Junctophilin-2 abundance is unaltered in human heart failure samples with disrupted T-tubules and contractility.

T-tubules are structural components of the cardiomyocyte plasma membrane that are imperative for efficient excitation-contraction coupling and cardiac contraction. Junctophilin-2 (JPH2) has been postulated in multiple preclinical models to play a key role in T-tubule structure development, maintenance, and organisation by anchoring the sarcoplasmic reticulum to the sarcolemma and maintaining the spatial distance between excitation contraction coupling proteins. Animal studies have shown that JPH2 downregulation leads to T-tubule dysfunction and impaired cardiac contractility, and upregulation

Carrier-Free Berberine/Nitidine Chloride Self-Assembled Nanoparticles Induce Ferroptosis to Overcome Bortezomib Resistance in Multiple Myeloma.

Therapeutic relapse driven by bortezomib resistance represents a formidable clinical barrier in the management of multiple myeloma (MM). Here, a carrier-free, supramolecular nanoplatform was engineered through the spontaneous co-assembly of two natural alkaloids, berberine (BBR) and nitidine chloride (NC), termed BBR/NC-SAPs, to counter this malignancy. Driven by cooperative π-π stacking and van der Waals forces, BBR/NC-SAPs display superior anti-MM efficacy and optimized biosafety as compared to the free-drug combination. Quantitative proteomics and functional landscapes showed that BBR/NC-SA

Diazonium-Enabled Post-Synthetic Construction of Enantiomeric Single-Atom Nanoagents Used for Alzheimer's Disease Treatment.

Alzheimer's disease (AD) is driven by a self-amplifying interplay between oxidative stress and neuroinflammation, in which mitochondrial DNA (mtDNA) leakage-induced activation of the cGAS-STING pathway plays a central role. Strategies that solely eliminate reactive oxygen species (ROS) are insufficient to suppress downstream inflammatory cascades. Here, we report a pair of chiral dual-functional single-atom nanoagents that can simultaneously scavenge ROS and sequester leaked mtDNA. A diazonium-enabled post-synthetic modification strategy is employed to graft benzoic acid linkers onto a single-

Wnt signaling dysregulation drives Stargardt disease pathogenesis.

Stargardt disease, the most common inherited juvenile macular degeneration, is primarily caused by ABCA4 mutations, yet the cellular origin of pathology remains debated. Here, we identify a critical role for Wnt signaling dysregulation, specifically non-canonical Wnt pathway suppression, in driving retinal pigment epithelium (RPE) dysfunction as a central mechanism of disease. Using patient-derived iPSC-RPE harboring distinct ABCA4 mutations, we demonstrate that ABCA4 loss disrupts RPE hexagonal morphology, reduces pigmentation, and aberrantly activates neural retinal differentiation. Mechanis

An Emerging Paradigm for Safer and Faster Recovery: A Narrative Review on Opioid Sparing Anesthesia in Surgery.

Opioid-free anesthesia (OFA) replaces opioid use with many non-opioid drugs, such as dexmedetomidine, lidocaine, esketamine, regional techniques, and enhanced recovery after surgery (ERAS)-aligned strategies to control pain while minimizing opioid-related adverse effects. Across surgical procedures, we conducted a narrative review of the literature, which showed that OFA is consistently associated with lower postoperative nausea and vomiting, faster recovery of gastrointestinal function, and reduced rescue opioid use, with similar post-anesthesia care unit (PACU) stay and pain scores in many t

Self-Fluorescent Lone Tryptophan Nanoparticles as Theranostic Agents Against Alzheimer's Disease.

Aggregation of β-amyloid (Aβ42) peptide in the neural extracellular space leads to cellular dysfunction, resulting in Alzheimer's disease (AD). The hydrophobic core of the amyloidogenic Aβ42 peptide contains aromatic residues that play an important role in the self-assembly and subsequent aggregation of the peptide. Hence, targeting these hydrophobic core residues by potent low molecular agents can be a promising therapeutic approach toward AD. In the current work, we have developed self-fluorescent solo tryptophan nanoparticles (TNPs) as nanotheranostic systems against AD. We demonstrated tha

Leptospirosis: a neglected tropical zoonotic infection of public health importance-an updated review.

