Canine visceral leishmaniasis (CVL) is a zoonotic disease of great worldwide importance and can be prevented by vaccinating seronegative dogs. The objective of the present systematic review and meta-analysis is to verify the efficacy rate of vaccines tested in dogs against CVL or L. infantum infection. We used PRISMA guidelines for this review and Pubmed, Web of Science, Scopus, Cochrane, Scielo and CABI to find studies about vaccines against CVL in dogs. Articles were analysed and grouped according to the antigens used. The risk of bias analysis was performed using SYRCLE's RoB tool and meta-

Several new antituberculous vaccine candidates that are effective against primary infection in preclinical animal models have now entered the early phases of clinical trials. Many of these clinical trials involve subunit vaccines, recombinant bacillus Calmette-Guérin (BCG), or improvement of BCG immunity by boosting with subunit vaccines or recombinant viral vectors expressing immunodominant TB antigens. The burning question at this stage is: will the current vaccines be effective in the endemic world where the diverse and complex challenges of TB exist? These challenges include protection of

White spot syndrome virus (WSSV) has been extensively investigated since the white spot disease outbreak in shrimp in 1990. These investigations have mainly focused on determining the function of viral structural proteins, and on utilizing the envelope proteins as subunit vaccines to protect the host. These studies have shown that a WSSV vaccine can be a practical and effective approach for controlling WSSV infection. The development of such a vaccine has become an intense focus of research in the field. This research has resulted in significant progress in the development of WSSV vaccines. Ho

Despite progress in the prevention and treatment of infectious diseases, they continue to present a major threat to public health. The frequency of emerging and reemerging infections and the risk of bioterrorism warrant significant efforts towards the development of prophylactic and therapeutic countermeasures. Vaccines are the mainstay of infectious disease prophylaxis. Traditional vaccines, however, are failing to satisfy the global demand because of limited scalability of production systems, long production timelines and product safety concerns. Subunit vaccines are a highly promising alter

The emergence of SARS-CoV-2 and its subsequent variants in addition to the previous SARS-CoV-1 outbreak indicates the importance of developing broadly protective sarbecovirus vaccines. To date, broadly protective vaccines have primarily focused on clade 1 sarbecoviruses including SARS-CoV-2 variants and SARS-like animal viruses. The discovery of clade 2 and clade 3 sarbecoviruses capable of infecting human cells highlights a need to preemptively develop vaccines that can protect against these viruses. Here, we develop stabilized multivalent subunit vaccines from clade 2 and clade 3 sarbecoviru

Intranasal vaccines potentially offer superior protection against viral infections compared with injectable vaccines. The immunogenicity of intranasal vaccines including adenovirus vector (AdV), has room for improvement, while few options are available for safe execution. In this study, we demonstrate that modifying a basic parameter of vaccine formulation, i.e., osmolarity, can significantly enhance the immunogenicity of intranasal vaccines. Addition of glycerol to AdV intranasal vaccine solutions, unlike other viscous additives, enhanced systemic and mucosal antibodies as well as resident me

Influenza virus afflicts millions of people worldwide on an annual basis. There is an ever-present risk that animal viruses will cross the species barrier to cause epidemics and pandemics resulting in great morbidity and mortality. Zoonosis outbreaks, such as the H7N9 outbreak, underscore the need to better understand the molecular organization of viral immunogens, such as recombinant influenza virus hemagglutinin (HA) proteins, used in influenza virus subunit vaccines in order to optimize vaccine efficacy. Here, using cryo-electron microscopy and image analysis, we show that recombinant H7 HA

OBJECTIVES: Existing Brucella vaccines are attenuated and can cause vaccine-associated brucellosis; and these safety concerns have affected their application. Although subunit vaccines have the advantages of safety, efficacy, low cost, and rapid production, they are usually poorly immunogenic and insufficient to trigger persistent immunity. Therefore, we added layered double hydroxide (LDH) as an adjuvant to Brucella subunit vaccine formulations to enhance the immune response to the antigen.
MATERIALS AND METHODS: LDH and Freund's adjuvant were combined with Brucella outer-membrane vesicles (

Nanoparticles have emerged as the platform of choice to improve the efficacy and safety of subunit vaccines. A major challenge underlying the use of nanomaterials in vaccines lies in the particle designs that can efficiently target and activate the antigen-presenting cells, especially dendritic cells. Here we show a toll-like receptor 9 (TLR-9) agonist and antigen coloaded, silica nanoparticles (SiNPs) are able to accumulate in antigen presenting cells in the draining lymph nodes after injection. Vaccine loaded SiNPs led to dramatically enhanced induction of antigen-specific B and T cell respo

