Developmental exposure to cobalt chloride affected mouse testis via altered iron metabolism in adulthood.

INRODUCTION: Cobalt (Co) is known to interfere with iron (Fe) metabolism that is essential for differentiating male germ cells. Our aim was to study the effect of developmental chronic cobalt exposure on mouse testis through changes in iron homeostasis in adulthood. METHODS: Pregnant ICR mice were exposed to 75 mg (low dose) or 125 mg (high dose)/kg b.w. cobalt chloride (CoCl2) with drinking water for 3 days before delivery and treatment continued until postnatal day 90 of the pups. Age-matched control animals obtained regular tap water. Testes of control and Co-treated mice were processed fo

Deficient LRRC8A-dependent volume-regulated anion channel activity is associated with male infertility in mice.

Источник пометил публикацию как отозванную

Ion channel-controlled cell volume regulation is of fundamental significance to the physiological function of sperm. In addition to volume regulation, LRRC8A-dependent volume-regulated anion channel (VRAC) activity is involved in cell cycle progression, insulin signaling, and cisplatin resistance. Nevertheless, the contribution of LRRC8A and its dependent VRAC activity in the germ cell lineage remain unknown. By utilizing a spontaneous Lrrc8a mouse mutation (c.1325delTG, p.F443*) and genetically engineered mouse models, we demonstrate that LRRC8A-dependent VRAC activity is essential for male g

Emerging Contaminants and Male Reproductive Health: Mechanistic Insights, Clinical Evidence, and Research Priorities.

Male infertility contributes to around 40-50% of the infertility cases worldwide. Emerging environmental pollutants (microplastics, engineered nanoparticles, radiofrequency electromagnetic radiation (RF-EMR), pharmaceuticals and personal care products (PPCPs)) have received significant attention because of their environmental persistence and potential adverse effects on male reproductive health. This review critically synthesizes evidences from experimental, epidemiological and systematic studies, with emphasis on mechanistic pathways. Micro- and nanoplastics are now detected in human blood, t

A novel inducible Cre mouse model for genetic manipulation in nonciliated cells of efferent ductules†.

The efferent ductules (ED) function as essential conduits for spermatozoa transport from the rete testis to the epididymis. The nonciliated and ciliated cells within the efferent ductal epithelium are responsible for fluid reabsorption and stirring the luminal fluid to prevent sperm agglutination, respectively. Dysfunction in either cell type can result in obstructive azoospermia. To systematically investigate the molecular mechanisms underlying efferent ductal development and function, we successively developed two novel knock-in mouse models via CRISPR/Cas9-mediated insertion of Cre-P2A or C

Melatonin in Livestock Production: Bridging Chronobiology, Health, and Sustainable Outcomes.

Melatonin (N-acetyl-5-methoxytryptamine) is a phylogenetically ancient hormone synthesized primarily in the pineal gland in response to light-dark cycles, where it regulates circadian rhythms, but it is also produced in peripheral tissues, including the gastrointestinal tract. In livestock, melatonin extends beyond chronobiology to modulate immune function, reproduction, metabolism, and adaptation to environmental stressors such as heat, transport, and high production intensity. This review consolidates current knowledge on melatonin's application in animal nutrition, tracing from its discover

The Generation of a Testicular Peritubular Cell Line from Giant Pandas (Ailuropoda melanoleuca).

Giant pandas (Ailuropoda melanoleuca), a flagship endangered species under priority preservation in China, remain poorly understood in terms of their testicular physiology and the mechanisms underlying spermatogenesis. Testicular peritubular cells (TPTCs), a crucial somatic cell type surrounding seminiferous tubules, secrete growth factors such as GDNF and CSF1 and release inflammatory factors such as IL-6 and IL-1β, contributing to the testicular niche and immune homeostasis. The contraction of TPTCs also facilitates the transport of sperm towards the epididymis. Nonetheless, TPTCs tend to un

Canine spermatozoa-What do we know about their morphology and physiology? An overview.

