Preparation, Structure Characteristics, Antioxidant, Antitumor, and Immunomodulatory Activity of Polysaccharides From Rheum officinale Baill. and Their Derivatives.

In this study, three polysaccharides (RHC, RHH, and RHA) were sequentially extracted from Rheum officinale Baill., under different conditions, and their sulfated, phosphorylated, and selenated derivatives were successfully synthesized. Results indicated that the sequential extraction method effectively separated polysaccharides with distinct structures. According to their charge properties, these three polysaccharides were identified as acidic polysaccharides with different structural characteristics. Activity assays showed that the phosphorylation modification of polysaccharides significantly

Selective requirement for CD40-CD154 in drug-induced type 1 versus type 2 responses to trinitrophenyl-ovalbumin.

CD154 is transiently expressed by activated T cells and interacts with CD40 on B cells, dendritic cells, macrophages, and monocytes. This costimulatory receptor-ligand couple seems decisive in Ag-driven immune responses but may be differentially involved in type 1 vs type 2 responses. We studied the importance of CD40-CD154 in both responses using the reporter Ag popliteal lymph node assay in which selectively acting drugs generate clearly polarized type 1 (streptozotocin) or type 2 (D-penicillamine, diphenylhydantoin) responses to a constant coinjected Ag in the same mouse strain. Oxidative modification of apolipoprotein E in human very-low-density lipoprotein and its inhibition by glycosaminoglycans.

The mechanism of metal ion-catalyzed oxidative modification of apolipoprotein E (apoE) in human very-low-density lipoprotein (VLDL) and its inhibition by glycosaminoglycan (GAG) was investigated in vitro. The VLDL oxidation catalyzed by Cu2+ led to the lipid peroxidation, the formation of aggregates, and covalent modification of apoE. The modified apoE lost heparin-binding activity. These results suggest that the lipid peroxidation of VLDL and modification of apoE cause impairment of lipid uptake by cells and deposit the oxidized lipids in the tissues. The lipid peroxidation and oxidative modi
