Differential alterations in peripheral tryptophan pathways in methamphetamine versus MDMA users are linked to their contrasting psychiatric symptoms.

Methamphetamine (METH, "Crystal Meth") and 3,4-methylenedioxymethamphetamine (MDMA, "Ecstasy") are two types of substituted amphetamines that share structural-chemical similarities but exhibit contrasting acute and chronic effects including addictive liability. Tryptophan (TRY) pathways are involved in pleiotropic physiological functions at the interface of brain-body connections. Preclinical evidence suggests that amphetamines may modulate these pathways and, thus, indirectly influence brain functions via persistent alterations of peripheral metabolites. However, little is known about alterat

Assessing the Concurrent Validity of the Australian Treatment Outcomes Profile in a Methamphetamine Dependent Treatment-Seeking Population.

INTRODUCTION: The Australian Treatment Outcomes Profile (ATOP) is a brief clinical tool assessing substance use, health and well-being used in Australian alcohol and other drug treatment services. It is validated for use with clients using alcohol, opioids and cannabis, but not yet for clients who primarily use methamphetamine. METHODS: An embedded validation study was undertaken in treatment-seeking adults enrolled in a randomised double-blind placebo-controlled trial of lisdexamfetamine for methamphetamine dependence with sites in New South Wales, South Australia and Victoria. Participant d

Right Atrial Perforation in the Setting of Methamphetamine-Associated Cardiomyopathy.

Right atrial perforation is a rare sequela of a few clinical situations, such as acute myocardial infarction, invasive cardiac procedures, or cardiac malignancies. This case is a novel example of a right atrial perforation due to methamphetamine-associated cardiomyopathy. The decedent was a 44-year-old male with a history of methamphetamine use who suffered a witnessed collapse and was pronounced on scene. His autopsy showed findings consistent with acute and chronic sympathomimetic drug use, such as cardiomegaly with dilated cardiomyopathy, necrosis, perforation of the right atrium, and hepat

Exploring the mechanism of trait depression and cognitive impairment on the formation of among individuals with methamphetamine use disorder under varying degrees of social support.

BACKGROUND: Methamphetamine stands as one of the most widely abused drugs globally. Methamphetamine Use Disorder not only impairs the physical and mental wellbeing of addicts but also elevates their risk of suicide. Despite the high suicide rate among individuals with methamphetamine use disorder, research on their clinical characteristics and suicide risk factors remains scarce. Therefore, it is imperative to investigate the risk factors associated with suicidal ideation in individuals with methamphetamine use disorder. METHODS: Employing Respondent Driven Sampling (RDS), a total of 11,825 i

Sex-specific alterations in brain network topology in methamphetamine use disorder: A graph theory-based fMRI study.

BACKGROUND: Methamphetamine use disorder (MUD) imposes severe neurological and societal challenges, yet the sex-specific alterations in brain network topology remain poorly understood. METHODS: Resting-state fMRI data were acquired from 78 patients with MUD (49 male, 29 female) and 65 demographically matched healthy controls (HCs). Functional connectivity matrices were constructed using RESTplus V1.30, and graph metrics (global efficiency, nodal centrality) were computed using GRETNA V2.0.0. Group comparisons (MUD vs. HCs; male vs. female) and correlation analyses with Barratt Impulsiveness S

Coupling analysis of diffusion tensor imaging analysis along the perivascular space (DTI-ALPS) with abnormal cerebral blood flow in methamphetamine-dependent patients and its application in machine-learning-based classification.

BACKGROUND: Diffusion tensor imaging (DTI) analysis along the perivascular space (ALPS) index is currently widely employed to evaluate the neurophysiological activity in various neuropsychiatric disorders. However, there remains a scarcity of studies assessing the DTI-ALPS index in individuals with methamphetamine (MA) dependence. Recent studies on animals have demonstrated a significant correlation between glymphatic activity and alterations in cerebral blood flow (CBF). Hence, investigating the DTI-ALPS index and its coupling with CBF could yield novel insights for MA-dependent patients. ME

Childhood abuse and craving in methamphetamine-dependent individuals: the mediating role of alexithymia.

