Fabrication of hierarchically porous Cu/PVDF/CeO2 membranes via synergistic NIPS-spraying for robust oil-water separation.

High-efficiency oil-water separation membranes based on low-mesh copper mesh offer advantages in cost-effectiveness, scalability, and adaptability to complex separation scenarios. However, their inherently large pore size usually results in insufficient separation precision. In this work, the Cu/PVDF/CeO2 composite membrane is fabricated on the copper mesh substrate by combining the non-solvent induced phase separation method with spraying and immersion techniques. By adjusting the porogen content, the pore size of the composite membrane is effectively regulated, leading to enhanced separation…

Design and Characterization of Self-Assembling Peptide (SAP)-Polyethylene Glycol (PEG) Multiblock Copolymers (MBCP) Thin Films.

Self-assembling peptides (SAPs) provide a versatile, biocompatible platform for soft biomaterials, but their application in mechanically rob … peptide fiber-rich domains embedded in a soft, amorphous PEG matrix. At room temperature, only microphase separation persisted, while PEG crystallization produced a semicrystalline matrix in which peptide fibers restricted large crystal growth and were likely segregated at crystallite interfaces. Extensive mechanical testing revealed resilient films with shape-rec …

Regulating phase inversion behavior in cellulose acetate membranes using hydroxyethyl cellulose: Suppression of macrovoids and development of uniform sponge-like porous structures.

Porous cellulose acetate (CA) membranes fabricated via the nonsolvent-induced phase separation (NIPS) process often exhibit finger-like macrovoids and heterogeneous pore structures arising from rapid solvent-nonsolvent exchange. In this study, hydroxyethyl cellulose (HEC), a water-affinity cellulose derivative, was introduced at low concentration as a functional additive to regulate phase inversion behavior during the NIPS process. The incorporation of HEC transformed the conventional macrovoid-containing morphology of Porous_CA into a uniform sponge-like porous structure while increasing memb…

Water Under Molecular Crowding: From Femtosecond H-Bond Networks Dynamics to Biological Function.

Cells are densely packed with macromolecules, creating a crowded environment that fundamentally alters water's hydrogen bond (H-bond) networks. H-bond networks, typically dynamic and tetrahedral in bulk water, are disrupted by macromolecular surfaces, confinement, and competitive solute interactions. Subtle shifts in H-bond lifetimes, connectivity, and reorientation dynamics cascade into large-scale biological consequences-modulating protein folding, enzymatic function, and liquid-liquid phase separation. This chapter highlights how recent advances in time-resolved vibrational spectroscopy (e.…

Biomolecular condensates and disease pathogenesis.

Biomolecular condensates or membraneless organelles (MLOs) formed by liquid-liquid phase separation (LLPS) divide intracellular spaces into discrete compartments for specific functions. Dysregulation of LLPS or aberrant phase transition that disturbs the formation or material states of MLOs is closely correlated with neurodegeneration, tumorigenesis, and many other pathological processes. Herein, we summarize the recent progress in development of methods to monitor phase separation and we discuss the biogenesis and function of MLOs formed through phase separation. We then present emerging proo…

Size of Biomolecular Condensates Dictates Fate in Liquid-Solid Phase Transitions through Amorphous-Amyloid Competition.

Proteins function not only through intramolecular folding and intermolecular complex formation but also through phase transitions driven by intermolecular interactions. Such phases, including liquid-like condensates, amorphous aggregates (AAs), and amyloid fibrils, are linked to distinct biological functions and pathologies. Although the transition from liquid-like condensates to amyloids has been extensively studied, the kinetic relationships between amyloids and other metastable solid states under cell-sized confinement remain unclear, which may hinder the establishment of effective therapeu

One-stop solution for wide polar range compounds: Preparation and application of quaternary ammonium salt molecular cage stationary phase.

The separation and analysis of complex samples with wide polar range on a singular column is always a difficult problem in separation and analysis science. In this study, the RCC3-R molecular cage modified with the quaternary ammonium salt stationary phase (RCC3-GQ@silica) was successfully prepared and applicated in the separation of wide polar range compounds. In the Reverse Phase Liquid Chromatography (RPLC) mode, due to the hydrophobicity and π-π interactions provided by the cyclohexane and benzene rings in the molecular cage structure, this stationary phase demonstrated effective separatio

Disentangling the events that constitute the main phase transition of predominantly neutral lipid bilayers containing anionic lipids by measuring their macroscopic properties.

The thinning of lipid bilayers during their main phase transition, as the outcome of the weakening of van der Waals interactions between their hydrocarbon chains and the hydration of the polar headgroup region, displays a peak at the specific temperature (Tm). The most common technique used for Tm determination is differential scanning calorimetry (DSC), which provides the required information straightforwardly and rapidly, but it cannot provide any details on the molecular-level events that underlie the main phase transition. To uncouple the impact of hydrogen bonding and (de)protonation in t

Enhanced Condensation of RNA Repeats Induced by Terahertz Oscillatory Fields.

