Mineralocorticoid Receptor Antagonists for Treatment of Hypertension and Heart Failure.

Spironolactone and eplerenone are both mineralocorticoid-receptor antagonists. These compounds block both the epithelial and nonepithelial actions of aldosterone, with the latter assuming increasing clinical relevance. Spironolactone and eplerenone both affect reductions in blood pressure either as mono- or add-on therapy; moreover, they each afford survival benefits in diverse circumstances of heart failure and the probability of renal protection in proteinuric chronic kidney disease. However, as use of mineralocorticoid-blocking agents has expanded, the hazards inherent in taking such drugs

Diversity of Potassium Channel Ligands: Focus on Scorpion Toxins.

Potassium (K+) channels are a widespread superfamily of integral membrane proteins that mediate selective transport of K+ ions through the cell membrane. They have been found in all living organisms from bacteria to higher multicellular animals, including humans. Not surprisingly, K+ channels bind ligands of different nature, such as metal ions, low molecular mass compounds, venom-derived peptides, and antibodies. Functionally these substances can be K+ channel pore blockers or modulators. Representatives of the first group occlude the channel pore, like a cork in a bottle, while the second gr

Simultaneous quantification of voriconazole and posaconazole in human plasma by high-performance liquid chromatography with ultra-violet detection.

A sensitive and selective high-performance liquid chromatographic (HPLC) method with ultra-violet detection has been developed and validated for the simultaneous determination of posaconazole and voriconazole, two systemic anti-fungal agents. An internal standard diazepam was added to 100 microL of human plasma followed by 3 mL of hexane-methylene chloride (70:30, v/v). The organic layer was evaporated to dryness and the residue was reconstituted with 100 microL of mobile phase before being injected in the chromatographic system. The compounds were separated on a C8 column using sodium potassi

Contact allergy to gold in dental patients.

Gold sodium thiosulphate (GSTS) in patch test series commonly yields positive reactions. In this study of dental patients, the clinical relevance of these positive reactions, the suitability of gold compounds as patch test materials, and value of the lymphocyte proliferation test in gold contact allergy, were evaluated. The frequency of positive patch test reactions to GSTS was 12.4% in the dental series. Fifty-two patients, only two of whom were male, were studied. Thirteen (25%) had had symptoms from jewellery or dental restorations. However, in most cases gold allergy was subclinical. A 10%

The Significant Therapeutic Effects of Chinese Scorpion: Modern Scientific Exploration of Ion Channels.

Chinese scorpion (CS), a traditional animal-based medicine used for over a millennium, has been documented since AD 935-960. It is derived from the scorpion Buthus martensii Karsch and is used to treat various ailments such as stroke, epilepsy, rheumatism, and more. Modern research has identified the pharmacological mechanisms behind its traditional uses, with active components like venom and proteins showing analgesic, antitumor, antiepileptic, and antithrombotic effects. Studies reveal that CS affects ion channels, crucial for cellular functions, through interactions with sodium, potassium,

Effects of hypervitaminosis of vitamin B3 on silkworm biology.

A high-dose of vitamin B(3) in silkworm diet interrupts larval feeding and normal growth. High mortality of larvae occurs during molting and they cannot complete this process normally. Also the larvae exhibit nicotinamide hypervitaminosis symptoms such as immobility, dyspepsia, darkening of the skin, inability to excrete normally, exerting brownish fluid from anus and swelling of rectal muscles. Maximum larval weights in 1, 2 and 3 g/l treatments were 2.9, 1.6 and 1.2 g respectively, while maximum larval weight in the control was 5.6 g. Larval stage compared to control had increased 18, 26 and

Chemical oxidation of N-hydroxyguanidine compounds. Release of nitric oxide, nitroxyl and possible relationship to the mechanism of biological nitric oxide generation.

N omega-Hydroxy-L-arginine was found to cause vasodilation in arginine-depleted rabbit aorta. It is, therefore, likely to be a biosynthetic intermediate in the conversion of arginine to nitric oxide in this tissue. N-Hydroxyalkylguanidine compounds, including N omega-hydroxy-L-arginine were oxidized with various oxidizing agents and examined for their ability to release nitric oxide. All oxidizing agents tested were capable of oxidizing the N-hydroxyguanidine function but only lead tetra-acetate (Pb(OAc)4) and potassium ferricyanide/hydrogen peroxide (K3FeCN6/H2O2) were capable of generating s

Hypokalemic paralysis as an initial presentation of Sjogren syndrome.

Sjogren syndrome (SS) is a systemic autoimmune disorder with predominant exocrine gland involvement leading to sicca symptoms. Among extraglandular manifestations, renal disease is the most common. Tubular interstitial nephritis and renal tubular acidosis (RTA) are the common presentations. Mild hypokalemia associated with distal RTA is common in SS, however, severe hypokalemia causing paralysis is unusual. We report the case of a 26-year-old female who presented with hypokalemic paralysis. On evaluation, distal RTA was diagnosed. Further evaluation showed positive SS-a/SS-b antibodies in high

Enhanced emulsifying properties of octenyl succinic anhydride starch by homogeneous modification in KOH/urea aqueous solution.