Spermatozoa are unique cells because of their morphological and physiological characteristics. They are produced during the process called spermatogenesis. Spermatogenesis consists of three phases: spermatocytogenesis, spermiogenesis and spermiation, during which spermatozoa undergo several changes. Spermatogenesis takes place within the seminiferous tubules containing two types of cells-the germ cells and the Sertoli cells-that alongside the Leydig cells, which play an important role when it comes to normal fertility. Everything is regulated by the hypothalamic-pituitary-gonadal axis and spec

Exposure to non-ionizing electromagnetic radiation of public risk prevention instruments threatens the quality of spermatozoids.

The use of artificial insemination in cattle breeding has evolved to global extent, and insemination doses are often shipped via air transport which requires strict radiation-based examinations. For the determination of effect of non-ionizing radiation (NIR), to which are beings frequently exposed due to protection of airport or cultural event security, freshly ejaculated and cryopreserved bovine spermatozoa were used as experimental model. Following radiation with hand-held metal detector in various exposition times (0, 10 s, 15, 30 and 60 min-groups FR, FR10, FR15, FR30 and FR60) the spermat

Morphological and ultrastructural study of the efferent ductules in the Chinese soft-shelled turtle Pelodiscus sinensis.

Comparative study of the turtle excurrent duct system increases our understanding the evolution of sperm motility and fertility maintenance in higher vertebrates. Therefore, in this study we observed the histology and ultrastructure organization of efferent ductules in the Pelodiscus sinensis using light and transmission electron microscopy. The efferent ductules are extra- testicular and 22-28 in number originate from rete testis. The epithelium is entirely composed of two types of cells, the predominant non-ciliated and ciliated cells. The ciliated cells have long cilia that protrude into th

Open chromatin mapping identifies transcriptional networks regulating human epididymis epithelial function.

The epithelium lining the epididymis in the male reproductive tract maintains a luminal environment that promotes sperm cell maturation. This process is dependent on the coordinated expression of many genes that encode proteins with a role in epithelial transport. We previously generated genome-wide maps of open chromatin in primary human epididymis epithelial (HEE) cells to identify potential regulatory elements controlling coordinated gene expression in the epididymis epithelium. Subsequent in silico analysis identified transcription factor-binding sites (TFBS) that were over-represented in

A mutation in the mouse ttc26 gene leads to impaired hedgehog signaling.

The phenotype of the spontaneous mutant mouse hop-sterile (hop) is characterized by a hopping gait, polydactyly, hydrocephalus, and male sterility. Previous analyses of the hop mouse revealed a deficiency of inner dynein arms in motile cilia and a lack of sperm flagella, potentially accounting for the hydrocephalus and male sterility. The etiology of the other phenotypes and the location of the hop mutation remained unexplored. Here we show that the hop mutation is located in the Ttc26 gene and impairs Hedgehog (Hh) signaling. Expression analysis showed that this mutation led to dramatically r

Aquaporin 9 expression in the developing rat epididymis is modulated by steroid hormones.

Fluid and solute transport across the epithelium of the male excurrent duct is important for sperm maturation and storage. Aquaporin 9 (AQP9), which allows permeation of water and neutral solutes, is abundant throughout the male reproductive tract, where it is expressed at the apical membrane of rat epididymal principal cells as early as at 1 week of age. We evaluated the effect of neonatal exposure to: 1) a GNRH antagonist (GNRHa); 2) diethylstilbestrol (DES); 3) ethinyl estradiol (EE); 4) DES plus testosterone (DES+TE); and 5) the anti-androgen flutamide on AQP9 expression in the epididymis

A biphasic pattern of 45Ca2+ uptake by mouse spermatozoa in vitro correlates with changing functional potential.