Liquid-liquid phase separation (LLPS) of RNA drives the formation of membraneless organelles, and its dysregulation is closely linked to major human diseases, including cancer, neurodegenerative disorders, and various rare genetic diseases. Current strategies for modulating LLPS often require the introduction of exogenous molecules or specific genetic modifications. Here, coarse-grained molecular dynamics (CGMD) simulations suggest that terahertz (THz) oscillatory fields may influence the condensation of pathogenic G4C2 RNA repeats under the simulated conditions. Oscillatory fields at 10 THz a…

NTA-Ni lipids affect membrane phase behavior and fluorescence of lipid-like dyes.

The nitrilotriacetic acid group coordinated with nickel, NTA(Ni), is the standard chelator for purifying fusion proteins containing poly-histidine tags. The development of lipids with NTA(Ni) headgroups has enabled the specific attachment of His-tagged peptides and proteins to model membranes. Recently, these lipids have become essential for the study of surface-mediated protein phase separation and membrane interaction with biomolecular condensates. While this approach is gaining popularity, the intrinsic effects of NTA(Ni) lipids on the properties of the host membrane have been very little s…

RNA-Protein Assemblies: A Review of Biophysical Principles and Coarse-Grained Modeling Approaches.

RNA-protein assemblies are fundamental organizers of eukaryotic cell biology, participating in diverse processes such as ribosome biogenesis in nucleoli, mRNA sequestration in nuclear speckles, and genome packaging in viral ribonucleoprotein (vRNP) complexes. Across these systems, multivalent RNA-protein interactions promote self-organization, contributing directly to liquid-liquid phase separation (LLPS) in nucleoli and nuclear speckles, while facilitating assembly and intermediate states in mature structurally organized vRNPs. This review critically examines the molecular grammar underlying …

In situ growth of 2D nanosheets of terbium-based metal organic frameworks as super-hydrophilic and super-oleophobic (underwater) selective layer on a ceramic support for efficient separation of stabilized oil-in-water emulsion.

The current study aimed to develop an active layer composed solely of 2-dimensional nanosheets of terbium-based metal organic frameworks (Tb-IPA-NH2 MOFs) on a ceramic support via an in situ growth approach. The ultimate aim of the current study was to develop a stable, defect-free active layer composed of 2-dimensional nanosheets that can be readily applied under cross-flow filtration conditions for the separation of surfactant-stabilized oil-in-water emulsions. In addition, the current approach for membrane fabrication focuses on reducing the need for high temperatures, high-polarity solvent

Susceptibility and regulation of biomolecular condensates by solutes.

Metabolites are abundant in cells, where condensation of proteins and nucleic acids can organize cellular contents without membranes. Condensates form and dissolve dynamically in response to diverse cellular processes. Understanding condensate phase behavior using full phase diagrams is hampered by the sheer number of distinct molecular species involved. To circumvent this limitation, we introduce susceptibility, a dimensionless descriptor that quantifies dilute-phase responses to solute perturbations and enables direct comparison across condensates. We measure how three model condensates, ass

Solvent Interaction Analysis: A New Lens for Protein Structure and Diagnostics.

Aqueous two-phase systems (ATPSs) provide a versatile, fully aqueous platform for probing solute-water interactions and protein structure. This review first surveys the diversity and phase behavior of biphasic aqueous systems formed by polymers and salts. We describe how phase diagrams characterize ATPS formation and composition and how both polymer chemistry and salt identity, rather than molecular size alone, govern phase separation by modulating the solvent properties of water. Building on a modified binodal model, we show that phase separation and solute partitioning can be understood in t…

Ultrastable Water-in-Water Emulsions by Addition of Polyion Complex Micelles.

We report the use of polyion complex micelles (PICM) obtained by complexation of poly(sodium acrylate) and poly(N-methyl-2-vinylpyridinium iodide-block-ethylene oxide) as novel stabilizers for water-in-water emulsions consisting in poly(ethylene oxide) (PEO) and dextran mixtures in water. PICM concentrations as low as 1 g/L lead to a strong increase of stability for both PEO-in-dextran and dextran-in-PEO emulsions, as they did not display any macroscopic phase separation after several months. Such long term stability is to our knowledge unprecedented in the literature dedicated to water-in-wat…

LLPS Inflammasome Metabo-Proteostatic Failure Axis in AD: Mechanistic and Translational Insights.

Alzheimer's Disease (AD) is a multifactorial and progressive neurodegenerative disorder for which effective disease-modifying therapies to halt or slow disease progression remain limited. Current pathogenic models primarily focus on the accumulation of amyloid-β (Aβ) and tau proteins; however, these do not fully explain the extensive neuroinflammation, metabolic dysfunction, and impaired protein homeostasis observed in affected individuals. Recent evidence suggests that liquid-liquid phase separation (LLPS) can represent an important biophysical mechanism linking cellular stress, protein aggre…
