Mouse models for actinic keratosis and squamous cell carcinoma.

This manuscript focuses on the use of mice to study the genetics and biology of cutaneous squamous cell carcinoma (SCC). Mice develop actini … e molecular network, and as with other complex diseases, the picture is being pieced together using systems biology strategies to which mouse tumour models are amenable. Such approaches do not focus on single genes or proteins but try to integrate the complex interactions of many types of genetic and biological information.

New trends in pharmacogenomic strategies against resistance development in microbial infections.

This review summarizes some of the new trends in the fight against drug resistant bacteria. We review Gram-positive (e.g., S.aureus) and Gra … eria, or a new way to attack efflux systems. Typical approaches from pharmacogenomics combined with systems biology and bioinformatics support these routes (simulations, metagenomics and metabolic network modeling), as well as the patient treatment (e.g., haplotyping and immune response).

Cardiopedia-Ligand: A ligand-receptor perturbation atlas of human cardiac organoid function and transcriptional state.

Large-scale perturbation atlases have transformed systems biology, yet no equivalent resource exists for the human heart, where contractile function and transcriptomic state must be measured together. Here, we establish Cardiopedia-Ligand, a comprehensive perturbation-function-transcriptome atlas generated by stimulating human cardiac organoids (hCOs) with 87 ligands targeting 98 cell-membrane receptors expressed in the human heart. We developed an automated high-throughput pipeline enabling individualized contractility measurements and single-organoid mRNA sequencing. We use this pipeline to …

A Brief Overview of Molecular Biology in Pituitary Adenomas with a Focus on Aggressive Lesions.

Arising from the anterior lobe of the hypophysis, pituitary neuroendocrine tumors (PitNETs), previously known as pituitary adenomas, constitute an intricate and heterogeneous entity. Although they are defined as benign pathology, these tumors can often invade neighboring structures and demonstrate aggressive behavior. The continuous advancement of molecular biology has begun to shed light on the genetic mutations behind the development and evolution of this pathology, providing a better understanding. Notwithstanding, the rising occurrence in recent decades can mainly be attributed to advanced

Experimental Evidence for Double Intermolecular Coulombic Decay in Bio-Relevant Molecular Dimers.

We report the experimental observation of double intermolecular Coulombic decay (dICD) and reveal its potential for radiation biology in some prototypical molecular dimers consisting of benzene, pyridine, and water. In dICD, the inner-shell vacancy is filled by an electron from an outer shell and the energy released is transferred to doubly ionize the neighboring molecule with the emission of two low-energy electrons. The system further relaxes by a three-body Coulomb explosion process, e.g., CH_{3}^{+}+C_{5}H_{3}^{+}+C_{6}H_{6}^{+} for benzene dimer. Through multicoincidence momentum imaging,

Culturing and Differentiation of Patient-Derived Ectocervical Epithelial Stem Cells Using Air-Liquid Interphase and Matrigel Scaffold.

The ectocervix acts as a multilayered defense barrier, protecting the female reproductive system from external pathogens and supporting fertility and pregnancy. To understand the complex cellular and molecular mechanisms of cervical biology and disease, reliable in vitro models are vital. We present an efficient method to isolate and cultivate epithelial stem cells from ectocervical tissue biopsies. This method combines enzymatic digestion, mechanical dissociation, and selective culturing to obtain pure ectocervical epithelial cells for further investigation. The protocol accommodates both 2D

Gaining biological perspectives from schistosome genomes.

Characterization of the genomic basis underlying schistosome biology is an important strategy for the development of future treatments and interventions. Genomic sequence is now available for the three major clinically relevant schistosome species, Schistosoma mansoni, S. japonicum and S. haematobium, and this information represents an invaluable resource for the future control of human schistosomiasis. The identification of a biologically important, but distinct from the host, schistosome gene product is the ultimate goal for many research groups. While the initial elucidation of the genome o

Molecular Mechanisms of APOL1-Associated Kidney Disease.

The discovery of apolipoprotein L1 (APOL1) risk polymorphisms has significantly changed our knowledge of kidney disease susceptibility and development in African American populations. Several non-diabetic kidney disorders, such as focal segmental glomerulosclerosis (FSGS), collapsing glomerulopathy, HIV-associated nephropathy (HIVAN), and accelerated chronic kidney disease (CKD) development, are significantly more likely to occur in people with two coding variations, G1 and G2. The significance of context-dependent pathogenic processes is highlighted by the poor penetrance and remarkable pheno

The β Common Cytokine Receptor Family Reveals New Functional Paradigms From Structural Complexities.

Cytokines are small proteins that are critical for controlling the growth and activity of hematopoietic cells by binding to cell surface receptors and transmitting signals across membranes. The β common (βc) cytokine receptor family, consisting of the granulocyte-macrophage colony-stimulating factor (GM-CSF), interleukin (IL)-3, and IL-5 cytokine receptors, is an architype of the heterodimeric cytokine receptor systems. We now know that signaling by cytokine receptors is not always an "all or none" phenomenon. Subtle alterations of the cytokine:receptor complex can result in differential or se

A Robust Marine Collagen Peptide-Agarose 3D Culture System for In Vitro Modeling of Hepatocellular Carcinoma and Anti-Cancer Therapeutic Development.

The development of physiologically relevant three-dimensional (3D) culture systems is essential for modeling tumor complexity and improving the translational impact of cancer research. We established a 3D in vitro model of human hepatocellular carcinoma (HCC) using a marine collagen peptide-based (MCP-B) biomimetic hydrogel scaffold optimized for multicellular spheroid growth. Compared with conventional two-dimensional (2D) cultures, the MCP-B hydrogel more accurately recapitulated native tumor biology while offering simplicity, reproducibility, bioactivity, and cost efficiency. At the Edge of Chaos: Complexity Science and Critical Care in the Era of Artificial Intelligence.

Critical care unfolds at the crossroads of highly complex, nonlinear systems. The intersection of pathophysiology, human behavior, and clinical operations exemplifies this complexity. We can frame critical illnesses along two dimensions of complexity: a dysregulation axis, marked by hub failures, superlinear effects, and system uncoupling; and a temporal axis, defined by unpredictable nonlinear dynamics and path-dependent trajectories. Artificial intelligence (AI) offers tools to make sense of this complexity and can help guide decision-making in the intensive care unit. At the same time, comp

Cardiovascular imaging with total-body PET.

Cardiovascular disease remains the leading cause of morbidity and mortality worldwide. PET enables non-invasive imaging of cardiovascular biology at the molecular level, providing insights into pathological processes that precede structural changes detectable by conventional imaging. While conventional PET has been used to investigate isolated cardiac pathologies, its short axial field-of-view (SAFOV) limits the exploration of multi-organ axes influencing cardiovascular disease. The advent of total-body PET systems represents a major advancement in molecular imaging, providing enhanced sensiti

Engineering the human endometrium at the intersection of development and reproduction.

The human endometrium plays a central role in embryo implantation, pregnancy, and reproductive and systemic health, making it of great biological and clinical interest. Animal models of the endometrium often fail to mimic many intrinsic characteristics at the base of human physiology and pathology. Although two-dimensional cell-culture systems have yielded foundational insights, recent advances in three-dimensional modelling techniques have unlocked long-term culture of hormone-responsive, multicellular systems that more closely reproduce endometrial physiology. Here we trace the evolution of

Multidimensional characterization of inducible promoters and a highly light-sensitive LOV-transcription factor.

The ability to independently control the expression of different genes is important for quantitative biology.