Mouse sperm capacitation in vitro, leading to hyperactivated motility, acrosomal exocytosis and rapid fertilization, takes approximately 120 min in a medium containing sufficient Ca2+. During that period, spermatozoa incubated in 45Ca2+ exhibited a biphasic pattern of Ca2+ uptake, with the first and lower peak occurring from 10 to 50 min and the second and higher peak from 60 to 90 min. When the exogenously supplied glucose was reduced from 5.56 mmol l-1 to 5.56 mumol l-1, the latter supporting capacitation but not fertilization, only the first peak of 45Ca2+ uptake was observed. Increasing th

Uterine infection-induced transcriptomic changes in the bovine endometrium.

BACKGROUND: Uterine infections negatively impact cow health and productivity, increasing herd health costs, infertility, and culling. They also interfere with immune efficiency, sperm transport, embryonic development, and implantation, thus leading to pregnancy failure and infertility. Most previous studies have focused on transcriptomic changes induced by uterine infection in dairy cows. The present study aimed to investigate the changes in the endometrial transcript caused by uterine infections in beef cows. METHODS: To achieve this, we examined the reproductive tract of over 200 Angus cows…

Seminal plasma alters the endometrium during early embryo development in cattle†.

Seminal plasma is commonly known to serve as a survival medium to transport sperm cells through the male and then female reproductive tract. In rodents, swine and humans, seminal plasma actively modulates the cellular and molecular maternal environment at the time of conception and early embryo development. Artificial insemination in cattle minimizes female exposure to seminal plasma and deposits semen directly into the uterus, bypassing the vagina as the natural site of insemination. Seminal plasma exposure at insemination increases heifer birth weight, but the mechanisms are unknown. We hypo

PRMT3 restricts porcine epidemic diarrhea virus replication by disrupting the interaction between VAPA and the viral nucleocapsid protein.

Porcine epidemic diarrhea virus (PEDV) represents a severe threat to the global swine industry. Its infection process involves intricate virus-host interactions and immune evasion mechanisms, but effective therapeutic targets remain elusive. In this study, we identified protein arginine methyltransferase 3 (PRMT3) as a novel regulatory factor that significantly modulates PEDV infection via genome-wide CRISPR/Cas9 knockout library screening. Knockout or inhibition of PRMT3 markedly enhanced PEDV infection in multiple cell lines, including LLC-PK1, IPEC-J2, and primary porcine intestinal epithel

MiRNA-seq and mRNA-seq revealed the mechanism of fluoride-induced cauda epididymal injury.

The widespread presence of fluoride in water, food, and the environment continues to exacerbate the impact of fluoride on the male reproductive health. However, as a critical component of the male reproductive system, the intrinsic mechanism of fluoride-induced cauda epididymis damage and the role of miRNAs in this process are still unclear. This study established a mouse fluorosis model and employed miRNA and mRNA sequencing; Evans blue staining, Oil Red O staining, TEM, immunofluorescence, western blotting, and other technologies to investigate the mechanism of miRNA in fluoride-induced caud

Chorein deficiency promotes ferroptosis.

Ferroptosis is a type of programmed cell death owed to an intracellular accumulation of iron resulting in the generation reactive oxygen species, which in turn can cause peroxidation of plasma membrane lipids and ultimately result in cell death. We investigated the potential involvement of VPS13A deficiency in ferroptosis. The VPS13A gene encodes for chorein, and its deficiency is a molecular cause of chorea-acanthocytosis (ChAc), a Huntington-like disease with neurodegeneration in the striatum. In our previous study, we found male infertility characterized by increased malondialdehyde stainin

Seminal plasma modulates expression of endometrial inflammatory meditators in the bovine†.

Seminal plasma has conventionally been viewed as a transport and survival medium for mammalian sperm; however, its role now extends beyond this process to actively targeting female tissues. Studies in rodents, swine, and humans demonstrate that seminal plasma induces molecular and cellular changes within the endometrium or cervix following insemination. Seminal-plasma-induced alterations to the maternal environment have been theorized to facilitate embryo development, modulate maternal immunity toward the conceptus, and potentially improve pregnancy success. It is unknown if bovine seminal pla
