Rationale for an experimental treatment of retinitis pigmentosa: 140-month test of hypothesis with one patient.

BACKGROUND: Numerous mutations in over 100 rod genes are the well-established cause of apoptotic death of these cells and development of night blindness in retinitis pigmentosa (RP). Cone death is either concomitant or follows rod death with resultant loss of critical peripheral and central day vision. As cones are generally not encumbered by genetic mutations, the causes of their death and its prevention are the central problems of RP research. Currently no FDA-approved medications are available for retarding RP progression. HYPOTHESIS: It is proposed that cones, which are outnumbered 20:1 b

Progressive cone dystrophy with deutan genotype and phenotype.

PURPOSE: To study the electroretinographic signals originating in the long-wavelength-sensitive (L) and middle-wavelength-sensitive (M) cone pathways by means of large-field and multifocal cone type-specific electroretinograms (ERGs) in a patient with progressive cone dystrophy. METHODS: A 65-year-old male patient with colour vision disturbances (age at onset 10 years), loss of visual acuity (14 years), and central visual field defects (40 years) was investigated. Large-field flicker-ERG responses to stimuli that exclusively modulated the L-cones or the M-cones, or the two simultaneously (bot

Putative Photosensitivity-Associated Sexual Dimorphism in Compound Eye Structure of Lymantria xylina (Lepidoptera: Erebidae).

Lymantria xylina is a major pest in coastal casuarina shelterbelts and a species subject to quarantine regulations by countries to which it is non-native. Phototaxis is fundamental to the insect's surveillance and risk assessment analysis, and it exhibits pronounced sexual dimorphism in compound eye structure. This dimorphism was investigated using scanning and transmission electron microscopy. Males displayed significantly larger compound eyes, characterized by greater ommatidial areas and a higher total number of facets per eye compared to females. From the distal to proximal end, the ommati

Electrophysiology in the investigation of acquired retinal disorders.

Electrophysiological research on acquired retinal disorders, both common and rare, is reviewed. Age is a major factor influencing electroretinogram (ERG) and electro-oculogram (EOG) findings. Bipolar or Müller cell death in the aging retina could account for much of the amplitude decline that is observed with age. In diabetic retinopathy, the oscillatory potentials can monitor the progression of the disease and indicate neuronal alterations rather than diabetic angiopathy of the retina. Human ERG studies on glaucoma concentrated on ERG measures that are dominated by inner retinal contributions

The co-expression of the depolarizing and hyperpolarizing mechanosensitive ion channels in mammalian retinal neurons.

INTRODUCTION: The elevation of the intraocular and extraocular pressures is associated with various visual conditions, including glaucoma and traumatic retinal injury. The retina expresses mechanosensitive channels (MSCs), but the role of MSCs in retinal physiology and pathologies has been unclear. METHODS: Using immunocytochemistry, confocal microscopy, and patch-clamp recording techniques, we studied the co-expression of K+-permeable (K-MSCs) TRAAK and big potassium channel BK with the epithelial sodium channel ENaC and transient receptor potential channel vanilloid TPRV4 and TRPV2 favorabl

A teenager with nightblindness and cystic maculopathy: enhanced S cone syndrome (Goldmann-Favre syndrome).

Poor night vision and pigmentary retinopathy are hallmarks of retinitis pigmentosa, but variations in the type of pigmentation and in the characteristics of the electroretinogram (ERG) can point to other diagnoses. The case below (enhanced S cone syndrome) is a good example of this, and demonstrates the diagnostic importance of clinical electrophysiology.

Physiological Postnatal Neurogenesis, Development, and Maturation in the Regenerable Retina.

The retina enlarges postnatally by ∼440% in axolotls and ∼150% in humans, predicting a reduction in neuron densities by 81.5% and 60%, respectively, if neurogenesis is absent. The concurrent retinal remodeling is poorly characterized. We studied retinas of 2-15 cm axolotls. Results showed that hatchling (∼2 cm) retinas, particularly the peripheral (Peri) had 1-2 more soma rows in the ganglion cell (GCL) layer and outer (ONL) and inner nuclear (INL) layers. Cell density reduced in GCL (GCs, -49%) in juveniles (4-12 cm) but increased in INL (74%) and ONL (55.5%) in young juveniles (4-6 cm), demo

Ciliary neurotrophic factor (CNTF) protects retinal cone and rod photoreceptors by suppressing excessive formation of the visual pigments.

The retinal pigment epithelium (RPE)-dependent visual cycle provides 11-cis-retinal to opsins in the photoreceptor outer segments to generate functional visual pigments that initiate phototransduction in response to light stimuli. Both RPE65 isomerase of the visual cycle and the rhodopsin visual pigment have recently been identified as critical players in mediating light-induced retinal degeneration. These findings suggest that the expression and function of RPE65 and rhodopsin need to be coordinately controlled to sustain normal vision and to protect the retina from photodamage. However, the

Interactions of cone abundancies, opsin expression, and environmental lighting with emmetropization in chickens.

We had previously found that M to L cone abundancy ratios in the chicken retina are correlated with vitreous chamber depth and refractive state in chickens eyes, when they have normal visual exposure but not when they develop deprivation myopia. The finding suggests an interaction between cone abundancies and emmetropization. In the current study, we analyzed how stable this correlation was against changes in environmental variables and strain differences. We found that the correlation was preserved in two chicken strains, as long as they were raised in the laboratory facilities and not in the

[Macular Telangiectasia Type 2 (MacTel) - Therapeutic Options and New Developments].

ABSTRACT: Macular telangiectasia type 2 (MacTel) is a rare, bilateral neurodegenerative disease of the macula associated with secondary vascular alterations. Pathophysiologically, Müller cell dysfunction plays a central role. Their loss leads to progressive degeneration of central cone photoreceptors and may secondarily contribute to the development of macular neovascularization (MNV), which affects approximately 10% of patients. ABSTRACT: A major pathophysiologic mechanism is a disturbance in serine and sphingolipid metabolism. Patients with MacTel have significantly lower blood serine level

Gene editing prospects for treating inherited retinal diseases.

Retinal diseases (RD) include inherited retinal dystrophy (IRD), for example, retinitis pigmentosa and Leber's congenital amaurosis, or multifactorial forms, for example, age-related macular degeneration (AMD). IRDs are clinically and genetically heterogeneous in nature. To date, more than 200 genes are known to cause IRDs, which perturb the development, function and survival of rod and cone photoreceptors or retinal pigment epithelial cells. Conversely, AMD, the most common cause of blindness in the developed world, is an acquired disease of the macula characterised by progressive visual impa

Neonatal and Juvenile Ocular Development in Göttingen Minipigs and Domestic Pigs: A Histomorphological and Immunohistochemical Study.

Pigs are considered one of the relevant animal models for ocular research as they share several histological and anatomical similarities with the human eye. With the increasing interest in juvenile animal models, this study aimed to describe the postnatal development of ocular structures in 16 Göttingen minipigs and 25 F2 domestic pigs, between birth and 6 months of age, using histopathology and immunohistochemistry against Ki-67, caspase-3, calbindin, glial fibrillary acidic protein, rhodopsin, and synaptophysin. All ocular structures in both pig breeds were incompletely developed at birth an
