BACKGROUND: T cells are closely linked to the clinical manifestations of subjects with Mycobacterium tuberculosis (MTB) infection. T cell receptor beta variable (TCRBV) is a signal and indicative molecule on the membrane of T lymphocytes, reflecting the composition and specificity of T cells. The molecular profiles of TCRBV in peripheral blood mononuclear cells (PBMCs) and their subpopulations (CD4+ and CD8+ T cells) from subjects with active tuberculosis (TB) or latent TB infection (LTBI) have not been well described.
METHODS: In 42 subjects with active TB or LTBI, PMBCs and their subsets we

Atherosclerosis is a chronic immune-inflammatory disease of the arterial wall in which antigen-presenting cells (dendritic cells, macrophages and B cells) present locally generated antigens, such as oxidized low-density lipoprotein, to plaque-infiltrating T cells. Full T-cell activation requires costimulation through the B7 family ligands CD80 (B7-1) and CD86 (B7-2), which engage CD28 to activate T cells and cytotoxic T-lymphocyte-associated protein 4 (CTLA-4) to restrain them. Although long considered functionally redundant, CD80 and CD86 are now known to be structurally and mechanistically d

Wound healing requires coordinated interactions between macrophages and fibroblasts, yet how contact-dependent signaling integrates with paracrine pathways to regulate their reciprocal behavior is not well defined. Here, we investigated macrophage-fibroblast communication using complementary 2D and 3D in vitro wound healing models combined with live-cell calcium imaging and single-cell RNA sequencing (scRNA-seq). We show that bone marrow-derived macrophages (BMDMs) promote fibroblast scratch closure in a contact-dependent manner independent of connexins, whereas fibroblasts reciprocally regula

Radiolabeled nanocarriers that can simultaneously deliver therapeutic and diagnostic agents are promising for targeted cancer therapy. In this study, we employed molecular dynamics and metadynamics simulations to investigate how the chemical identity of drug molecules influences the behavior of technetium-99 m (⁹⁹ᵐTc)-loaded carbon nanotube (CNT) nanocarriers at the atomic scale. Four clinically relevant drugs-methotrexate, diclofenac, ketotifen, and piroxicam-were conjugated to a single-walled CNT (SWCNT) via a bifunctional chelating group. Our results revealed that drug structure significant

BACKGROUND: Cancer-associated fibroblasts have become a new target for therapy. Fibroblasts present within malignancies express the fibroblast activation protein (FAP). Inhibitors to FAP (FAPI) are small molecules recently developed as a theranostic agents for imaging and radiotherapy. All currently used FAPI rely on a linker-chelator complex attached to the 'inhibitor'. We describe a new automated method of the direct attachment of the radioisotope to the inhibitor, resulting in a >50% MW reduction with the hope of an improved tumor-to-background ratio and tumor uptake.

In this Perspective, we review our recent work on rotationally inelastic collisions of highly vibrationally excited NO molecules prepared in single rotational and parity levels at v = 10 using stimulated emission pumping (SEP). This state preparation is employed in a recently developed crossed molecular beam apparatus where two nearly copropagating molecular beams achieve an intersection angle of 4° at the interaction region. This near-copropagating beam geometry of the molecular beams permits very wide tuning of the collision energy, from far above room temperature down to 2 K where we test t

This study presents an alternative approach to directly synthesizing magnetite nanoparticles (MNPs) in the presence of Vitis vinifera, Vaccinium corymbosum, and Punica granatum derived from natural sources (grapes, blueberries, and pomegranates, respectively). A modified co-precipitation method that combines phytochemical techniques was developed to produce semispherical MNPs that range in size from 7.7 to 8.8 nm and are coated with a ~1.5 nm thick layer of polyphenols. The observed structure, composition, and surface properties of the MNPs@polyphenols demonstrated the dual functionality of th

Bacterial infection seriously restricts the wound healing process due to severe inflammation and delayed wound healing. Unfortunately, the overuse or improper use of antibiotics leads to the advent of multidrug-resistant strains and intractable biofilms, severely affecting the therapeutic effect. Therefore, there is an urgent need to develop antibiotic-free strategies to accelerate the healing process of wounds with bacterial infection. Considering that single photothermal therapy (PTT) or photodynamic therapy (PDT) cannot fully meet the requirements of clinical sterilization and accelerating

Patterning chemical reactivity with a high spatiotemporal resolution and chemical versatility is critically important for advancing revolutionary emergent technologies, including nanorobotics, bioprinting, and photopharmacology. Current methods are complex and costly, necessitating novel techniques that are easy to use and compatible with a wide range of chemical functionalities. This study reports the development of a digital light processing (DLP) fluorescence microscope that enables the structuring of visible light (465-625 nm) for high-resolution photochemical patterning and simultaneous f

H2S and H2O2 participate in various biochemical reactions as active signaling molecules and maintain cellular homeostasis during oxidative stress. Current methods for detecting H2S and H2O2 mostly rely on their respective single-detection systems, while the research on dual-detection systems for both substances remains less explored. Herein, we rationally constructed a new naphthalimide-based fluorescence probe P1 for the simultaneous detection of H2S and H2O2 using the dual-lock strategy. P1 consists of naphthalimide fluorephore, the disulfide bond-containing ferrocene (first lock) and the bo