Leptospirosis is a zoonotic and waterborne disease worldwide. It is a neglected, reemerging disease of global public health importance with respect to morbidity and mortality both in humans and animals. Due to negligence, rapid, unplanned urbanization, and poor sanitation, leptospirosis emerges as a leading cause of acute febrile illness in many of the developing countries. Every individual has a risk of getting infected as domestic and wild animals carry leptospires; the at-risk population varies from the healthcare professionals, animal caretakers, farmers and agricultural workers, fishermen

Immune dysregulation may contribute to disease pathogenesis in spinal muscular atrophy mice.

Spinal muscular atrophy (SMA) has long been solely considered a neurodegenerative disorder. However, recent work has highlighted defects in many other cell types that could contribute to disease aetiology. Interestingly, the immune system has never been extensively studied in SMA. Defects in lymphoid organs could exacerbate disease progression by neuroinflammation or immunodeficiency. Smn depletion led to severe alterations in the thymus and spleen of two different mouse models of SMA. The spleen from Smn depleted mice was dramatically smaller at a very young age and its histological architect

The novel human β-defensin 114 regulates lipopolysaccharide (LPS)-mediated inflammation and protects sperm from motility loss.

Lipopolysaccharide (LPS) is an important pathological factor involved in serious inflammatory diseases and male reproductive impairments. Emerging evidence demonstrates that antimicrobial peptides possess protective activity in response to LPS-induced inflammation. However, the LPS-binding and/or immunosuppressive activity of β-defensins (DEFBs) has been underestimated. In the present work, we characterized a novel human defensin, DEFB114, which was expressed predominantly in the epididymis and gingival cells at the RNA level. Homogenous recombinant DEFB114 peptides were prepared and character

In vivo delivery of DN:REST improves transcriptional changes of REST-regulated genes in HD mice.

Current therapeutic strategies for Huntington's disease (HD) are focused on symptom management of disease progression. Transcriptional dysregulation is one of the major characteristics in HD. REST is a transcriptional repressor that silences gene expression through binding to RE1/NRSE sites found in the regulatory regions of numerous neuronal genes. Dysregulation of REST and its targeted genes has been reported in different cell and mouse HD models, as well as in biopsies from human patients. In this work, we characterized transcriptional dysregulation associated with REST in two different HD

Targeted deletion of the zebrafish obscurin A RhoGEF domain affects heart, skeletal muscle and brain development.

Obscurin is a giant structural and signaling protein that participates in the assembly and structural integrity of striated myofibrils. Previous work has examined the physical interactions between obscurin and other cytoskeletal elements but its in vivo role in cell signaling, including the functions of its RhoGTPase Exchange Factor (RhoGEF) domain have not been characterized. In this study, morpholino antisense oligonucleotides were used to create an in-frame deletion of the active site of the obscurin A RhoGEF domain in order to examine its functions in zebrafish development. Cardiac myocyte

Akt pathway mediates a cGMP-dependent survival role of nitric oxide in cerebellar granule neurones.

Apoptotic death results from disrupting the balance between anti-apoptotic and pro-apoptotic cellular signals. The inter- and intracellular messenger nitric oxide is known to mediate either death or survival of neurones. In the present work, cerebellar granule cells were used as a model to assess the survival role of nitric oxide and to find novel signal transduction pathways related to this role. It is reported that sustained inhibition of nitric oxide production induces apoptosis in differentiated cerebellar granule neurones and that compounds that slowly release nitric oxide significantly r

Sugar on the Coat: Site-Specific O-GlcNAc and Myoferlin Adapt COPII for Collagen Export.

Recent work by Hirata and colleagues identifies site-specific O-GlcNAc modification of the cargo-selecting COPII subunit Sec24D as a dynamic regulatory switch that promotes collagen export from the endoplasmic reticulum (ER), providing new insight into how COPII accommodates cargoes far larger than canonical transport vesicles. Two glycosites in the poorly characterized N-terminal intrinsically disordered region (IDR) of Sec24D, T9 and S13, display strikingly different dynamics: S13 is constitutively glycosylated, whereas T9 is rapidly modified following a synchronized burst of collagen transp

Cardiac Efficiency as a Potential Marker of Firefighter Expertise: A Pilot Study of Heart Rate Variability Monitoring in Realistic Search-and-Rescue Training.

