Paxlovid Is Associated With Lower Rates of Long COVID-19 Smell and Taste Disorders.

BACKGROUND: Research is needed on treatments that prevent progression to long COVID-19 olfactory and gustatory dysfunction, which millions continue to suffer from. We sought to explore the utility of Paxlovid in decreasing rates of long COVID-19 smell and taste loss. DESIGN: This case-control study at a single tertiary medical center examined patients who were acutely infected with COVID-19 and received Paxlovid from December 2021 to September 2023 and age-matched infected patients who did not receive Paxlovid. Occurrence of nasal congestion, rhinorrhea, facial pain/pressure and smell/taste l…

Expression and activity of the umami taste receptor (TAS1R1/TAS1R3) in rat corpus cavernosum.

The activation of umami taste receptors (TAS1R1/TAS1R3) promotes smooth muscle (SM) relaxation in the mouse stomach. The nitric oxide (NO)/cGMP signaling pathway is crucial for penile erection. Phosphodiesterase type 5 (PDE5) inhibitors, which specifically target cGMP, are the primary treatment for erectile dysfunction (ED). However, these drugs are ineffective in a significant number of patients, highlighting the need for alternative pharmacological targets for ED. Since umami taste receptors regulate SM contractility, this study investigates the role of TAS1R1/TAS1R3 in rat erectile tissue. …

Exploring the eating experience of a pneumatically-driven edible robot: Perception, taste, and texture.

This study investigated the effects of animated food consumption on human psychology. We developed a movable, edible robot and evaluated the participants' impressions induced by the visualization of its movements and eating of the robot. Although several types of edible robots have been developed, to the best of our knowledge, the psychological effects associated with the eating of a robot have not been investigated. We developed a pneumatically driven edible robot using gelatin and sugar. We examined its perceived appearance and the participants' impressions when it was eaten. In the robot-ea

Factors associated with loss and recovery of smell and taste after COVID-19 infection.

OBJECTIVE: To identify predictors associated with loss and recovery of smell and taste after COVID-19 infection. METHODS: The Integrated Public Use Microdata Series (IPUMS) 2021 National Health Interview Series (NHIS) database was used to analyze factors associated with loss and recovery of smell and taste in respondents who had a previous COVID-19 infection. Significant variables from univariate analysis were included in a stepwise backward regression model to identify independent predictors. RESULTS: Of the 3844 individuals who answered yes to having contracted COVID-19, 51.1% and 48.9% re…

The role of Eya1 and Eya2 in the taste system of mice from embryonic stage to adulthood.

Members of the Eya family, which are a class of transcription factors with phosphatase activity, are widely expressed in cranial sensory organs during development. However, it is unclear whether these genes are expressed in the taste system during development and whether they play any role in specifying taste cell fate. In this study, we report that Eya1 is not expressed during embryonic tongue development but that Eya1-expressing progenitors in somites or pharyngeal endoderm give rise to tongue musculature or taste organs, respectively. In the Eya1-deficient tongues, these progenitors do not …

Expression of taste sentinels, T1R, T2R, and PLCβ2, on the passageway for olfactory signals in zebrafish.

The senses of taste and smell detect overlapping sets of chemical compounds in fish, e.g. amino acids are detected by both senses. However, so far taste and smell organs appeared morphologically to be very distinct, with a specialized olfactory epithelium for detection of odors and taste buds located in the oral cavity and lip for detection of tastants. Here, we report dense clusters of cells expressing T1R and T2R receptors as well as their signal transduction molecule PLCβ2 in nostrils of zebrafish, i.e. on the entrance funnel through which odor molecules must pass to be detected by olfactor…

Enhancement of taste by retronasal odors in patients with Wolfram syndrome and decreased olfactory function.

Wolfram syndrome is a rare disease characterized by diabetes, neurodegeneration, loss of vision, and audition. We recently found, in a young sample of participants (mean age 15 years), that Wolfram syndrome was associated with impairment in smell identification with normal smell sensitivity and whole-mouth taste function. However, these senses were assessed separately, and it is unknown whether smell-taste interactions are altered in Wolfram syndrome, which was the focus of this study. Participants with Wolfram syndrome (n = 36; 18.2 ± 6.8 years) and sex-age-equivalent healthy controls (n = 34…

Gene expression analyses of TAS1R taste receptors relevant to the treatment of cardiometabolic disease.

