Scorpionism at the human-environment interface: an eco-epidemiological synthesis and conceptual framework.

INTRODUCTION: Scorpion envenomation is a major but persistently under-recognized public health problem in tropical and subtropical regions. Its occurrence arises from complex interactions among climatic, ecological, socioeconomic, and health-system drivers. Despite sustained high incidence and expanding risk associated with climate and land-use change, these relationships remain insufficiently synthesized. Addressing this gap requires an integrative framework capable of capturing the multi-level determinants shaping scorpionism risk and outcomes. METHODS: Drawing on Human Ecology Theory and s

Stinging Salvation: Harnessing Scorpion Venom Peptides for Revolutionary Pain Relief.

Peptides from scorpion venom, mainly in species such as Olivierus martensii (formerly Olivierus martensii Karsch, often designated BMK) (BmK) and Tityus serrulatus from the Buthidae family, show real promise as painkillers that skip opioids altogether. They work by hitting specific ion channels and dialing down inflammation. This review gathers information on their molecular setups: disulfide-bridged types and those without, weighing in at 3 to 10 kilodaltons (kDa). Structural features include motifs stabilized by cysteines. In pain signaling, they block voltage-gated sodium channels (NaV) suc

Broad-Spectrum Antiviral and Antibacterial Activity of the Scorpion Venom Peptide HP1090.

HP1090 is a short, cationic, amphipathic peptide derived from scorpion venom and previously described as a membrane-active antiviral compound. Here, we primarily characterize the antiviral activity of HP1090 and assess whether additional antibacterial effects are consistent with membrane-disruptive properties. Chemically synthesized HP1090 exhibited dose-dependent virucidal activity against multiple enveloped viruses, including herpes simplex virus type 1 and 2 (HSV-1, HSV-2), human immunodeficiency virus type 1 (HIV-1), and Zika virus (ZIKV), with IC50 values ranging from 14.7 to 56.1 µg/mL.

Clinical and hematological predictors of severity in pediatric patients with Centruroides scorpion stings.

Introduction. In Mexico, scorpion stings are the leading cause of death from contact with venomous animals among children under 5 years of a … ogistic regression, with a significance level of p <0.05. Results. A total of 349 patients stung by scorpions of the genus Centruroides were analyzed over 10 years. A negative correlation was observed between age, weight, and height and the severity of poisoning (p = 0.001); that is, the younger the patient and the lower their weight and height, the greater …

Scorpion stings and broken hearts: an underdiagnosed etiology of inverted Takotsubo cardiomyopathy.

INTRODUCTION: Severe cases of scorpion envenomation are commonly characterized by pulmonary oedema and heart failure. One mechanism of this acute heart failure is Takotsubo cardiomyopathy. The inverted type is reported less, and its management remains controversial. CASE REPORTS: We present six patients diagnosed with inverted Takotsubo cardiomyopathy following scorpion envenomation. They were aged 5-55 years and presented with clinical signs of acute heart failure. Echocardiography revealed apical hyperkinesia and basal hypokinesis, consistent with inverted Takotsubo cardiomyopathy. Manageme

Scorpion venom peptides: Novel therapeutic approaches for inflammatory and hepatic disorders.

Chronic hepatic disorders, such as metabolic dysfunction associated steatohepatitis (MASH), alcohol associated liver disease (ALD), and viral hepatitis (Hepatitis B virus [HBV]/Hepatitis C virus [HCV]), are primarily driven by persistent immune-mediated inflammation and hepatic stellate cell activation leading to fibrosis, yet conventional therapies lack tissue and molecular specificity. Scorpion venom peptides, refined through evolutionary selection, provide highly potent, target specific scaffolds capable of modulating intrahepatic inflammatory networks. Recent in vivo preclinical studies in

Targeted Metabolomics of Tityus Scorpion Venoms: Unveiling Small-Molecule Components.

Scorpion venoms consist of proteins, peptides, and various low molecular weight (LMW) organic compounds, which act as toxins. Despite their potential significance, these compounds in scorpion venoms have been little investigated and their full range has not been well characterized. In this work, a targeted metabolomic approach was used in combination with an HPLC-QTOF-MS methodology to create a library of 55 LMW standard compounds, for the analysis of venoms from three Tityus species scorpions. This strategy enabled reliable identification of 45 compounds, including 20 amino acids, 4 organic a…

Design of antinociceptive peptide by grafting domains between scorpion β-neurotoxins.

Neurotoxic peptides from venomous animals have been pointed out as antinociceptive therapeutic leads. Venom peptides have modulation properties on isoforms of voltage-gated sodium channels (VGSC), associated with pathologies such as neuropathic or inflammatory pain. The β-neurotoxins obtained from scorpion venoms are peptides that can alter the kinetics of VGSC by binding to its receptor site 4. CeII8 is a non-lethal scorpion β-neurotoxin that has been reported to interact with hNaV1.7, a VGSC isoform related to the codification and processing of painful stimulus (nociception), and it is invol

Beyond the venom: Exploring the antimicrobial peptides from Androctonus species of scorpion.

Prevalent worldwide, the Androctonus scorpion genus contributes a vital role in scorpion envenoming. While diverse scorpionisms are observed because of several different species, their secretions to protect themselves have been identified as a potent source of antimicrobial peptide (AMP)-like compounds. Distinctly, the venom of these species contains around 24 different AMPs, with definite molecules studied for their therapeutic potential as antimicrobial, antifungal, antiproliferative and antiangiogenic agents. Our review focuses on the therapeutic potential of native and synthetic AMPs ident

Harnessing the potency of scorpion venom-derived proteins: applications in cancer therapy.

Despite breakthroughs in the development of cancer diagnosis and therapy, most current therapeutic approaches lack precise specificity and sensitivity, resulting in damage to healthy cells. Selective delivery of anti-cancer agents is thus an important goal of cancer therapy. Scorpion venom (SV) and/or body parts have been used since early civilizations for medicinal purposes, and in cultures, SV is still applied to the treatment of several diseases including cancer. SV contains numerous active micro and macromolecules with diverse pharmacological effects. These include potent anti-microbial, a

Acetylcholinesterase inhibitory potential of scorpion venom in Aedes aegypti (Diptera: Culicidae).

Scorpion venom contains a variety of neurotoxins which interact with ion channels and affect their activities. The present study was designed to evaluate the potential of scorpion venom as acetylcholinesterase (AChE) inhibitor by using Aedes aegypti as model organism. Venoms of two species, Hottentota tamulus (Fabricus, 1798) and Androctonus finitimus (Pocock, 1897) were selected for this study. Two peptides (36 kDa from H. tamulus and 54 kDa from A. finitimus) were separated from scorpion venom by using HPLC. Selected peptides caused significantly higher mortality in larvae and adults of Aede
