Hospital or population controls for case-control studies of severe childhood diseases?

There are few empirical data to determine which control group seems best in a case-control study for a severe disease: population controls or hospital controls. The author conducted a case-control study of leukemia in children using two control groups, population and hospital controls (cancers other than leukemia and severe blood diseases), between 1980 and 1993 in Québec, Canada. Maternal, paternal, and child factors not known to be associated with leukemia as well as factors possibly associated were selected for analysis. Most factors were taken directly from parental interviews, but two fac

Hierarchical Algorithms for Constructing Mother-Father-Infant Linkages Under Modern Privacy Constraints.

BACKGROUND: Post-marketing safety surveillance of medications used during pregnancy depends on accurate reconstruction of mother-father-infant triads in administrative healthcare data. However, global privacy regulations increasingly restrict the use of direct familial identifiers, and few parent-infant linkage algorithms have been validated against a reference standard. OBJECTIVE: We aimed to develop and validate a hierarchical deterministic algorithm that constructs mother-infant and father-infant linkages from variables widely available in administrative healthcare data internationally. Sharp Foreign Bodies of the Aero-Digestive Tract: Endoscopic Removal by the 'Kangaroo' Technique.

BACKGROUND: Our prevalence study on Palestinian school children aged 6-12 years showed lower rates for asthma and asthma symptoms than economically developed and industrialized countries. Reasons for such differences are largely unknown, and could possibly be related to different environmental and lifestyle factors. OBJECTIVE: To investigate familial, early life exposures and indoor environmental determinants for asthma in children in Palestine. METHODS: From the population of our previous study, a group of 273 children with wheeze in the past 12 months (of whom 99 children had physician-dia

The Cancer, Lifestyle and Evaluation of Risk Study (CLEAR): Rationale and design of an unmatched "case-spouse control" study of over 10,000 participants in New South Wales, Australia.

INTRODUCTION: The New South Wales (NSW) Cancer, Lifestyle and Evaluation of Risk Study (CLEAR) is an open epidemiological bioresource, using an all cancer unmatched case-spouse control design. Participant characteristics and selected confirmed associations are compared to published estimates: current smoking and lung cancer; country of birth and melanoma; body mass index (BMI) and bowel cancer; and paternal history of prostate cancer and prostate cancer, to illustrate the validity of this design. MATERIAL AND METHODS: Cases are NSW residents, ≥18 years, with an incident cancer of any type. Co

Prenatal Androgen Excess Induces Multigenerational Effects on Female and Male Descendants.

BACKGROUND: It is still unelucidated how hormonal alterations affect developing organisms and their descendants. Particularly, the effects of androgen levels are of clinical relevance as they are usually high in women with Polycystic Ovary Syndrome (PCOS). Moreover, it is still unknown how androgens may affect males' health and their descendants. OBJECTIVES: We aimed to evaluate the multigenerational effect of prenatal androgen excess until a second generation at early developmental stages considering both maternal and paternal effects. DESIGN AND METHODS: This is an animal model study. Fema

Susceptibility to e-cigarette adoption among tobacco-naïve youths: a cross-sectional study in Shenzhen, China.

BACKGROUND: The rise in e-cigarette use among youth is a significant global public health issue. It is important to identify those at increased risk and implement effective strategies to reduce e-cigarette popularity among the youth. OBJECTIVE: This study aims to identify predictors of e-cigarette uptake in youths with no prior tobacco use, considering individual, familial and the broader societal environmental factors. METHODS: For this investigation, a group of 2,487 tobacco-free youths was selected from 15 high schools in Shenzhen, China. Susceptibility to e-cigarettes was determined by a

Exploration of gene-environment interactions, maternal effects and parent of origin effects in the etiology of hypospadias.

PURPOSE: Hypospadias is a common congenital malformation of the male external genitalia. Association studies for single nucleotide polymorphisms in genes encoding steroid 5alpha-reductase, estrogen receptors 1 and 2, and activating transcription factor 3 have been equivocal. We examined whether nonreplication of findings for 4 single nucleotide polymorphisms in these genes could be due to interaction with environmental exposures. MATERIALS AND METHODS: We genotyped 712 Dutch hypospadias case-parent triads for the 4 single nucleotide polymorphisms, used questionnaire information to determine e

Digital Pacifier for Parents: A Cross-Sectional Study of Mobile Phone Use, Patterns, and Parental Attitudes in Young Children in India.

Background The increasing reliance on smartphones and digital media to calm or occupy young children, often referred to as the "digital pacifier," has become widespread in Indian households. While this approach provides immediate convenience to caregivers, emerging research indicates potential adverse impacts on sleep patterns, behavior, and early developmental outcomes. Sociodemographic factors are likely to significantly influence this practice. Objective This study aims to characterize the sociodemographic characteristics and associated factors influencing the use of digital devices as tool

A biomarker-based study of prenatal smoking exposure and autism in a Finnish national birth cohort.

