Meclizine is neuroprotective in models of Huntington's disease.

Defects in cellular energy metabolism represent an early feature in a variety of human neurodegenerative diseases. Recent studies have shown that targeting energy metabolism can protect against neuronal cell death in such diseases. Here, we show that meclizine, a clinically used drug that we have recently shown to silence oxidative metabolism, suppresses apoptotic cell death in a murine cellular model of polyglutamine (polyQ) toxicity. We further show that this protective effect extends to neuronal dystrophy and cell death in Caenorhabditis elegans and Drosophila melanogaster models of polyQ t

Outcomes of Pediatric Patients with Excessive Postural Tachycardia After Attending an Intensive Interdisciplinary Pain Program: A Pilot Study.

BACKGROUND/OBJECTIVES: Adolescents with autonomic disorders who attend Intensive Interdisciplinary Pain Treatment (IIPT) programs report improvements in functioning. However, it is unclear whether they experience corresponding improvements in physiological measures. As such, the aim of this pilot study was to examine changes in physiological measures in youth attending an IIPT program who demonstrated excessive postural tachycardia on an active stand test. The secondary goal was to examine associations between physiological measurements and self-reported measures of chronic orthostatic intoler

The structural analogue of apelin-12 prevents energy disorders in the heart in experimental type 1 diabetes mellitus.

Type 1 diabetes mellitus (T1DM) is the most severe form of diabetes, which is characterized by absolute insulin deficiency induced by the destruction of pancreatic beta cells. The aim of this study was to evaluate the effect of a structural analogue of apelin-12 ((NαMe)Arg-Pro-Arg-Leu-Ser-His-Lys-Gly-Pro-Nle-Pro-Phe-OH, metilin) on hyperglycemia, mitochondrial (MCh) respiration in permeabilized cardiac left ventricular (LV) fibers, the myocardial energy state, and cardiomyocyte membranes damage in a model of streptozotocin (STZ) diabetes in rats. Metilin was prepared by solid-phase synthesis u

Identifying the key role of mitochondrial respiration and lipid metabolism in regulating axillary osmidrosis through proteomics analysis.

Axillary osmidrosis (AO) affects a large number of young people in Asia, resulting from a combination of body and bacterial metabolism. This study aimed to explore the pathogenesis of AO through proteomics. Apocrine gland tissues from 3 mild and 3 severe AO patients were analyzed using 4D label-free proteomics, followed by bioinformatics analysis. The RNA and protein levels of the predicted key regulators were further validated by qPCR and immunohistochemistry in additional AO tissues. A total of 5066 proteins were identified, of which 323 were significantly upregulated and 412 were downregula

Feasibility of cellular bioenergetics as a biomarker in porphyria patients.

Porphyria is a group of metabolic disorders due to altered enzyme activities within the heme biosynthetic pathway. It is a systemic disease with multiple potential contributions to mitochondrial dysfunction and oxidative stress. Recently, it has become possible to measure mitochondrial function from cells isolated from peripheral blood (cellular bioenergetics) using the XF96 analyzer (Seahorse Bioscience). Mitochondrial respiration in these cells is measured with the addition of activators and inhibitors of respiration. The output is measured as the O2 consumption rate (OCR) at basal condition

Induced schizophrenic like breathing pattern leads to impaired cardiorespiratory coupling in healthy subjects.

Schizophrenia is referred to as one of the most severe mental disorders in the world, and patients with this condition are associated with high cardiac mortality rates. However, the reasons for this high mortality rates are still under debate. One major contributing factor seems to be that a dysfunction of the autonomic nervous system (ANS) is evident in schizophrenia. Thereby, recent investigations focused on the analyses of respiration and cardiorespiratory coupling (CRC) in these patients. The objective of this study was to characterize the causal strength and direction of CRC applying the n

[Definitions and clinical classifications of sleep disorders].

Sleep disorders are defined as disorders which symptoms or pathophysiology are related with sleep regardless of comorbid physical and/or mental disorders. Sleep disorders are classified into 6 major categories: sleep related breathing disorders which exhibit abnormal breathing during sleep, sleep related movement disorders which show involuntary movements and/or abnormal sensations during sleep and/or nighttime, hypersomnia of central origin not due to other sleep disorders, circadian rhythm sleep disorders due to desynchronization between sleep-wake pattern and required social schedule, paras

Cabergoline counteracts adipose and skeletal muscle lipid accumulation: A novel therapeutic approach to obesity?

OBJECTIVE: Obesity represents a global health problem with large costs for public health. Cabergoline (CAB), a potent dopamine agonist with a high affinity for type 2 dopamine receptor (D2R), usually used in clinical practice for hyperprolactinemia management, has been reported to significantly decrease metabolic syndrome prevalence and visceral adiposity associated with improved insulin sensitivity in prolactinoma patients. Nevertheless, scant data are reported about CAB effects on obesity condition. METHODS: The current preclinical study investigated the in vivo chronic effects of CAB on ob

Obesity-induced arterial redox imbalance involving mitochondrial NOX4, endothelial dysfunction, and ER stress underlie kidney injury compensated by enhanced mitochondrial bioenergetics.

