A novel bi-enzyme electrochemical biosensor for selective and sensitive determination of methyl salicylate.

An amperometric sensor based on a bi-enzyme modified electrode was fabricated to detect methyl salicylate, a volatile organic compound released by pathogen-infected plants via systemic response. The detection is based on cascadic conversion reactions that result in an amperometric electrochemical signal. The bi-enzyme electrode is made of alcohol oxidase and horseradish peroxidase enzymes immobilized on to a carbon nanotube matrix through a molecular tethering method. Methyl salicylate undergoes hydrolysis to form methanol, which is consumed by alcohol oxidase to form formaldehyde while simult

Diphtheria-toxin based anti-human CCR4 immunotoxin for targeting human CCR4(+) cells in vivo.

CC chemokine receptor 4 (CCR4) has attracted much attention as a promising therapeutic drug target for CCR4(+) tumor cells and Tregs. CCR4 is expressed on some tumor cells such as T-cell acute lymphoblastic leukemia (ALL), adult T-cell leukemia/lymphoma (ATLL), adult peripheral T cell lymphoma (PTCL) and cutaneous T cell lymphoma (CTCL). CCR4 is also expressed on majority of Tregs, mainly effector Tregs. In this study we have successfully developed three versions of diphtheria-toxin based anti-human CCR4 immunotoxins (monovalent, bivalent and single-chain fold-back diabody). Binding analysis b

Secretion of truncated recombinant rabies virus glycoprotein with preserved antigenic properties using a co-expression system in Hansenula polymorpha.

Rabies virus infection remains a serious public health threat in the developing world, where cost-concerns make wide-scale public health interventions impractical. The development of novel and inexpensive ELISA diagnostic antigens is critical in early detection and prevention of complications. The transmembrane glycoprotein (G) of rabies virus (RV) contains an external domain capable of inducing the synthesis of anti-rabies, virus-neutralizing antibodies, in infected or immunized hosts. In our study, the external G domain was synthesized and fused in-frame with a polyhistidine-tag coding seque

Unraveling the Synergistic Inhibition of Human Maltase-Glucoamylase by Baicalein and Acarbose: Integrated Pharmacodynamics and Computational Insights.

Background: Combining natural products with conventional antidiabetic agents to inhibit α-glucosidase activity is an effective strategy for preventing postprandial hyperglycemia. Baicalein, a natural flavonoid with well-documented low toxicity, showed potential synergistic effect with acarbose in diabetic models; however, the synergistic performance and mechanisms of the two agents targeting human maltase-glucoamylase (MGAM) remain unclear. Methods: Recombinant human MGAM-C and MGAM-N were expressed in Pichia pastoris for in vitro inhibition assays. Maltose-loaded mice were used to assess the

High-yield production of immunogenic PV3 virus-like particle in yeast.

The identification of thermally stabilized mutants of all three poliovirus serotypes (PV1, PV2, and PV3) has enabled the development of virus-like particle (VLP)-based next-generation poliovirus vaccines. PV3 stabilized mutant-derived VLPs (sVLPs) have been produced in several recombinant systems through co-expression of mutant P1 polyprotein with native or uncleavable viral protease 3CD, and have shown immunogenicity in animal models. However, their yields remain suboptimal, likely because of intrinsic 3CD toxicity and/or inefficient 3CD-mediated cleavage of P1 into capsid subunits VP0, VP3,

Clinical characteristics, antifungal exposure, and 90-day mortality of PCR-identified candidemia according to presumed species-level fluconazole susceptibility.

Rapid species identification frequently informs early antifungal decision-making before isolate-level antifungal susceptibility testing (AFST) becomes available. Clinicians therefore commonly rely on presumed species-level fluconazole susceptibility to guide initial therapy. Whether these presumed susceptibility categories identify clinically distinct patient populations with different treatment patterns and outcomes remains uncertain. We conducted a retrospective cohort study using TriNetX Research Network (May 2017-November 2023) including adults with PCR-identified candidemia. Patients were

Beyond Bacteria: The Emerging Role of the Gut Virome and Mycobiome in Celiac Disease.

