Prenatal diagnosis of intrauterine infection with parvovirus B19 by the polymerase chain reaction technique.

Human parvovirus B19 is a recently recognized cause of fetal hydrops and death. Efforts to characterize the natural history of fetal infection with this virus have been hampered by the lack of sensitive and specific tests for diagnosis in utero. Using the highly sensitive polymerase chain reaction (PCR) assay, we determined the fetal infection status in 56 pregnancies by testing amniotic fluid, fetal serum, and maternal serum for B19 DNA and antibodies. Factors associated with a high risk of B19 infection were fetal disease, exposure to persons with erythema infectiosum, or signs or symptoms o…

Severe anemia due to B19 parvovirus infection in children with acute leukemia in remission.

Two children developed severe anemia with reticulocytopenia while on maintenance chemotherapy for acute lymphoblastic leukemia. Bone marrow … as tested 16 months after the episode of anemia, he had elevated IgG and IgM antibodies against B19 parvovirus. The patient did not experience recurrent viremia or anemia over a two year period. Thus, patients with leukemia are at risk to develop severe anemia when infected with B19 parvovirus.

Management and outcomes of pregnancies complicated by human B19 parvovirus infection: a prospective study.

During a large statewide outbreak of fifth disease in Connecticut in 1988, 39 pregnant women were identified who had serologic evidence of recent human B19 parvovirus infection. The patients were followed up prospectively with targeted fetal ultrasonographic examinations to detect signs of fetal hydrops. Of these 39 pregnant women, 37 had healthy infants and two patients had miscarriages. None of the fetuses developed hydrops. We propose that pregnant women exposed to B19 parvovirus be tested for evidence of IgG and IgM B19-specific antibodies and that targeted fetal ultrasonography be conside…

Frequency of bluetongue and bovine parvovirus infection in cattle in South Carolina dairy herds.

A survey of South Carolina diary cattle in 12 commercial herds was conducted, using serologic testing to determine the frequency and impact on reproduction of bluetongue (BT) and bovine parvovirus (BFV) infections. Results of the study of serum antibodies titers and dairy cattle health records indicated that the majority of the cattle surveyed was adequately protected against the major reproductive tract diseases through vaccination programs. The frequency of reproductive dysfunction was common in vaccinated herds, however. Six of the 12 herds in the survey had cattle which were BT serotest re…

Development of canine parvovirus-2-based recombinant pseudoviruses expression system: a potential vaccine platform.

Canine parvovirus-2 (CPV-2) and canine distemper virus (CDV) are two highly infectious and pathogenic pathogens that harm canids and various carnivores, and often cause co-infections in clinical settings. Although commercial attenuated live vaccines against CPV-2 and CDV have been widely used to prevent these viral infections, there are still issues of biosafety and insufficient protection against new variants, thus requiring novel vaccines. In this study, we developed a recombinant pseudovirus expression system based on the CPV-2 full-length infectious clone, and found that the recombinant CP…

Bilateral Parsonage-Turner Syndrome in a Young Adult Man Due to Parvovirus B19 Infection.

Parsonage-Turner syndrome (PTS) is a rare neuromuscular disorder. Parvovirus B19 infection commonly causes a childhood exanthematous illness. Most previous case reports have documented unilateral parvovirus B19-associated PTS along with the systemic manifestations of parvovirus infection. This report highlights parvovirus B19-associated bilateral PTS without systemic manifestations. A 20-year-old gentleman was asymptomatic one and a half months ago when he developed an acute onset of excruciating right shoulder pain. After 7-10 days, he developed weakness and wasting in the right shoulder gird…

Intrauterine transfusion in monochorionic twins with parvovirus B19 infection: Evidence of intertwin blood exchange.

BACKGROUND: Parvovirus B19 infection can cause severe fetal anemia. Management in monochorionic twin pregnancies is challenging due to placental vascular connections. CASE: We report a case of severe fetal anemia in both fetuses of a monochorionic diamniotic twin pregnancy following maternal parvovirus B19 infection. Elevated middle cerebral artery peak systolic velocity (MCA-PSV) was first detected at 16 weeks in twin A and reached 2.32 multiples of the median (MoM) in twin A and 2.30 MoM in twin B at 21+6 weeks. These findings were associated with polyhydramnios in both fetuses and ascites …

Peripheral polyneuropathy in three puppies temporally associated with canine parvovirus infection resembling acute polyradiculoneuritis.

Molecular mimicry is hypothesized to play a key role in the pathogenesis of acute polyradiculoneuritis (APRN) in dogs, as in Guillain-Barré syndrome (GBS). GBS is linked to Campylobacter jejuni and cytomegalovirus, while Parvovirus B19 shows temporal and serological associations. In dogs, the cause of APRN is often unknown, although recent vaccination or ingestion of raw poultry contaminated with Clostridium perfringens is documented. Antiganglioside antibodies are potential serum biomarkers for APRN. Eight 2.5-month-old Sprocker littermates and a 3-month-old female Cockapoo developed severe g…

Adenovirus, herpesvirus, and parvovirus survey in Iberian wolves (Canis lupus signatus) of Cantabria, Spain.