Mitochondrial reactive oxygen species (mtROS) are key pathogenic factors in the microvascular complications of metabolic disorders including nephropathy. However, the effects of obesity on kidney vascular mitochondria and endothelial function remain unclear. We assessed here the specific impact of obesity on endothelial function, mtROS-derived oxidative stress and mitochondrial bioenergetics of kidney preglomerular arteries in rat models of high fat diet (HFD)-induced obesity and endoplasmic reticulum (ER) stress. Arterial function was assessed in microvascular myographs, mitoSOX and Amplex Re

Measurement of respiratory-swallowing coordination using an oronasal facemask in healthy individuals.

Respiratory-swallowing coordination (RSC) is well established as an essential airway-protective mechanism. Previous studies have used nasal airflow and/or kinematic rib cage and abdominal measures to assess respiration surrounding swallowing, meaning that the direct influence of oral respiration on RSC remains unknown. This study used a partitioned oronasal facemask to compare respiratory phase patterns measured using isolated nasal airflow with those measured using combined oronasal airflow during non-ingestive and ingestive swallowing tasks. Twenty-four healthy individuals with no respirator

Gαq modulates the energy metabolism of osteoclasts.

INTRODUCTION: The bacterial protein toxin Pasteurella multocida toxin (PMT) mediates RANKL-independent osteoclast differentiation. Although these osteoclasts are smaller, their resorptive activity is high which helps in efficient destruction of nasal turbinate bones of pigs. METHODS: The proteome of bone marrow-derived macrophages differentiated into osteoclasts with either RANKL or PMT was analysed. The results were verified by characterizing the metabolic activity using Seahorse analysis, a protein translation assay, immunoblots, real-time PCR as well as flow cytometry-based monitoring of m

Arctigenin, a natural compound, activates AMP-activated protein kinase via inhibition of mitochondria complex I and ameliorates metabolic disorders in ob/ob mice.

AIMS/HYPOTHESIS: Arctigenin is a natural compound that had never been previously demonstrated to have a glucose-lowering effect. Here it was found to activate AMP-activated protein kinase (AMPK), and the mechanism by which this occurred, as well as the effects on glucose and lipid metabolism were investigated. METHODS: 2-Deoxyglucose uptake and AMPK phosphorylation were examined in L6 myotubes and isolated skeletal muscle. Gluconeogenesis and lipid synthesis were evaluated in rat primary hepatocytes. The acute and chronic effects of arctigenin on metabolic abnormalities were observed in C57BL

Heartbeat-evoked potentials reveal interoceptive dysfunction in clinical disorders: Experimental frameworks and promising applications.

Interoception refers to the ability to perceive and integrate physiological signals originating from within the body, such as heartbeat and respiration. This process involves both bottom-up and top-down. As a key neurophysiological marker of interoception, the heartbeat-evoked potential (HEP) reflects the cortical processing of cardiac signals in the brain. In this review, we first outline the neural mechanisms underlying interoception and HEP, followed by a comprehensive overview of the methodologies commonly employed in HEP research. Based on the directionality of interoceptive information f

Recent advances in high relative humidity strategy for preservation of postharvest fruits and vegetables: A comprehensive review.

Postharvest water loss is a major factor resulting in quality deterioration and physiology disorders of fruits and vegetables, which is effectively inhibited by high relative humidity (RH) storage. High RH storage could retain quality attributes and stress tolerance, also influence physiological metabolisms and cellular level responses of postharvest horticultural products, involving cell ultrastructure integrity, degradation of bioactive compounds, antioxidant response, respiration processes, ripening and senescence. In addition, challenges for the implementation of high RH strategies in the

Thoracic Wall Reconstruction with Dorsal Diaphragmatic Traction and Preservation of Diaphragmatic Attachments in a Dog with Resection of the 11-13th Ribs.

A 9-year-old, 4.7 kg, spayed female Chihuahua presented with a 3.5 cm soft tissue sarcoma on the dorsal right thoracic wall. The tumor was resected, including the 11−13th ribs, resulting in a caudal dorsal thoracic wall defect. The defect was reconstructed with direct traction of part of the diaphragm dorsally, preserving the diaphragmatic attachments to the body wall, and the diaphragm was sutured to the surrounding ribs and muscles. Possible respiratory complications, including paradoxical respiration and exercise intolerance, were not observed during the perioperative or postoperative obser

A laser-engraved wearable sensor for sensitive detection of uric acid and tyrosine in sweat.

Wearable sweat sensors have the potential to provide continuous measurements of useful biomarkers. However, current sensors cannot accurately detect low analyte concentrations, lack multimodal sensing or are difficult to fabricate at large scale. We report an entirely laser-engraved sensor for simultaneous sweat sampling, chemical sensing and vital-sign monitoring. We demonstrate continuous detection of temperature, respiration rate and low concentrations of uric acid and tyrosine, analytes associated with diseases such as gout and metabolic disorders. We test the performance of the device in
