Immune function was studied in four patients (two girls and two boys, aged 30 months to 24 years) with documented Bloom's syndrome. Three patients had a decreased serum concentration of at least one subclass of immunoglobulins. All had normal or elevated proportions of circulating B cells but two of them had a decreased proportion of CD4-positive helper-inducer T cells. We consistently found a severely impaired in vitro proliferative lymphocyte response to the plant lectin pokeweed mitogen (PWM). This could not be overcome by using suboptimal or supraoptimal doses of PWM, or by adding recombin

In this commentary, we highlight the findings described in a recent paper regarding the mechanism of H2S regulation of macroautophagy/autophagy in mammalian cells and discuss the similarities/divergencies with plant cells. The main outcome is that the posttranslational modification of thiol groups of cysteine residues to form persulfides is a conserved molecular mechanism.

Non-model organisms often lack the methodological and technological tools that are available for model systems. This is particularly true for plant-parasitic nematodes. Despite causing significant agricultural losses worldwide, their mechanisms of parasitism remain incompletely understood. One major limitation is the difficulty in validating the spatial expression of genes involved in pathogenesis, known as effectors. In situ hybridization (ISH) remains one of the most accessible and cost-effective approaches for gene expression analysis. However, traditional chromogenic ISH on fragmented nema

Bisphenol A (BPA) is one of the most used plasticizers and a known endocrine disrupting chemical. In plants, BPA is easily conjugated to form conjugates such as glycosyl BPA, and humans can be exposed to such conjugates when ingesting BPA-contaminated plant foods. This study considered the potential of deconjugation of glycosyl BPA in a simulated human gastrointestinal tract system and evaluated the subsequent changes in biological activity using the Caco-2 colon cell model. Glycosyl BPA was found in both soil and lettuce with concentrations ranging from not detected (ND) to 82.51 ng/g dry wei

Numerous studies have clarified the impacts of magnesium (Mg) on leaf photosynthesis from the perspectives of protein synthesis, enzymes activation and carbohydrate partitioning. However, it still remains largely unknown how stomatal and mesophyll conductances (gs and gm, respectively) are regulated by Mg. In the present study, leaf gas exchanges, leaf hydraulic parameters, leaf structural traits and cell wall composition were examined in rice plants grown under high and low Mg treatments to elucidate the impacts of Mg on gs and gm. Our results showed that reduction of leaf photosynthesis unde

Brassinosteroids (BRs) are essential hormones for plant growth and development that are perceived at the plasma membrane by a group of Leucine-Rich Repeat Receptor-Like Kinases (LRR-RLKs) of the BRASSINOSTEROID INSENSITIVE 1 (BRI1) family. The BRI1 receptor was first discovered by genetic screenings based on the dwarfism of BR-deficient plants. There are three BRI1 homologs, named BRI1-like 1, 2 and 3 (BRLs), yet only BRL1 and BRL3 behave as functional BR receptors. Whereas the BRI1 pathway operates in the majority of cells to promote growth, BRL receptor signaling operates under specific spat

BIR1 is a receptor-like kinase that functions as a negative regulator of basal immunity and cell death in Arabidopsis. Using Arabidopsis thaliana and Tobacco rattle virus (TRV), we investigate the antiviral role of BIR1, the molecular mechanisms of BIR1 gene expression regulation during viral infections, and the effects of BIR1 overexpression on plant immunity and development. We found that SA acts as a signal molecule for BIR1 activation during infection. Inactivating mutations of BIR1 in the bir1-1 mutant cause strong antiviral resistance independently of constitutive cell death or SA defens

Cell expansion is crucial for organ morphogenesis in multicellular organisms. Apoplast acidification triggers plant cell expansion. Plant hormones and transcription factors such as TEOSINTE BRANCHED, CYCLOIDEA, and PROLIFERATING CELL NUCLEAR ANTIGEN BINDING FACTORs (TCPs) control cell expansion. However, details regarding the regulatory mechanism of cell expansion for organ morphogenesis remain unclear. In this study, we used molecular, biochemical, cellular, genetic, atomic force microscopy, and tensile testing analyses and showed that miR319-targeted TCPs integrated cell expansion with organ

Terrestrial carbon uptake under climate change is significantly constrained by nitrogen (N) availability. While plants can alleviate this limitation through flexible leaf N allocation, the global patterns of such partitioning remain poorly understood. Here, by analyzing a global dataset of 520 seed plant species spanning 98 families, we find that plants averagely allocate 38.7% of leaf N to photosynthetic components (including Rubisco, bioenergetics, and light‑capture machinery) and 13.1% to cell walls. The leaf N allocation pattern exhibits a distinct evolutionary shift: early-diverging plant

