral third to half of SCG sections (average number 239 per ganglion). Incubation of the sections with neuropeptide Y (NPY) antiserum showed that the vast majority of neurons exhibit NPY-like immunoreactivity (NPY-LI). The comparison of cells

Arginine-vasopressin binding to isolated hippocampal microvessels of rats with different endogenous concentrations of the neuropeptide.

The binding of [125I]arginine-8-vasopressin (AVP) to hippocampal microvessels isolated from brains of normal Wistar rats, animals after water deprivation and heterozygous as well as homozygous diabetes-insipidus rats (Brattleboro strain) were measured. Data from binding experiments from the microvessels of the different groups of animals in each case revealed a single class of high affinity binding sites. However, the binding parameters between the different groups of rats were different. The affinity constants differs by a factor of 5.1, ranging from KD = 1.18 nmol X L-1 (animals after water

The effect of neuropeptide Y and peptide YY on electrogenic ion transport in rat intestinal epithelia.

1. Neuropeptide Y (NPY), peptide YY (PYY) and, to a lesser extent, human pancreatic polypeptide (HPP) reduced short-circuit current (SCC) in … K+-2Cl- co-transport inhibitor). These results may indicate that the reduction in SCC caused by the neuropeptides is due to a net increase in chloride movement in the apical to basolateral direction. 5. 36Cl-flux studies identified an inhibition of chloride secretion as the predominant mechanism of action of NPY and PYY, together with a smaller stimulation of c …

Co-release and functional interactions of neuropeptide Y and noradrenaline in peripheral sympathetic vascular control.

1. The immunohistochemical results suggest that NPY-LI co-exists with NA in sympathetic periarterial nerves of skeletal muscle. The corresponding veins are associated with few or no sympathetic nerves. 2. SNS evoked a detectable overflow of NPY-LI from the dog gracilis muscle in vivo during high-frequency but not low-frequency stimulation. After the administration of alpha-adrenoceptor antagonists, the overflow of NPY-LI was enhanced, indicating prejunctional inhibition of release via a mechanism similar to that for NA. Reserpine pretreatment depleted both NA and NPY-LI from skeletal muscle. T

Differential effects of reserpine and 6-hydroxydopamine on neuropeptide Y (NPY) and noradrenaline in peripheral neurons.

The effects of 6-hydroxydopamine (6-OHDA) and reserpine pretreatment on peripheral neuropeptide Y (NPY)- and noradrenaline (NA)-containing neurons were studied in guinea-pigs. Ten days after 6-OHDA pretreatment, a 60-80% reduction of the NA content was observed in the right atrium of the heart, stellate ganglion and spleen. The content of NPY-like immunoreactivity (LI) was reduced by about 50% in the heart, not changed in the spleen while it increased to 200% of control in the stellate ganglion. Immunohistochemistry showed a pronounced loss of NPY- and tyrosinehydroxylase (TH)-immunoreactive (

Plant volatile TMTT suppresses reproduction of the brown planthopper by affecting the neuropeptide receptor CapaR.

BACKGROUND: The brown planthopper, Nilaparvata lugens, is a major rice pest whose management heavily depends on chemical insecticides, raising concerns over resistance and ecological safety. Herbivore-induced plant volatiles (HIPVs) are key components of plant defense and can influence insect behavior and physiology. However, their molecular effects on insect reproduction remain poorly understood. This study aimed to investigate how the plant volatile (3E,7E)-4,8,12-trimethyltrideca-1,3,7,11-tetraene (TMTT) affects the reproduction of N. lugens and to elucidate the underlying molecular mechani

RNA interference targeting neuropeptide F inhibits the lipid metabolism and reproduction in Drosophila suzukii.

Neuropeptide F (NPF) is an important signaling molecule that acts as a neuromodulator to regulate diverse physiological and behavioral processes across vertebrates and invertebrates. However, the role of NPF in the globally invasive pest of soft-skinned fruits, Drosophila suzukii, remains unknown. Here, we identified and characterized cDNA encoding one NPF precursor from D. suzukii. DsNPF was most highly expressed during the adult stage and showed the highest expression levels in the midgut. Three-dimensional models of the DsNPF-DsNPFR complex generated by AlphaFold 3 revealed that three resid

A neuropeptide receptor-enriched transcriptional state characterizes resilient dopaminergic neurons in Parkinson's disease.

Dopaminergic neurons (DANs) exhibit subtype-specific vulnerability in Parkinson's disease (PD), but the molecular basis of selective resilience remains poorly understood. Here, we investigated the role of neuropeptide receptor (NPR) signaling in DAN survival using single-nucleus RNA sequencing of 8065 DANs from postmortem substantia nigra of individuals with PD and matched controls. Despite pronounced neuronal loss in PD, surviving DANs showed a higher NPR transcript burden and increased NPR gene co-expression per cell. Using a 25-gene NPR score, we stratified DANs into high-, mid-, and low-NP

Dependence of energy balance and hypothalamic neuropeptide gene expression on initial tumor load in mice.

INTRODUCTION: Animal models of cancer anorexia/cachexia are instrumental in investigating the underlying mechanisms. However, even among reports on the same cancer model, experimental procedures can vary, hindering the comparisons between studies. METHODS: To determine the impact of the number of implanted cancer cells on experimental outcomes, we directly compared tumor-bearing (TB) mice inoculated with Lewis-lung carcinoma at 1x106 cells (TBhigh) or 0.5x106 cells (TBlow). RESULTS: While both TB groups showed lower weight gain and adiposity than vehicle controls, TBlow mice displayed a larg

Sexually dimorphic effects of angiopoietin-like 2 on energy metabolism and hypothalamic neuropeptide regulation.

BACKGROUND: Adipokines regulate body weight and metabolism by targeting the hypothalamus, influencing feeding, energy expenditure (EE) and insulin sensitivity. Angiopoietin-like 2 (Angptl2) is a pro-inflammatory adipokine linking obesity to insulin resistance. Both Angptl2 and its receptor are expressed in the central nervous system. Yet, the contribution of Angptl2 to the regulation of energy metabolism and relevant hypothalamic neuropeptides in male and female mice is unknown. We aim at determining the impact of Angptl2 knockdown (KD) on energy balance, nutrient partitioning and hypothalamic…

Subcellular interactions of neuropeptide Y and corticotropin-releasing factor in the central nucleus of the amygdala in the mouse.

Neuropeptide Y (NPY) is ubiquitously distributed throughout the central nervous system. Recognized as a mediator of stress resilience, NPY has been shown to counteract the excitatory effects of the neuropeptide corticotropin-releasing factor (CRF), that orchestrates the stress response. In the mouse, while NPY and CRF exhibit a high degree of neuroanatomical association in the central nucleus of the amygdala (CeA) indicating potential significant interactions, the synaptic organizations of these neuropeptides have not been elucidated. In the present study, we determined the interactions betwee…
