Affinity Chromatography With Selective Nucleic Acid Hybridization: Direct Coupling With Size Exclusion Chromatography.

Affinity liquid chromatography (aLC) with immobilized oligonucleotides provides high selectivity for nucleic acid targets through on-column hybridization. We describe the preparation and evaluation of prototype 2.1 mm × 20 mm affinity columns packed with a 3.5 µm nonporous polystyrene/divinylbenzene sorbent covalently modified with streptavidin. Two different affinity columns were prepared by immobilizing biotinylated oligonucleotides on-column via biotin-streptavidin bond: (i) biotinylated 25-nt oligodeoxythymidine, dT25 or (ii) biotinylated 21 nt TP53 oligonucleotide. dT25 column was used fo

Invalid Nucleic Acid Test Results May Limit Deceased Donor Organ Utilization.

BACKGROUND: Nucleic acid testing (NAT) is essential for detecting transmissible viral infections in deceased organ donors and guiding safe organ allocation. However, invalid discriminatory NAT (dNAT) results may lead to unnecessary donor exclusion and reduced organ availability. METHODS: We performed a retrospective analysis of HIV-1/2, HBV, and HCV NAT results from deceased organ donors in the United States. Donor specimens were tested at eleven laboratories using FDA-approved donor screening assays. Donor serologic results and organ utilization outcomes were reviewed to evaluate the clinica

Application of aptamer nucleic acid test strips in the detection of tumor markers for colorectal cancer.

At present, the detection methods of tumor markers rely on ELISA, gene sequencing, and other technologies. However, these methods require specialized equipment and technical personnel, and the low concentrations of early tumor markers limit their sensitivity. This study developed a method for detecting colorectal cancer tumour markers. It uses a test strip and isothermal amplification to detect low concentrations of tumour markers. The test strip is easy to use and cost-effective. The test strip developed in this study can detect isothermal amplification products in 3 min, and the detection li

Ultrasensitive CRISPR Platform Enables Rapid and Extraction-Free Detection of Viral Nucleic Acid.

CRISPR-based molecular diagnostics offers great potential for the rapid identification of pathogens. However, existing one-pot detection systems remain constrained by their restricted versatility and operational complexity. Herein, we report a novel strategy termed interspaced phosphorothioate primer-mediated one-pot detection (iPSOT), which substitutes conventional phosphodiester primers with interspaced phosphorothioate (iPS) primers. The iPS primers demonstrate strong compatibility with AapCas12b during loop-mediated isothermal amplification, enabling robust fluorescence signal generation w

Mechanistic advances in nanomedicine, nucleic acid therapies, and AI-driven research on cervical cancer.

Cervical cancer pharmacotherapy is significantly limited by physiological and cellular barriers that restrict drug access to therapeutic targets, resulting in suboptimal biodistribution, systemic toxicity, and the emergence of drug resistance. This review provides a mechanistic and biopharmaceutics-centered analysis of how advanced drug delivery systems are being engineered to overcome these limitations. We critically examine the role of nanocarriers, including lipid-based vesicles, polymeric nanoparticles, and inorganic hybrid systems, in modulating absorption, distribution, and tumor-targeti

Elimination of cis-cleavage in CRISPR diagnostics for one-pot rapid nucleic acid detection.

Current one-pot clustered regularly interspaced short palindromic repeats diagnostics are limited by the cis-cleavage activity of Cas nucleases, which leads to amplicon degradation during amplification. Here, we report a streamlined strategy that overcomes this limitation. By integrating a bipartite split-crRNA into Cas12a (SCas12a), we separate target recognition from PAM dependency and completely eliminate cis-cleavage while preserving robust trans-cleavage. This strategy is broadly applicable for one-pot testing, compatible with recombinase polymerase amplification, RT-RPA, and loop-mediate

Ozone disinfection effectively reduces pathogenic microbial nucleic acid background in mNGS laboratories.

For the first time, this report confirmed under real-laboratory-contamination scenarios that ozone disinfection effectively reduces microbial nucleic acid background in mNGS laboratories, with consistent and efficient disinfection performance. With the global advancement of standardized clinical application of mNGS technology, the results provide a practical strategy to control laboratory contamination, reduce false-positive risks, and maintain a low nucleic acid baseline. This method addresses the difficulty of eliminating persistent contamination in mNGS laboratories and offers a direct refe

An in-house nucleic acid test for detection of Anaplasma phagocytophilum in whole blood donor samples.