Octenyl succinic anhydride starch (OSA-starch) is a widely used food-grade emulsifier. However, the heterogeneous synthesis process resulting from insoluble starch granules limits the functional properties of OSA-starch (HeOSA-starch). In this study, KOH/urea solution was used to dissolve starch to prepare OSA-starch via homogeneous modification (HoOSA-starch), and its emulsifying properties were investigated. Homogeneous modification significantly improved the degree of substitution and dispersibility of OSA-starch. Emulsions stabilized by HoOSA-starch exhibited superior stability than those

KOH-activated carbon from pomelo peels for CO2 adsorption: influence of activation and adsorption process parameters.

This study examines pomelo peel-based activated carbon (PMAC) as a promising and sustainable carbon dioxide (CO2) adsorbent through a systematic exploration of the influence of activation and adsorption process parameters. Investigations into the effects of the activation parameters revealed that the best potassium hydroxide (KOH) activation conditions were achieved with a KOH to pomelo peel-based char mass ratio of 3:1, an activation time of 1.5 h, and an activation temperature of 700 °C. The resulting activated carbon, designated as PMAC-3-1.5-7 (where 3, 1.5, and 7 represent the KOH ratio,

Vitamin C Mitigates Potassium Perchlorate Exposure-Induced Disruption of Spermatogenesis in Medaka.

Endocrine-disrupting chemicals (EDC) can cause metabolic and reproductive impairments. It is essential to develop strategies to mitigate EDC-exposure-induced endocrine disruption. Potassium perchlorate (KClO4), widely used in industrial and military applications, is an emerging environmental contaminant known to disrupt thyroid function. Its potential impact on male reproductive health remains underexplored. In this study, we investigated the testicular impairments caused by chronic KClO4 exposure in medaka (Oryzias latipes) and examined whether KClO4-induced testicular impairment can be mitig

Biochar, zeolite, and ferric chloride effectively separate phosphorus and nitrogen (plus potassium) in swine manure: A coagulation-flocculation-sedimentation approach.

We studied the effectiveness of different coagulants (0.15 M FeCl3, 0.40 M HCl, and 2.5% H3PO4) in separating key plant macronutrients (N as NH4+, P, and K) from swine manure into solid and liquid fractions for their sustainable use in agriculture. This study assesses how these coagulants impact nutrient recovery and separated fraction's characteristics, aiming to improve the solid-liquid separation process using biochar and zeolite. The FeCl3 treatment, biochar, and zeolite showed optimum nutrient recovery among the tested coagulants for solid (93% of total P) and liquid (90% and 50% of total

Crown-Ether-Based Artificial K+ Selective Ionic Filter.

In this study, we have successfully synthesized bis (cholesterol-dibenzo-18-crown-6-ether)-pillar[5]arene compound 1 through a click reaction, which could spontaneously insert into lipid bilayers to form ion channel due to the membrane anchor cholesterol group and show significant transport activity of K+ superior to Na+, with a permeability ratio of K+/Na+ equal to 4.58. Compound 1 two crown ether modules act as selective filters similar to natural K+ channel, which are determined to 1 : 2 binding stoichiometry to K+ by Job's plot and NMR titration. This structurally unambiguously unimolecule

Interplay between edaphic and climatic factors unravels plant and microbial diversity along an altitudinal gradient.

Altitude influences biodiversity and physiochemical soil attributes in terrestrial ecosystems. It is of immense importance to know the patterns of how interactions among climatic and edaphic factors influence plant and microbial diversity in various ecosystems, particularly along the gradients. We hypothesize that altitudinal variation determines the distribution of plant and microbial species as well as their interactions. To test the hypothesis, different sites with variable altitudes were selected. Analyses of edaphic factors revealed significant (p < 0.001) effects of the altitude. Soil am

A Novel PIFA/KOH Promoted Approach to Synthesize C2-arylacylated Benzothiazoles as Potential Drug Scaffolds.

To discover an efficient and convenient method to synthesize C2-arylacylated benzothiazoles as potential drug scaffolds, a novel [bis(trifluoroacetoxy)iodo]benzene(PIFA)/KOH synergistically promoted direct ring-opening C2-arylacylation reaction of 2H-benzothiazoles with aryl methyl ketones has been developed. Various substrates were tolerated under optimized conditions affording the C2-arylacylation products in 70-95% yields for 38 examples. A plausible mechanism was also proposed based on a series of controlled experiments.

Effects of the gaseous signalling molecule nitroxyl (HNO) on myenteric neurons governing intestinal motility.

OBJECTIVES: The main function of myenteric neurons is the control of gut motility. As we recently showed that nitroxyl (HNO) induces intestinal smooth muscle relaxation, it was of interest to evaluate the effects of this signalling molecule on myenteric neurons in order to distinguish its properties in regard to myocytes. METHODS: Myenteric neurons isolated from the ileum of 4-10 days old rats were used. HNO-induced changes in intracellular concentration of Ca2+ or membrane potential and ion currents were measured using the Ca2+-sensitive fluorescent dye fura-2 AM or by electrophysiological w

Polyphosphate accumulation dynamics in a population of Synechocystis sp. PCC 6803 cells under phosphate overplus.

In this study, a simple and rapid DAPI-based protocol was developed and optimized to visualize polyphosphates (polyPs) in the cyanobacterium Synechocystis sp. PCC 6803. The optimum dye concentration and incubation time were determined, and formaldehyde fixation was shown to significantly improve polyP detection in Synechocystis cells. Using the developed protocol, for the first time, it was shown that 80% of Synechocystis cells under phosphate overplus were able to accumulate phosphorus as polyP 3 min after the addition of K2HPO4. After 1 h, the number of cells with polyP began to decrease, an
