Genome-Wide Identification of the APRR2 Gene Family and Rind Color Trait Analysis in Zucchini (Cucurbita pepo).

Rind color is an important quality trait in zucchini (Cucurbita pepo). As a core transcription factor in plant pigment biosynthesis, APRR2 p … photomorphogenesis. Crucially, transcriptome analysis during rind development (0 and 10 days after pollination) in green (GR) and white (WR) rind lines demonstrated profound functional divergence. Expression clusters indicated temporal shifts in metabolism and enriched "circadian rhythm-plant" and "photosynthesis" pathways in WR at 0 DAP. Specific APRR2 gene …

Speed breeding: protocols, application and achievements.

One of the limiting factors in breeding and genetic research is the time required to develop pure lines. This is due, on the one hand, to the prolonged vegetative period of a single generation and, on the other hand, to the specifics of inbreeding, which typically requires 4-6 consecutive generations of self-pollination in plant material. Researchers have always sought approaches that enable the rapid development of homozygous plant lines. Consequently, methods such as greenhouse cultivation during the autumn-winter period, single-seed descent, shuttle breeding, embryo culture, and doubled hap…

Germination of pollen grains in the oesophagus of individuals with eosinophilic oesophagitis.

BACKGROUND: Eosinophilic oesophagitis (EoE) is characterized by oesophageal dysfunction and, histologically, by eosinophilic inflammation. There is no a clear aetiologic treatment. EoE exacerbations are often seasonal. We hypothesized that the inflammatory response of the oesophageal mucosa in patients with high levels of antibodies to pollen allergens and worsened seasonal EoE might be due to swallowing airborne pollen and the intrusion into the oesophageal mucosa of pollen allergens and pollen tubes, which encounter a pH and humidity resembling the stigma at pollination. OBJECTIVE: The aim …

Molecular characterization of emerging bacterial communities associated with honey bees.

Honey bees are vital pollinators essential for ecosystems and global agriculture, but their populations are declining due to pressures caused by pests. This study reports the presence of Escherichia coli, Klebsiella pneumoniae, Proteus terrae, and Pseudomonas spp. in four different honey bee species in Pakistan for the first time, uncovering a previously neglected aspect of honey bee microbiology. From January 2023 to June 2024; 1190 honey bees were collected from four districts in Khyber Pakhtunkhwa and they were identified. Molecular screening based on PCR targeting the microbial 16 S rDNA r

Semi-Quantitative Monitoring of Plant-Arthropod Interactions by eDNA Metabarcoding of Individual Flowers and Leaves.

Environmental DNA (eDNA) analysis has revolutionized our ability to study plant-arthropod interaction diversity. However, the method has one main limitation: while it accurately recovers the presence of individual species, it has limited quantitative recording capabilities. Recent work suggests that this problem can be overcome by processing replicate eDNA samples at individual sites. The naive occupancy of a species within a replicate sample approximates its relative abundance or its interaction strength with the sampled matrix. However, this approach will lead to a significant increase in th

Interface of Climate and Environmental Change on Food Systems-A Report from the "Agriculture and Diet: Value Added for Nutrition, Translation, and Adaptation in a Global Ecology" (ADVANTAGE) Project Working Group 3.

As the climate changes, investigation at the intersection of climate and food systems is critical to understand broader impacts on human nutrition and health. This work seeks to 1) identify reciprocal interactions of climate and environmental change (CEC) and food systems, 2) illuminate research gaps in CEC interactions with food systems, and 3) identify paths to respond to and mitigate the impact of CEC on food systems. We discuss CEC and food system interactions via 4 pathways. Food systems are influenced by CEC through shifting climate normals and altered instances of extreme events. The ph

Enhancing agronomic traits and bioactive compound accumulation in Thai Andrographis paniculata via EMS-induced variation.

BACKGROUND: Andrographis paniculata is a self-pollinating medicinal plant with limited natural genetic diversity, which restricts the development of improved cultivars with desirable agronomic traits and stable accumulation of key bioactive compounds, particularly andrographolide (AP1). To overcome this limitation, ethyl methanesulfonate (EMS) mutagenesis was applied to native Thai germplasm to induce phenotypic and phytochemical variation and to generate novel genetic resources for breeding improvement and gene function analysis. RESULTS: Optimal EMS treatment conditions (1.0% and 1.5% EMS fo

Faba bean populations already contain the inbreds needed for breeding.

Highly inbred individuals are naturally present in faba bean populations and should be used for rapid and cost-efficient development of line and synthetic cultivars. Producing inbred lines in faba bean (Vicia faba L.) breeding is, although crucial, a long, costly and laborious process due to the partial allogamy of the crop. However, in any genetically diverse and open pollinating faba bean population, this particular reproduction system leads to the so-called identity disequilibrium, resulting in a significant proportion of individuals being highly inbred. This study presents a cost-efficient

Effects of Atmospheric Pollutants on Volatile-Mediated Insect Ecosystem Services.