The sweet taste receptor (STR) is a G protein-coupled receptor (GPCR) responsible for mediating cellular responses to sweet stimuli. Early evidence suggests that elements of the STR signaling system are present beyond the tongue in metabolically active tissues, where it may act as an extraoral glucose sensor. This study aimed to delineate expression of the STR in extraoral tissues using publicly available RNA-sequencing repositories. Gene expression data was mined for all genes implicated in the structure and function of the STR, and control genes including highly expressed metabolic genes in …

Anatomophysiological relationships and clinical considerations of taste and smell loss in patients with COVID-19.

BACKGROUND: There are numerous conflicting discussions about the outbreak of the new coronavirus 2019 (COVID-19). AIM: To present some anatomical and physiological considerations about two of the symptoms reported by patients: The loss or reduction of smell and taste. METHODS: The loss or reduction of smell and taste is presented in a peculiar way, with some cases of persistence even after COVID-19. For this, it was searched in three databases, PubMed/MEDLINE, Web of Science, and Scopus, using the following keywords: "Smell", "Taste", "Smell AND COVID-19", "Taste AND COVID-19", with no publi…

Dedicated Olfaction and Taste Items do not Improve Psychometric Performance of the SNOT-22.

OBJECTIVE: Previous work has shown the chemosensory dysfunction item of the 22-item Sinonasal Outcome Test (SNOT-22) that assesses problems with "taste/smell" has poor psychometric performance compared with other items on the SNOT-22, which we have hypothesized is due to the simultaneous assessment of two different senses. Our aim was to determine whether distinct smell and taste items in the SNOT-22 would improve psychometric performance. METHODS: One hundred and eighty-one CRS patients were recruited and completed the SNOT-22. Additional items querying problems with the senses of "smell" an…

[Reliability of whole-mouth taste test in assessment of gustatory function in healthy adults].

Objective:The purpose of this study was to evaluate the test-retest reliability of gustatory function using whole-mouth taste tests in healthy adults. Methods:Fifty healthy subjects were tested by whole-mouth taste test. The taste test involved 5 tastants(sour, sweet, salty, umami and bitter) and 7 concentrations. The tastant solutions were administered in a pseudo-random order starting with the lowest concentration. Record the perception and recognition scores of each tastant separately. The same operator performed test again by the same method two weeks later. The relative reliability and ab…

Smell and taste disorders in COVID-19: From pathogenesis to clinical features and outcomes.

Patients with COVID-19 often complain of smell and taste disorders (STD). STD emerge early in the course of the disease, seem to be more common in SARS-CoV-2 infection than in other upper respiratory tract infections, and could in some cases persist for long after resolution of respiratory symptoms. Current evidence suggests that STD probably result from a loss of function of olfactory sensory neurons and taste buds, mainly caused by infection, inflammation, and subsequent dysfunction of supporting non-neuronal cells in the mucosa. However, the possible occurrence of other mechanisms leading t…

Quantitative analysis of taste disorder in COVID-19 patients, the hypersensitivity to salty quality.

Recently, many of the studies have illustrated that the new pandemic SARS-CoV-2 can affect Central Nervous System through the olfactory bulb. In addition to investigating anosmia or hyposmia induced by this virus, a quantitative analysis was needed to clarify the taste and smell disorder of the new coronavirus. The four basic taste quality with five concentrations for sweet, sour, bitter, and salty were administered to 75 subjects divided into three groups: COVID-19 patients with taste disorder, COVID-19 patients without taste disorder, and control group. The results indicated the increment of…

The effect of radiotherapy on taste sensation in head and neck cancer patients - a prospective study.

BACKGROUND: One of the main side effects of head and neck (H&N) radiation therapy (RT) is alteration in taste sensation. It causes significant morbidity and has a major effect on quality of life (QoL). The aim of this study was to prospectively define the effect of RT on taste sensation (general, and four basic tastes) and correlate these findings with changes in saliva secretion and QoL questionnaires. METHODS: Patients with H&N cancer treated with RT, in which the oral cavity was expected to receive a mean dose of 30 Gray (Gy). Patients were evaluated by Whole-Saliva Sialometry, validated T…

Conditioned taste avoidance induced by the combination of heroin and cocaine: Implications for the use of speedball.

Speedball (heroin + cocaine) is a prevalent drug combination among intravenous drug users. Although its use is generally discussed to be a function of changes in the rewarding effects of either or both drugs, changes in the aversive effects of either drug may also be impacted (weakened) by the combination. To address this latter possibility and its potential role in the use of speedball, the present studies examined the interaction of cocaine and heroin in taste avoidance conditioning. In Experiment 1, male Sprague-Dawley rats were given access to a novel saccharin solution and then injected w…