Maternal exposure to tobacco smoke during pregnancy is a common and persistent exposure linked to adverse neurodevelopmental outcomes in the offspring. However, previous studies provide mixed evidence regarding the relationship between prenatal smoking and offspring autism. This study used cotinine level, a biomarker for nicotine, to investigate the relationship between prenatal smoking and autism. The authors conducted a population-based case-control study nested in a national cohort of all births in Finland from 1987 to 2005. Cases diagnosed with childhood autism (ICD-10/9 code F84.0/299.0)

Tobacco smoke exposure and the risk of childhood acute lymphoblastic and myeloid leukemias by cytogenetic subtype.

BACKGROUND: Tobacco smoke contains carcinogens known to damage somatic and germ cells. We investigated the effect of tobacco smoke on the risk of childhood acute lymphoblastic leukemia (ALL) and myeloid leukemia (AML), especially subtypes of prenatal origin such as ALL with translocation t(12;21) or high-hyperdiploidy (51-67 chromosomes). METHODS: We collected information on exposures to tobacco smoking before conception, during pregnancy, and after birth in 767 ALL cases, 135 AML cases, and 1,139 controls (1996-2008). Among cases, chromosome translocations, deletions, or aneuploidy were iden

The Chromatin Configuration Promotes Constitutional Chromosomal Rearrangements in Post-Meiotic Sperm.

The complex nuclear packaging of sperm DNA, known as sperm chromatin organization, is essential for protecting the paternal genome during fertilization. Recent studies suggest that changes in the post-meiotic chromatin structure, a specific arrangement that forms after meiosis is completed, may be associated with an increased risk of constitutional chromosomal rearrangements (CCRs) in offspring. This review examines the potential mechanisms by which abnormal post-meiotic organization may impair DNA stability in spermatogonial cells, thereby leading to CCR formation. We discuss the interplay be

Seminal plasma exposure modulates peripheral T cell-associated transcription during early pregnancy in the mare.

Successful establishment of pregnancy requires maternal immune tolerance to paternal antigens introduced by insemination. Regulatory T cell (Treg)-associated responses are thought to play a central role in this process, whereas pro-inflammatory T helper 1 (Th1) responses may disrupt pregnancy success. In other species, seminal plasma (SP) has been shown to modulate maternal immune adaptation; however, this relationship remains poorly defined in mares, where SP is frequently reduced during routine breeding practices. The objectives of this study were to (1) characterize temporal changes in syst

Ethylene glycol monomethyl ether altered rat sperm small RNAs with critical developmental roles.

Ethylene glycol monomethyl ether (EGME) is a testicular germ cell toxicant that selectively targets spermatocytes. In rats, male-only EGME exposure reduces mating success and can lead to an increase in resorbed fetuses. In a previous study, 5-d exposure to 50, 60, or 75 mg/kg/d EGME in male rats led to a decrease in sperm motility and increase in retained spermatid heads with a lowest observed adverse effect level (LOAEL) of 75 mg/kg/d. At 60 mg/kg/d, EGME exposure altered the proportion of sperm small RNA reads mapped to different small RNA categories and the distribution of read lengths. To

Prenatal antidepressant exposure and emotional disorders until age 22: a danish register study.

BACKGROUND: Selective serotonin reuptake inhibitors (SSRIs) are the most frequently prescribed antidepressants in pregnancy. Animal and some clinical studies have suggested potential increases in depression and anxiety following prenatal SSRI exposure, but the extent to which these are driven by the medication remains unclear. We used Danish population data to test associations between maternal SSRI use during pregnancy and children outcomes up to age 22. METHODS: We prospectively followed 1,094,202 single-birth Danish children born 1997-2015. The primary exposure was ≥ 1 SSRI prescription fi

Parental mood during pregnancy and post-natally is associated with offspring risk of Tourette syndrome or chronic tics: prospective data from the Avon Longitudinal Study of Parents and Children (ALSPAC).

Little is known about risk factors for Tourette syndrome (TS) and chronic tic disorders (CT) but maternal psychological morbidity in pregnancy may be associated with TS/CT. We examined whether pre- and post-natal parental anxiety and/or depression are associated with risk of TS/CT in the Avon Longitudinal Study of Parents and Children. We compared self-reported anxiety and depression measures collected prospectively at four time points (18 and 32 weeks prenatally, and 8 weeks and 8 months post-natally) among parents of children who subsequently met criteria for TS/CT at 13 years of age as comp

Pregnancy outcomes of women whom spouse fathered children after tyrosine kinase inhibitor therapy for chronic myeloid leukemia: A systematic review.

INTRODUCTION: The introduction of tyrosine kinase inhibitors (TKIs) has revolutionized the therapy of chronic myeloid leukemia (CML). Although the efficacy of TKIs is beyond dispute, conception-related safety issues are still waiting to be explored, particularly in males. This systematic review aimed to summarize all available evidence on pregnancy outcomes of female spouses of male CML patients who fathered children after TKI treatment for CML. METHODS: We performed a systematic search in seven electronic databases for studies that reported on male CML patients who did or did not discontinue
