Ten different killer sensitivity types are distinguished within Cryptococcus neoformans, namely four in var. neoformans and six in var. gattii. All strains of the var. gattii investigated were inhibited by killer toxins of C. laurentii CBS 139, whereas those of the var. neoformans were not. Killer sensitivity patterns are an easy-to-use method to differentiate between the two varieties of the clinically important yeast C. neoformans, and may be of help in epidemiological surveys.

Adhesion is regarded as an important step in the pathogenesis of several microorganisms. Thus, the ability to recognize extracellular matrix proteins, such as laminin or fibronectin, has been correlated with invasiveness. Studying the already characterized laminin-binding protein of Paracoccidioides brasiliensis, the 43 kDa glycoprotein (gp43), we evaluated whether MAb 1.H12, raised against the laminin-binding protein from Staphylococcus aureus, cross-reacts with that fungal protein. Cryptococcus neoformans is an opportunistic pathogen infecting AIDS patients. However, infection caused by C. neoformans var. gattii in AIDS patients is reportedly rare. We report herein the first culture proven C. neoformans var. gattii serotype B infection in an AIDS patient in India.

The glycoprotein gp43 from Paracoccidioides brasiliensis is the main antigenic component in paracoccidioidomycosis (PCM) because it is recognized by 100% of PCM patients. It has also been shown that different fungal strains produce gp43 with at least four isoform profiles. In this study, different isoform profiles from gp43, with pIs ranging from 5.8 to 8.5, were affinity purified from various P. brasiliensis (B-339, S.S., 1925 and I9) exoantigens. Because of the isoform heterogeneity, we questioned whether those isoform profiles could be similarly recognized by acute or chronic PCM patients.

A case is reported in a 9-year-old Anglo-Arab horse with guttural pouch mycosis caused by Emericella nidulans. Acute death occurred by exsanguination following erosion of the external carotid artery. Histopathological examination of the mycotic plaque demonstrated septate hyphae, conidial heads, hülle cells and mature cleistothecia containing characteristic ascospores. Specific identification was confirmed by culture. In accordance with previous reports, Emericella nidulans should be considered as the major agent of guttural pouch mycosis in Equidae.

Necrotizing fasciitis caused by the zygomycete Apophysomyces elegans of the anterior abdominal wall of a young healthy woman following a lower segment caesarean section is reported. Early clinical diagnosis and laboratory identification followed by appropriate management involving extensive tissue debridement and adequate doses of amphotericin B were eventually successful in controlling the infection and saving her life.

Expression of the 43 kDa glycoprotein (gp43) was analysed in several Paracoccidioides brasiliensis isolates. Using one- and two-dimensional analysis of crude cellular extracts, it was shown that protein expression in yeast and mycelium was dependent on the isolate analysed. In two strains, in both yeast and mycelium cells. gp43 was present, whereas expression was restricted to the yeast phase of two other strains. The clinical implications of this phase-specific gp43 expression are uncertain.

We report the identification of Blastomyces dermatitidis by microscopic examination of a direct faecal smear from a dog with pulmonary blastomycosis. A simultaneously obtained faecal culture grew Blastomyces dermatitidis. The fungus was also cultured from a transtracheal sample from this same dog. This report suggests that yeast-phase cells of B. dermatitidis may be recovered in the stool of dogs with pulmonary blastomycosis following transit through the gastrointestinal tract of swallowed infected sputum. Implications regarding the ecology of Blastomyces dermatitidis are discussed.

By using multilocus enzyme electrophoresis (MLEE) we have analyzed the genetic diversity encountered among chlamydospore-positive Candida albicans strains. While the type II strains of the former C. stellatoidea were genetically indistinguishable from those of C. albicans, type I strains constituted a distinct subgroup compared with C. albicans strains. Nevertheless, all these strains remained genetically very closely related compared with other species of Candida (e.g. C. tropicalis, C. krusei and C. glabrata). Using a class-specific enzyme immunoassay IgG and IgA anticryptococcal antibody was measured in 400 serum specimens at 1 week to 11.7 years after diagnosis from 43 immunocompetent subjects with confirmed active cryptococcosis. We determined the ploidy of Cryptococcus neoformans (28 strains) isolated from patients and nature. Fungal isolates from the peritoneal cavity, kidney and liver, respectively, of three premature Australian babies are described as Rhizopus azygosporus. All three babies died. The 18S ribosomal RNA gene of Cryptococcus neoformans was amplified by polymerase chain reaction (PCR). The primers CPL1 and CPR4 were tested for their ability to amplify DNA from 30 strains of C. neoformans and 27 specimens of cerebrospinal fluid (CSF) from patients with cryptococcal meningitis.