BACKGROUND: Anaplasma phagocytophilum is a tick-borne pathogen that causes anaplasmosis. Increased incidences of this disease in Canada and cases of transfusion-transmitted anaplasmosis in the United States have been reported. Currently, there are no Health Canada licensed methodologies available for detection of A. phagocytophilum nucleic acids (NAs) in blood donors. Here we describe a method for NA extraction and detection of A. phagocytophilum in whole blood (WB) donor samples. STUDY DESIGN AND METHODS: Plasmids carrying the A. phagocytophilum msp2 and gltA genes and genomic A. phagocytoph…

Mismatch-Tuned Plasmonic Nanogap Networks on Block Copolymers for Ultrasensitive Nucleic Acid Quantification.

In localized surface plasmon resonance (LSPR), rational nanogap engineering in metallic nanostructures enables strong plasmonic coupling and efficient confinement of incident electromagnetic fields, thereby significantly enhancing optical responses for biosensing applications. Traditional approaches to fabricating small gaps have relied on localizing dielectric spacers between gold nanoparticles (AuNPs). However, doing so has encountered challenges in producing high-density, clean gaps across large surface areas. Here, we demonstrate a straightforward, self-assembly-guided method for the consi

Benchmarking Denoising Strategies and Nucleic Acid Templates for ITS2 Amplicon Sequencing of Gut Mycobiome.

The human gut mycobiome plays a critical role in host immunity, yet standardized workflows for fungal internal transcribed spacer (ITS) amplicon sequencing remain elusive. Unlike the bacterial 16S rRNA gene, where the impact of DNA versus RNA templates is well-documented, such comparative benchmarks are lacking for fungal studies. Furthermore, fungal ITS regions exhibit significant length heterogeneity, posing unique challenges for denoising algorithms that were originally designed for conserved bacterial amplicons. We performed fungal ITS2 sequencing on six fecal samples, generating 12 matche

Exploring Nucleic Acid Structure and Function with Molecular Dynamics and Fragment Molecular Orbital Methods.

In recent years, nucleic acid therapeutics have attracted increasing attention as a new modality for drug discovery and as a novel therapeutic approach for rare and intractable diseases. The G-quadruplex structure of nucleic acids is of interest as both a target in drug development and as a candidate for nucleic acid therapeutics, as it can inhibit replication, transcription, reverse transcription, translation, and elongation reactions. The G-quadruplex structures are known to undergo significant changes in both conformation and stability in response to environmental factors such as ionic conc…

Nucleic acid-based therapeutic strategies for modulator-refractory cystic fibrosis-causing variants.

Cystic fibrosis (CF) is a life-limiting autosomal recessive disorder caused by pathogenic variants in the CF transmembrane conductance regulator (CFTR) gene that impair epithelial chloride ion transport, leading to progressive lung dysfunction among other systemic manifestations. Small molecule CFTR modulators have dramatically improved outcomes for people with CF (pwCF) by promoting the trafficking of defective CFTR protein to the cell surface and enhancing its activity. However, a substantial proportion of pwCF harbor genotypes which remain refractory to CFTR modulators and thus lack approve

Low-cost, handheld, multi-pulse electroporators for simplified nucleic acid delivery in skin.

Electroporation-mediated delivery offers a promising alternative to carrier-based nucleic acid delivery methods for vaccination and therapeutic applications. Carrier-based systems like lipid nanoparticles and viral vectors often suffer from poor in vivo stability, immunogenicity, toxicity, and off-target effects. To overcome the high cost, bulkiness, lack of portability, and painful administration of traditional electroporators, we developed the RotoPatch family of small, low-cost, hand-held piezoelectric electroporators that use microneedle electrodes for intradermal delivery of nucleic acids…

Point-of-care nucleic acid testing - a step forward in controlling the HIV epidemic: A review.

INTRODUCTION: The HIV/AIDS epidemic, with 85.6 million infections and 40.4 million AIDS-related deaths globally, remains a critical public health challenge. Current diagnostic methods, primarily fourth-generation immunoassays, have limitations due to their long window periods, and most viral load assays require centralized testing protocols that result in delays, especially in remote regions. NUCLEIC ACID TESTING: Point-of-care (POC) nucleic acid amplification testing (NAAT) presents a transformative approach by reducing the window period for detection to one week and significantly shortening
