BACKGROUND: The global burden of liver cirrhosis is increasing, with 2.1 million incident cases and nearly 1.5 million deaths in 2019. Despite the enormous progress in our understanding of the etiology of liver cirrhosis, significant cases of the disease have been reported in Eastern Ethiopia due to unidentified causes. Hence, this study aimed to identify predictors of liver cirrhosis of unknown etiology in Eastern Ethiopia.
METHODS: A score of 7 out of 11 possible points on the ultrasound-based cirrhosis scale was used as a diagnostic criterion to include 127 liver cirrhosis patients. The st

A growing area of research in pharmacology and toxicology is concerned with the role of adrenal glucocorticosteroids (predominantly corticosterone in rats and mice, and cortisol in humans) in modulating toxicological responses. These steroids are secreted from the adrenal cortex, and in laboratory rodents secretion occurs particularly in response to stressful environmental change or noxious challenge, which can include a toxic insult. Glucocorticoids have profound biochemical effects in diverse tissues (e.g., inhibition of DNA and RNA synthesis; effects on carbohydrate metabolism, protein cata

Although selenium was once considered to be a toxic, undesirable and carcinogenic element, it is now recognized as an essential element with anticarcinogenic properties. Epidemiological studies in the United States have shown an inverse relationship between selenium intake and certain forms of cancer in humans, but other factors must be considered since cancer is not higher in people living in selenium-deficient areas of the world (Finland, New Zealand, and Keshan disease area, China). Under most dietary conditions, selenium has been shown to reduce the spontaneous mammary tumor incidence in a

Coix seed, a traditional medicinal and edible crop, is susceptible to mycotoxin contamination posing potential health risks, yet systematic risk assessments for its dual dietary and medicinal pathways remain limited. Fifty batches were collected from five major production regions in China. UPLC-MS/MS was used to quantify eight mycotoxins in raw materials and decoctions. A Monte Carlo simulation model assessed long-term (20-year) health risks via both pathways, and acceptable daily intake (ADI) levels were derived using a combined margin of exposure (MOE) threshold of 10,000. Results indicated

Epidemiological studies have suggested that human exposure to extremely low-frequency electromagnetic fields from the electric power and to mobile phone radiofrequency electromagnetic fields induce an increased risk of developing malignant tumours. However, no adequate laboratory data, in particular long-term carcinogenicity bioassays to support the epidemiological evidence, have yet been available. This motivated the Ramazzini Institute to embark on a first project of four large life-span carcinogenic bioassays conducted on over 7000 Sprague Dawley rats exposed from prenatal life until natura

Although biliary excretion is one of the biological elimination processes for foreign compounds, intake of high-protein diets was hypothesized to enhance their detoxification rates. Hence, this study investigates the effect of differential dietary protein intake on toxicokinetics and biliary excretion in rats following exposure to N-nitrosodiethylamine (NDEA) and aflatoxin B1 (AFB1). The animals were divided into five groups. Groups I and II were exposed to low and high dietary proteins following a single intraperitoneal dose of 43 µg NDEA/kg body weight, respectively. Groups III and IV were e

Surface-enhanced Raman scattering-based lateral flow immunoassays (SERS-LFIAs) have emerged as a promising platform for rapid food safety analysis. However, their quantitative reliability is often compromised by matrix-dependent variations during lateral flow, and the influence of probe size on analytical performance remains insufficiently understood. In this work, Au@DTNB immunoprobes with diameters of 15, 30, and 50 nm were prepared to systematically investigate the effect of nanoparticle size on the quantitative performance of SERS-LFIA for aflatoxin B1 (AFB1) detection. All three probe siz

Aflatoxin B1 (AFB1), a common contaminant in broiler feed, impairs hepatic function and poses a significant threat to the poultry industry by disrupting cellular autophagy. Ferulic acid (FA) is a naturally occurring phenolic compound with antioxidant, anti-inflammatory, and autophagy-modulating properties. However, whether FA alleviates AFB1-induced autophagy impairment in broiler liver remains unclear. In this study, an AFB1-induced liver injury model was established. A series of experiments were conducted to evaluate the effects of AFB1 and FA on autophagy-related processes. The results show