The study of infectious diseases in wild carnivores is essential to their conservation. Adenovirus, herpesvirus and parvovirus are DNA viruses able to cause severe disease in carnivores; however, data on their occurrence in Iberian wolves (Canis lupus signatus) are limited. Herein, 130 spleen samples from Iberian wolves in Cantabria, northern Spain, were tested for these viruses using broad-range nested PCRs and real time PCR. All positive amplicons were confirmed by direct Sanger sequencing in both directions. Additionally, exposure to adenovirus and Carnivore protoparvovirus 1 (CCPV1) was ev…

Evaluation of hematologic indices in parvovirus infected dogs with systemic inflammatory response syndrome (SIRS).

Canine parvoviral enteritis (CPE) is a highly contagious, acute viral disease that mainly affects young dogs and can lead to systemic inflammatory response syndrome (SIRS), sepsis, endotoxemia and even death. The study aimed to evaluate the diagnostic and prognostic potential of hematological indices such as MLR, NLR, PLR, PNR and SII in CPV-infected dogs with SIRS at first admission. The retrospective study was performed on 51 CPV-infected and 20 healthy dogs. MLR, NLR, PLR, PNR, and SII indices were determined and compared between groups. A receiver operating characteristic (ROC) curve analy

Parvovirus B19 Infection after Transfusion of a Pathogen-Reduced Platelet Concentrate: A Case Report.

INTRODUCTION: Parvovirus B19 (B19) exhibits moderate resistance to the pathogen reduction methods employed during the production of platelet concentrates. In Switzerland, nucleic acid testing (NAT) for B19 is conducted for plasma fractionation; however, its results are not mandatory for the release of blood products. CASE PRESENTATION: We describe a case of B19 infection in an immunosuppressed, seronegative patient who received a pooled platelet concentrate treated with UVA light and amotosalen. Although the patient remained clinically asymptomatic, the infection was confirmed through laborat…

Congenital Parvovirus B19 During the 2024 European Resurgence: A Prospective Single-Centre Cohort Study.

Parvovirus B19 (B19V) re-emerged across Europe in 2024, raising concerns about vertical transmission and neonatal morbidity. We undertook a prospective, single-centre cohort study to characterise the early clinical course of congenitally infected neonates born between April and December 2024. Seventy-one pregnancies with serologically or PCR-confirmed maternal infection were enrolled; seven neonates met laboratory criteria for congenital B19V infection and were followed with serial clinical, biochemical and imaging assessments through the first year of life. Troponin I and CK-MB were measured …

Human parvovirus B-19 meningitis: a sentinel event unmasking primary immunodeficiency in an infant.

Primary immunodeficiencies are genetic disorders that impair immune function, increasing the risk of infections, autoimmune diseases and cancer. Patients with severe viral infections are often not thoroughly evaluated for immunodeficiencies. This case presents a young infant with human parvovirus B-19 meningitis and persistent lymphocytopenia, whose immunophenotyping revealed low absolute counts of T, B and natural killer cells, leading to a diagnosis of underlying immunodeficiency. This case underscores the necessity for vigilant diagnosis when standard presentation is complicated by recurren…

Characterization of Natural Compounds as Inhibitors of NS1 Endonuclease from Canine Parvovirus Type 2.

Canine parvovirus type 2 (CPV-2) has high morbidity and mortality rates in canines. Nonstructural protein 1 (NS1) of CPV-2 has endonuclease activity, initiates viral DNA replication, and is highly conserved. Thus, it is a promising target for antiviral inhibitor development. We overexpressed a 41.9 kDa active recombinant endonuclease in Escherichia coli and designed a nicking assay using carboxyfluorescein and quencher-linked ssDNA as substrates. The optimal temperature and pH of the endonuclease were 37°C and pH 7, respectively. Curcumin, bisdemethoxycurcumin, demethoxycurcumin, linoleic acid…

Diagnosis of Peripheral Facial Palsy Associated with Parvovirus B19 Infection by Polymerase Chain Reaction.

Human parvovirus B19 (PVB19) infection causes neurological manifestations, including encephalitis, meningitis, and neuropathy, but facial nerve palsy is rare. Moreover, no case of facial nerve palsy related to PVB19 infection that was diagnosed by PCR and serology has been reported. A 19-month-old boy without the medical history developed facial nerve palsy and was treated with prednisolone and valacyclovir. On the 19th day, erythema appeared on his body, and the PVB19-specific IgM and PVB19 DNA were detected in the serum, leading to the diagnosis of infectious erythema associated with PVB19 i…

Safety and immunogenicity of parvovirus B19 virus-like particle vaccine lacking phospholipase A2 activity.

Parvovirus B19 (B19) belongs to the Erythroparvovirus genus and is known to cause the fifth disease in children. Primary infection of pregnant women is associated with a high risk of hydrops fetalis and stillbirth due to severe fetal anemia. Virus-like particle (VLP) vaccine candidates for B19 have been developed, although none have been approved so far. The B19 phospholipase A2 domain (B19 PLA2), located in the VP1 unique region, is believed to be associated with adverse inflammatory reactions, and previous effective attempts to improve this vaccine modality inserted a mutation to impair the …