The previously reported Arabidopsis dominant gain-of-function mutant accelerated cell death6-1 (acd6-1) shows spontaneous cell death and increased disease resistance. acd6-1 also confers increased responsiveness to the major defense signal salicylic acid (SA). To further explore the role of ACD6 in the defense response, we cloned and characterized the gene. ACD6 encodes a novel protein with putative ankyrin and transmembrane regions. It is a member of one of the largest uncharacterized gene families in higher plants. Steady state basal expression of ACD6 mRNA required light, SA, and an intact

The hypersensitive response (HR) displayed by resistant plants against invading pathogens is a prominent feature of plant-pathogen interactions. The Arabidopsis hypersensitive response like lesions1 (hrl1) mutant is characterized by heightened defense responses that make it more resistant to virulent pathogens. However, hrl1 suppresses avirulent pathogen-induced HR cell death. Furthermore, the high PR-1 expression observed in hrl1 remains unaltered after avirulent and virulent pathogen infections. The suppressed HR phenotype in hrl1 is observed even when an elicitor is expressed endogenously f

Plant-associated yeasts modulate host immunity to promote or prevent disease. Mechanisms of yeast perception by the plant innate immune system remain unknown, with progress hindered by lack of a model system with the model plant Arabidopsis thaliana (Arabidopsis). A yeast strain of Taphrina tormentillae, named M11, was previously isolated from wild Arabidopsis. Taphrina have been found on many non-host plants, and their complex ecology remains understudied. Here, the interaction of M11 with Arabidopsis was characterised. Infection of Arabidopsis with the birch pathogen T. betulina, used as a n

Presence of the cell wall and the lack of streamlined pathways for cellular delivery of external agents into plants is a core challenge of plant biotechnology and crop engineering development. However, both viral and bacterial transmission have their own restrictions and the few non-heavy metal nanodelivery platforms require external forces for tissue penetration. Such dependency limits any high-throughput application considering the large plant numbers to be treated in the field or even laboratory exercises. Herein, we demonstrate Aspartic acid (Asp) decorated poly(ethylene glycol)-block-poly

The genetic transformation of plant cells is critically dependent on the availability of efficient selectable marker gene. Sulfonamides are herbicides that, by inhibiting the folic acid biosynthetic pathway, suppress the growth of untransformed cells. Sulfonamide resistance genes that were previously developed as selectable markers for plant transformation were based on the assumption that, in plants, the folic acid biosynthetic pathway resides in the chloroplast compartment. Consequently, the Sul resistance protein, a herbicide-insensitive dihydropteroate synthase, was targeted to the chlorop…

1-Aminocyclopropane-1-carboxylic acid (ACC) is a precursor molecule of ethylene whose concentration is elevated in the plant subjected to biotic and abiotic stress. Several soil microorganisms are reported to produce ACC deaminase (ACCd) which degrades ACC thereby reducing stress ethylene in host plants. This study is aimed to apply ACCd producing beneficial rhizobacteria to improve biochemical parameters and cell wall properties of Panicum maximum exposed to salt and drought stress, focusing on bioethanol production. Thirty-seven ACCd producing bacteria isolated from rhizospheric soil of fiel

Their sessile lifestyle means that plants have to be exquisitely sensitive to their environment, integrating many signals to appropriate developmental and physiological responses. Stimuli ranging from wounding and pathogen attack to the distribution of water and nutrients in the soil are frequently presented in a localized manner but responses are often elicited throughout the plant. Such systemic signaling is thought to operate through the redistribution of a host of chemical regulators including peptides, RNAs, ions, metabolites, and hormones. However, there are hints of a much more rapid co

Lithium (Li) toxicity in plants is, at a minimum, a function of Li(+) concentration, exposure time, species and growth conditions. Most plant studies with Li(+) focus on short-term acute exposures. This study examines short- and long-term effects of Li(+) exposure in Arabidopsis with Li(+) uptake studies and measured shoot mRNA transcript abundance levels in treated and control plants. Stress, pathogen-response and arabinogalactan protein genes were typically more up-regulated in older (chronic, low level) Li(+)-treatment plants and in the much younger plants from acute high-level exposures. The prevalence of metabolic disorders characterized by chronic inflammation has been on a sharp rise for decades. As such, tools that address metabolic and inflammatory dysregulation are of great importance. Plant stanols are well-known for reducing intestinal cholesterol absorption and may also have direct anti-inflammatory effects. In this study, our aim was to investigate to what extent the benefits of dietary plant stanol supplementation depend on dietary cholesterol intake in an experimental mouse model for cholesterol-induced metabolic inflammation. Here, we used Ldlr-/- mice transplante