Hepatocellular carcinoma (HCC) represents a global public health burden of increasing magnitude, driven by risk factors including viral hepatitis, metabolic dysfunction-associated fatty liver disease and various environmental carcinogens, which may inactivate key tumor suppressor genes or activates oncogenes. Recent studies have linked various environmental exposures with inactivation of tumor suppressor gene CDKN2A (cyclin dependent kinase inhibitor 2A) and its encoded proteins p16INK4a and p14ARF to various molecular pathways leading to hepatocarcinogenesis. Current studies suggest p16INK4a

BACKGROUND: Guinea has the highest incidence of hepatocellular carcinoma (HCC) in Sub-Saharan Africa (> 21 cases/100,000 habitants/year). This high incidence is largely attributed to the combination of a high prevalence of chronic hepatitis B virus (HBV) infection and significant exposure to aflatoxin B1 (AFB1), a known carcinogen. However, the clinical presentation of HCC in Guinea has not been well-documented to date.
METHODS: We conducted a retrospective analysis of 216 HCC cases from patients who received care between 2015 and 2023 at the Donka University Hospital in Conakry, the main ter

Aflatoxin B1 (AFB1)-induced hepatotoxicity is a common toxic disease in poultry farming. However, there is currently a lack of effective pharmaceutical interventions for treating AFB1. Astaxanthin (AST), a natural carotenoid, exhibits potent antioxidant and immune-enhancing properties. In this study, the effect of AST on AFB1-induced hepatotoxicity was determined using chick embryo liver cells (CELs). The CCK-8 assay results showed that AST significantly attenuated the decrease in viability of CEL cells induced by AFB1, and reduced the activity of the enzymes GOT, GPT, and ALP (P < 0.01). More

The success of oltipraz (OPZ) [5-(2-pyrazinyl)-4-methyl-1,2-dithiole-3-thione] as a chemoprotective agent against aflatoxin B1 (AFB1)-induced hepatocarcinogenesis in the rat is thought to depend principally on its ability to enhance detoxication by inducing phase II enzymes, especially glutathione transferases. However, in primary cultures of human hepatocytes, we recently demonstrated that OPZ also has an important inhibitory effect on the major cytochromes P450 (CYPs) of human hepatic AFB1 metabolism. This has prompted a detailed study of the effect of OPZ on some CYPs involved in metabolism

Oxidative stress is an important molecular mechanism for kidney injury in aflatoxin B1 (AFB1) nephrotoxicity. Nuclear factor erythroid 2-related factor 2 (Nrf2) is a master transcription factor for regulating the cellular oxidative stress response, which has been confirmed in animal models. Lycopene (LYC), a natural carotenoid, has received extensive attention due to its antioxidant effect with the activation of Nrf2. However, the role of LYC in protecting against AFB1-induced renal injury is unknown. To evaluate the chemoprotective effect of LYC on AFB1-induced renal injury, forty-eight male

The dietary supplementation of fat has great potential in countering the toxic effects of aflatoxin (AF) in chickens, but the issue was less researched upon. An experiment was conducted to evaluate the response of broiler chickens to graded levels of AF B1 (0, 150 and 300 ppb) and sunflower oil (SFO) (0, 1.5 and 3.0%) in the diet in a 3 × 3 factorial manner to understand their interaction effects. A total of 360 broiler chickens divided into 9 equal groups were fed the diets during 0 to 35 days of age, and their response was evaluated in terms of performance, serum biochemical profile, organ w

Zearalenone (ZEA) and aflatoxin B₁ (AFB₁) are two main kinds of mycotoxins widely existing in grain and animal feed that cause a lot of economic loss and health problems for animals and humans. In order to alleviate the cytotoxic effects of AFB₁ and ZEA on swine jejunal epithelial cells (IPEC-J2), the combination of a cell-free supernatant of compound probiotics (CFSCP) with mycotoxin degradation enzymes (MDEs) from Aspergillus oryzae was tested. The results demonstrated that coexistence of AFB₁ and ZEA had synergetic toxic effects on cell viability. The cell viability was decreased with mycot

Aflatoxin B(1) (AFB(1)) may be present in moldy dust. Inhalation of contaminated dust particles may result in high local exposure of the nasal mucosa. The present study was designed to assess bioactivation and toxicity of AFB(1) in the nasal mucosa after intranasal administration of the mycotoxin in rats and also to examine if translocation of the mycotoxin occurs from the nasal mucosa to the brain along olfactory neurons. Female Sprague-Dawley rats were given (3)H-AFB(1) (0.2, 1 or 20 microg) intranasally and were sacrificed at various intervals (1 h to 20 d). Tissues were examined autoradiog