Background/Objectives: Celiac disease (CeD) is a complex autoimmune enteropathy triggered by gluten intake in genetically predisposed individuals. While the bacterial microbiome has been extensively studied in CeD, the role of the gut virome and mycobiome is less well defined. Emerging evidence suggests that viruses and fungi may influence mucosal immunity, intestinal permeability and gluten tolerance, acting as potential environmental cofactors in the onset of this disease. Methods: A narrative review of the literature was conducted to summarize current knowledge of the composition, developme

In Vitro and In Vivo Antifungal Efficacy and Safety of the CaDef2.1G27‑K44 Peptide against the Neglected and Drug-Resistant Pathogen Candida krusei.

The growing threat of fungal infections, driven by increasing drug resistance, has become a major global health concern. Candidiasis, a common human infection, is associated with high mortality, particularly in invasive cases. Among non-albicans Candida (NAC) species, Candida krusei (renamed Pichia kudriavzevii) is of clinical importance because of its intrinsic resistance to fluconazole, complicating treatment options. This study evaluated the antifungal efficacy and safety of the bioinspired peptide CaDef2.1G27‑K44 (CDF-GK) against NAC species, with a specific focus on C. krusei, through a s

Microbial and Metabolic Dynamics during Fermentation of Vinegar with Rye Bread.

Vinegar is a widely consumed acidic fermented product prepared from diverse raw materials. In this study, we investigated vinegar fermentation using rye bread as a substrate, focusing on the comparative dynamics of spontaneous and backslopping fermentation. Two fermentation batches were produced, each in triplicate. One batch was subjected to spontaneous fermentation, while the other was inoculated with a starter culture via backslopping. The pH, microbial communities, and metabolite profiles were analyzed at various stages of fermentation to characterize the fermentation process. In both batc

Wild-type MIC distributions and epidemiological cutoff values for 5-flucytosine and Candida species as determined by EUCAST broth microdilution.

OBJECTIVES: EUCAST has established clinical breakpoints and epidemiological cutoff values (ECOFFs) for Candida spp. However, limited data are available for 5-flucytosine (5-FC). We assessed the in vitro susceptibility of 5-FC against a large collection of clinical Candida species using EUCAST methodology and determined the associated ECOFFs. METHODS: A total of 5622 Candida isolates were collected from patients across the Netherlands between 2008 and 2024. 5-FC MICs were determined using the EUCAST microbroth dilution reference method. Furthermore, MICs were extracted from the EUCAST website.

Computational-guided discovery of UDP-glycosyltransferases for lauryl glucoside production using engineered E. coli.

Defining suitable enzymes for reaction steps in novel synthetic pathways is crucial for developing microbial cell factories for non-natural products. Here, we developed a computational workflow to identify C12 alcohol-active UDP-glycosyltransferases. The workflow involved three steps: (1) assembling initial candidates of putative UDP-glycosyltransferases, (2) refining selection by examining conserved regions, and (3) 3D structure prediction and molecular docking. Genomic sequences from Candida, Pichia, Rhizopus, and Thermotoga, known for lauryl glucoside synthesis via whole-cell biocatalysis,

Intranasal B5 promotes mucosal defence against Actinobacillus pleuropneumoniae via ameliorating early immunosuppression.

Actinobacillus pleuropneumoniae (APP) is an important pathogen of the porcine respiratory disease complex, which leads to huge economic losses worldwide. We previously demonstrated that Pichia pastoris-producing bovine neutrophil β-defensin-5 (B5) could resist the infection by the bovine intracellular pathogen Mycobacterium bovis. In this study, the roles of synthetic B5 in regulating mucosal innate immune response and protecting against extracellular APP infection were further investigated using a mouse model. Results showed that B5 promoted the production of tumour necrosis factor (TNF)-α, i

A thermostable and inhibitor resistant β-glucosidase from Rasamsonia emersonii for efficient hydrolysis of lignocellulosics biomass.

The present study reports a highly thermostable β-glucosidase (GH3) from Rasamsonia emersonii that was heterologously expressed in Pichia pastoris. Extracellular β-glucosidase was purified to homogeneity using single step affinity chromatography with molecular weight of ~ 110 kDa. Intriguingly, the purified enzyme displayed high tolerance to inhibitors mainly acetic acid, formic acid, ferulic acid, vanillin and 5-hydroxymethyl furfural at concentrations exceeding those present in acid steam pretreated rice straw slurry used for hydrolysis and subsequent fermentation in 2G ethanol plants.