Expertise in high-stakes occupations is typically evaluated through performance outcomes, potentially neglecting physiological efficiency-a dimension critical for career longevity in physically demanding work. In this pilot study, we investigated whether expert firefighters achieve comparable task performance with lower cardiovascular activation and faster autonomic recovery than novices during realistic search-and-rescue operations. Twenty-two male firefighters completed navigation tasks in a high-fidelity training facility under operational conditions (complete darkness, full protective equi

H19 enhances pancreatic cancer proliferation and invasion by reducing miR-29c-5p's inhibitory effects on ATF2/ECM1.

Pancreatic ductal adenocarcinoma is a lethal malignancy with few therapeutic options. Identifying novel molecular targets is essential for treating this illness. This work suggests that the long non-coding RNA H19 exhibits a carcinogenic role in pancreatic ductal adenocarcinoma (PDAC). Analysis of TCGA data and clinical samples revealed a significant elevation of H19 in PDAC tissues. This elevation was associated with a poor outcome and a heightened likelihood of metastatic spread. In this study, H19 was demonstrated to enhance the proliferation, migration, and invasion of PDAC cells via the c

Kisspeptin-54 confers renal protection in diabetic nephropathy by ameliorating endothelial permeability through ZEB1 inhibition.

BACKGROUND: Diabetic nephropathy (DN) is a leading cause of end-stage renal disease (ESRD). A central pathological feature is the breakdown of the glomerular endothelial barrier. Although the Kisspeptin-54/GPR54 pathway is known to regulate vascular function, its specific role and mechanism in DN-related renal endothelial damage are unclear. METHODS: This study used db/db mice (type 2 DN model) and human renal glomerular endothelial cells (HrGECs) exposed to high glucose (HG). We measured pathway components, assessed barrier function via albumin excretion and electrical resistance, and invest

Parkinson's disease-associated PINK1 loss disrupts ensheathing glia and causes dopaminergic neuron synapse loss.

Parkinson's disease (PD) is commonly associated with the loss of dopaminergic neurons in the substantia nigra, but many other cell types are affected even before neuron loss occurs. Recent studies have linked oligodendrocytes to early stages of PD, though their precise role is still unclear. PINK1 is mutated in familial PD, and through unbiased single-cell sequencing of the entire brain of Drosophila Pink1 models, we observed significant gene deregulation in ensheathing glia (EG), cells that share functional similarities with oligodendrocytes. We found that the loss of Pink1 leads to abnormali

Adhiron binders as molecular tools for inhibiting activation dependent, IL-7 mediated long-term survival of CD4+ T-cells.

Tools that enable functional inhibition of the long-term effects of immunological signalling cascades remain challenging to develop, in part because biological outcomes often arise long after the initiating receptor-ligand interaction and lack accessible experimental readouts. Interleukin-7 (IL-7) cytokine signalling through the IL-7 receptor (IL-7R) exemplifies this problem, as IL-7-mediated survival signals in activated CD4+ T cells emerge over extended timescales, yet play a critical role in human disease. Here, we describe the development of engineered Adhiron binders as molecular tools to

TREM2 Impedes Recovery After Spinal Cord Injury by Regulating Microglial Lysosomal Membrane Permeabilization-Mediated Autophagy.

Microglia, considered as the main immune responder, play an important role in regulating neuroinflammation in central nervous system (CNS) disorders. Our previous work found that TREM2 is highly expressed in microglia and is related to their functional state. However, the specific role of TREM2 in spinal cord injury has not yet been explored. To further investigate the potential mechanism of TREM2, we performed single-cell sequencing on wild-type (Wt) and Trem2-/- mice before and after spinal cord injury. Compared to Wt mice, the lysosome, autophagy and membrane-related pathways are more stron

Prothioconazole induced stereoselective developmental toxicity and liver injury in zebrafish embryos via ferroptosis.

The widespread use of prothioconazole (PTCZ), a globally applied triazole fungicide, raises concerns regarding ecological risks from environmental residues and highlights the critical gap in pesticide safety assessment concerning enantiomeric differences in toxicity. This study investigated the stereoselective toxicity and molecular mechanisms of PTCZ enantiomers in an aquatic model using a zebrafish embryo exposure system. The toxic effects were systematically analyzed through multidimensional endpoint assessments, which examined developmental malformations, liver histopathology, lipid metabo
