A novel tetrafunctional reagent, alpha,gamma,alpha',gamma'-tetra-succinimidyl-hexanediamide-di-glutamate ester (HDG(OSu)(4)), was successfully synthesized, and a well-defined cross-linked bovine hemoglobin (mainly 128 kDa) was prepared with this reagent. Horizontal gel electrophoresis has previously suffered from lack of an instrumental design that would allow for application of large sample volumes. The efficacy of second-line antiepileptic drugs (AEDs) was evaluated in the treatment of 66 patients with complex partial seizures who had previously failed first-line AEDs. Methsuximide, valproate, or clorazepate had eliminated seizures in 11% of the patients at the end of the study. However, these good results deteriorated on longer follow-up and were not expected to be permanent. It is recommended that suitable patients with partial epilepsy be referred for surgical evaluation after failing the first-line AEDs, and that second-line AEDs be reserved for nonsurgical candidates.

Twelve new N-aminosuccinimides were synthesized by the condensation of hydrazines with succinic anhydrides in glacial acetic acid. The compounds were evaluated in the maximal electroshock seizure and subcutaneous pentylenetetrazol seizure threshold tests for anticonvulsant activity and in the rotorod test for neurotoxicity in mice. The lowest dose at which several of the compounds exhibited anticonvulsant activity was 300 mg/kg.