Focused near-infrared femtosecond laser irradiation induces efficient photochemical breakdown of intracellular molecules, resulting in the formation of gas microbubbles that ultimately lead to cell death and removal. This study characterizes these microbubbles using high-speed imaging. Microbubbles generated within cells by single-pulse irradiation are consistently larger than those formed in the surrounding culture medium across a broad pulse energy range. Notably, the energy threshold for intracellular microbubble formation is several tens of nanojoules lower than in the culture medium, enab

Hydroxyurea (HU; hydroxycarbamide) is a chemotherapy medication used to treat various types of cancer and other diseases such as sickle cell anemia. HU inhibits DNA synthesis by targeting ribonucleotide reductase (RNR). Recent studies have suggested that HU also causes oxidative stress in living systems. In the present study, we investigated if HU could directly affect the activity and/or conformation of DNA. We measured in vitro gene expression in the presence of HU by adapting a cell-free luciferase assay. HU exhibited a bimodal effect on gene expression, where promotion or inhibition were o

53BP1 protein has been isolated in-vitro as a putative p53 interactor. From the discovery of its engagement in the DNA-Damage Response (DDR), its role in sustaining the activity of the p53-regulated transcriptional program has been frequently under-evaluated, even in the case of a specific response to numerous DNA Double-Strand Breaks (DSBs), i.e., exposure to ionizing radiation. The localization of 53BP1 protein constitutes a key to decipher the network of activities exerted in response to stress. We present here an automated-microscopy for image cytometry protocol to analyze the evolution of

As vital important bioactive species, human serum albumin (HSA) and sulfur dioxide (SO2) are essential molecules in the organisms and act a pivotal part in many biological events. Although studies have shown that SO2-induced HSA radicals can cause oxidative damage, the underlying mechanism of the synergistic effect of HSA and SO2 in various diseases is obscure, mainly because of the lack of powerful tools that can simultaneously detect HSA and SO2 in living systems. In this work, we report a novel single-site, double-sensing fluorescent probe 1 for the simultaneous detection of HSA and SO2. Th

In the adrenal gland, neuroendocrine cells that synthesize catecholamines and epithelial cells that produce steroid hormones are united beneath a common organ capsule to function as a single stress-responsive organ. The functional anatomy of the steroid hormone-producing adrenal cortex and the catecholamine-producing medulla is ill defined at the level of small molecules. Here, we report a comprehensive high-resolution mass spectrometry imaging (MSI) map of the normal human adrenal gland. A large variety of biomolecules was accessible by matrix-assisted laser desorption/ionization-Fourier tran

How appetite is modulated by physiological, contextual, or pharmacological influence is still unclear. Specifically, the discovery of appetite modulators is compromised by the abundance of side effects that usually limit in vivo drug action. We set out to identify neuroactive drugs that trigger only their intended single behavioral change, which would provide great therapeutic advantages. To identify these ideal bioactive small molecules, we quantified the impact of more than 10,000 compounds on an extended series of different larval zebrafish behaviors using an in vivo imaging strategy. Targeting both tumor cells and microenvironment is a promising anticancer strategy. FGFR1 and VEGFR2 represent important drivers of tumor progression and angiogenesis. We engineered multitarget proteins by fusing DR5‑specific TRAIL DR5-B with SRH and P9 peptides antagonizing VEGFR2 and FGFR1, respectively. SRH-DR5-B, DR5-B-P9, and SRH-DR5-B-P9 fusions demonstrated nanomolar affinity for receptors DR5, VEGFR2, and FGFR1 and reduced viability of FGFR1-positive gastric (MKN-45) and colon (HCT116, COLO 205) adenocarcinoma cells. DR5‑B-P9 and SRH‑DR5‑B‑P9 decreased phospho‑FGFR1 levels in FGF2‑stim

Single-molecule magnets (SMMs) and low-dimensional van der Waals (vdW) magnets are highly promising for quantum technologies, spintronics, and data storage, yet their integration into practical devices is frequently impeded by chemical reactivity, aggregation, or loss of magnetic function during processing. Encapsulation within carbon nanotubes (CNTs) and boron nitride nanotubes (BNNTs) has emerged as a versatile strategy to protect, isolate, align, and electronically address these fragile magnetic species at the atomic scale. This review provides a comprehensive overview of encapsulated magne

BACKGROUND: Prediction of stroke outcomes is challenging due to limited predictive properties of clinical and radiological variables. We aimed to investigate whether local cerebral neurofilament light chain (NfL) could provide additional prognostic value. METHODS: We prospectively enrolled 110 consecutive patients with large vessel occlusion of the anterior circulation who underwent endovascular thrombectomy (EVT). We collected two different arterial blood samples during EVT: (1) Local: the region of the occluded vascular compartment; (2) Systemic: femoral artery. We used ultrasensitive single

BACKGROUND: Sjögren's disease (SjD) is an autoimmune disease marked by lymphocytic infiltration of salivary and lacrimal glands, leading to glandular dysfunction, where CD4-positive helper T (Th) cells and their cytokines are crucial in the pathogenesis. Recent studies have demonstrated that Toll-like receptors (TLRs), particularly those recognizing immune complexes containing DNA and RNA, contribute to Th cell activation in various autoimmune diseases. This study explores the expression and function of these TLRs in SjD.
METHODS: DNA microarray analysis of salivary gland tissue from six SjD