The risk of plague as a bioweapon has prompted increasing research efforts to develop plague vaccines due to its extreme virulence and the ease of its transmission. Subunit vaccines that contain F1 and LcrV antigens of Y. pestis have been tested for safety and immunogenicity, but doubts have been raised about whether subunit vaccines that engender antibody responses will protect against pneumonic plague, which requires both humeral and cellular immune responses for protection. The live, attenuated vaccine EV76, a pgm locus deficient Y. pestis strain, has been used for a long time in the Former

Nonpigmented Yersinia pestis (pgm) strains are defective in scavenging host iron and have been used in live-attenuated vaccines to combat plague epidemics. Recently, a Y. pestis pgm strain was isolated from a researcher with hereditary hemochromatosis who died from laboratory-acquired plague. We used hemojuvelin-knockout (Hjv(-/-)) mice to examine whether iron-storage disease restores the virulence defects of nonpigmented Y. pestis. Unlike wild-type mice, Hjv(-/-) mice developed lethal plague when challenged with Y. pestis pgm strains. Immunization of Hjv(-/-) mice with a subunit vaccine that

Molecular adjuvants are vaccine delivery vehicle to increase specific antigens effectiveness. Herein, we concentrated on IgG Fc, an effective molecular adjuvant, to develop novel pseudorabies virus (PRV) subunit vaccines. Two major protective antigen genes of PRV were constructed and linked into the mouse IgG Fc fragment. The gD, gD-IgG2aFc, gB and gB-IgG2aFc proteins were expressed using a baculovirus system. Mice intranasally immunized with gD-IgG2aFc or gB-IgG2aFc subunit vaccine exhibited significantly higher PRV-specific antibodies, neutralizing antibodies and intracellular cytokines than

de Boer and colleagues present a cost-effectiveness analysis based in the Netherlands of two vaccines available for the prevention of herpes zoster. Zostavax® was the first vaccine available for the prevention of herpes zoster in older adults. A live-attenuated vaccine, Zostavax is not free of limitations, which include a relatively low efficacy that wanes over time and its contraindication among immunocompromised individuals. The recently available adjuvanted herpes zoster subunit vaccine Shingrix® overcomes some of these limitations. The herpes zoster subunit vaccine is more efficacious than

BACKGROUND: The African swine fever virus (ASFV), prevalent globally, causes high mortality and morbidity in domestic pigs. However, there is a lack of effective treatment or vaccines against ASFV infection despite the ongoing research in this field.
METHODS: In this systematic review and meta-analysis, we conducted a quantitative evaluation of the efficacy of non-replicating vaccines against ASFV. The vaccine efficacy (VE) was analyzed based on three key disease outcomes: mortality, fever, and clinical symptoms after infection.
RESULTS: The search of relevant electronic databases yielded 23

The heightened concern about the intentional release of variola virus has led to the need to develop safer smallpox vaccines. While subunit vaccine strategies are safer than live virus vaccines, subunit vaccines have been hampered by the need for multiple boosts to confer optimal protection. Here we developed a protein-based subunit vaccine strategy that provides rapid protection in mouse models of orthopoxvirus infections after a prime and single boost. Mice vaccinated with vaccinia virus envelope proteins from the mature virus (MV) and extracellular virus (EV) adjuvanted with CpG ODN and alu…

INTRODUCTION: Animal influenza viruses pose a danger to the general public. Eurasian avian-like H1N1 (EA H1N1) viruses have recently infected humans in several different countries and are often found in pigs in China, indicating that they have the potential to cause a pandemic. Therefore, there is an urgent need to develop a potent vaccine against EA H1N1.
METHODS: In this study, we report the effective intramuscular (i.m.) and/or intranasal (i.n.) vaccination of mice with a subunit influenza vaccine utilizing safe adjuvant gram-positive enhancer matrix (GEM) particles derived from the food-g

Subunit vaccines are among the most useful vaccine modalities; however, their low immunogenicity necessitates the addition of adjuvants. Although adjuvants improve immune responses induced by vaccines, they often cause adverse reactions. To address this, we developed an adjuvant-free subunit vaccine platform that uses pre-existing antibodies generated from past infections or vaccinations as carriers for the delivery of vaccine antigens. Although we have confirmed the usefulness of this platform for nasal vaccines, its suitability as a parenterally injectable vaccine remains uncertain. Here, we