Primary and secondary atmospheric pollutants, including carbon monoxide (CO), carbon dioxide (CO2), nitrogen oxides (NOx), ozone (O3), sulphur dioxide (SO2) and particulate matter (PM2.5/PM10) with associated heavy metals (HMs) and micro- and nanoplastics (MPs/NPs), have the potential to influence and alter interspecific interactions involving insects that are responsible for providing essential ecosystem services (ESs). Given that insects rely on olfactory cues for vital processes such as locating mates, food sources and oviposition sites, volatile organic compounds (VOCs) are of paramount im

Discovery of Novel Small Molecule Growth Inhibitors to Manage Pseudomonas Leaf Spot Disease on Peppers (Capsicum sp.).

Pseudomonas leaf spot (PLS) disease in peppers caused by Pseudomonas syringae pv. syringae (Pss) is an emerging seedborne phytopathogen. Pss infection can severely reduce the marketable yield of peppers in favorable environmental conditions and cause significant economic losses. The intensive use of copper-sulfate and streptomycin-sulfate to control PLS and other bacterial diseases is associated with antimicrobial-resistant Pss strains, making these control methods less effective. So, there is an urgent need to develop novel antimicrobials effective against Pss in peppers. Several studies, inc

Combined effect of a neonicotinoid insecticide and a fungicide on honeybee gut epithelium and microbiota, adult survival, colony strength and foraging preferences.

Fungicides, insecticides and herbicides are widely used in agriculture to counteract pathogens and pests. Several of these molecules are toxic to non-target organisms such as pollinators and their lethal dose can be lowered if applied as a mixture. They can cause large and unpredictable problems, spanning from behavioural changes to alterations in the gut. The present work aimed at understanding the synergistic effects on honeybees of a combined in-hive exposure to sub-lethal doses of the insecticide thiacloprid and the fungicide penconazole. A multidisciplinary approach was used: honeybee mor

A novel method for the detection and diagnosis of virus infections in honey bees.

In terms of infectious diseases caused by a variety of microorganisms, the ability to promptly and accurately identify the causative agents is the first step on the path to all types of effective management of such infections. Among the various factors that are affecting global bee health, viruses have often been linked to honey bee colony losses and they pose a serious threat to the fraction of agriculture that depends on the service of pollinators. Over the past few decades, PCR-based molecular methods have provided powerful tools for rapid, specific, and sensitive detection and the quantifi

Phylogenetic analyses of antibiotic-producing Streptomyces sp. isolates obtained from the stingless-bee Tetragonisca angustula (Apidae: Meliponini).

Many insects have been associated with actinobacteria in protective symbiosis where antimicrobial metabolites inhibit host pathogens. However, the microbiota of neotropical insects such as the stingless-bee Tetragonisca angustula is poorly explored. T. angustula is a meliponid bee widely distributed in Latin America, its honey is traditionally exploited because of its ethno-pharmacological properties and its antimicrobial activity has been demonstrated. Also, the well-structured nest of this species allows exploration of the microbiota of its different components. Even though Streptomyces spp.

Honey Bee Exposure to Pesticides: A Four-Year Nationwide Study.

Pollinators, including honey bees, are responsible for the successful reproduction of more than 87% of flowering plant species: they are thus vital to ecosystem health and agricultural services world-wide. To investigate honey bee exposure to pesticides, 168 pollen samples and 142 wax comb samples were collected from colonies within six stationary apiaries in six U.S. states. These samples were analyzed for evidence of pesticides. Samples were taken bi-weekly when each colony was active. Each apiary included thirty colonies, of which five randomly chosen colonies in each apiary were sampled fo

Micro-CT imaging of denatured chitin by silver to explore honey bee and insect pathologies.

BACKGROUND: Chitin and cuticle coatings are important to the environmental and immune defense of honey bees and insect pollinators. Pesticides or environmental effects may target the biochemistry of insect chitin and cuticle coating. Denaturing of chitin involves a combination of deacetylation, intercalation, oxidation, Schweiger-peeling, and the formation of amine hydrochloride salt. The term "denatured chitin" calls attention to structural and property changes to the internal membranes and external carapace of organisms so that some properties affecting biological activities are diminished.

Genetic diversity and population structure of Coffea arabica in Saudi Arabia revealed by whole-genome resequencing.

Gene banks are essential for conserving plant genetic resources, yet limited genomic characterization constrains their effective use in breeding. Coffea arabica L., an economically important crop in Saudi Arabia, remains poorly characterized at the genome-wide level. We performed whole-genome resequencing of 78 Saudi C. arabica accessions conserved at the Ministry of Environment, Water and Agriculture (MEWA) Seed Center and PGR Bank, generating 86 sequencing libraries, including eight technical replicates. Reads were aligned to the Cara_r6 reference genome, yielding 468, 133 variants (412, 043