Individuals with a history of childhood abuse (CA, including neglect and abuse by caregivers before the age of 18 years) have more severe substance dependence problems than those without a history of childhood abuse. However, whether a history of CA exacerbates craving and the mechanism of this effect remain largely unknown. The aim of this study was to explore the role of alexithymia in the effects of CA on craving in a large sample of methamphetamine-dependent individuals based on latent vulnerability theory. A total of 324 methamphetamine-dependent individuals who met DSM-5 criteria for sub

Lycium barbarum polysaccharide mitigated methamphetamine addiction and altered methamphetamine-induced gut microbiota dysbiosis.

Methamphetamine (MA) is a highly addictive mental stimulant, and MA abuse remains a significant public health problem worldwide, while effective treatment options are limited. Lycium barbarum polysaccharide (LBP), a major effective component extracted from Lycium barbarum, has potential health-promoting effects on the nervous system; however, its role in MA dependence remains unclear. In this study, the conditioned place preference (CPP) of MA addiction in adult male mice was established to detect changes in gut microbiota profiles after LBP treatment through 16S rRNA gene sequencing. Our resu

Oxytocin as an adolescent treatment for methamphetamine addiction after early life stress in male and female rats.

Early life stress (ELS) is associated with perturbed neural development and augmented vulnerability to mental health disorders, including addiction. How ELS changes the brain to increase addiction risk is poorly understood, and there are no therapies which target this ELS-induced vulnerability. ELS disrupts the oxytocin system, which can modulate addiction susceptibility, suggesting that targeting the oxytocin system may be therapeutic in this ELS-addiction comorbidity. Therefore, we determined whether adolescent oxytocin treatment after ELS could: (1) reduce vulnerability to anxiety, social d

CART in energy balance and drug addiction: Current insights and mechanisms.

Cocaine amphetamine related transcript (CART) is a neuropeptide first isolated and sequenced from ovine hypothalamus and later shown to have functions related to drug taking, drug seeking, and energy balance. Here we review the molecular features of CART, its anatomical distribution and provide an update on CART function in energy balance control and addiction. A common theme across these topics is that CART has diverse roles and functions depending on both the specific cells in which it is expressed as well as the location of these cells in distinct midbrain, hypothalamic, thalamic, and stria

Methamphetamine drug abuse and addiction: Effects on face asymmetry.

The abnormal aging mechanism associated with drug abuse results in poor performance of face recognition systems on illicit drug addicts (mainly methamphetamine). Consequently, the high correlation between drug addiction and crime exaggerates the urge for further investigations to originate and overcome this problem. Concurrently, face asymmetry was found to play a significant role in face recognition and age estimation. Therefore, facial asymmetry assessment for meth-addicts is highly serviceable, acknowledging how meth addiction accelerates biological aging and causes severe face distortion.

Longitudinal associations between outpatient medical care use and substance use among rural stimulant users.

BACKGROUND: Negative views toward substance use treatment among some rural substance users and limited treatment resources in rural areas likely affect substance use utilization. It is therefore important to determine whether accessing healthcare options other than substance use treatment, specifically outpatient medical care (OMC), is associated with reductions in substance use. OBJECTIVES: We examined whether use of OMC was associated with reductions in substance use among rural substance users over a three-year period. We also explored whether substance user characteristics, including subs

Acute buspirone dosing enhances abuse-related subjective effects of oral methamphetamine.

There is not an approved pharmacotherapy for treating methamphetamine use disorder. This study sought to determine the effects of acute buspirone treatment on the subjective and cardiovascular effects of oral methamphetamine in order to provide an initial assessment of the utility, safety, and tolerability of buspirone for managing methamphetamine use disorder. We predicted that acute buspirone administration would reduce the subjective effects of methamphetamine. We also predicted that the combination of buspirone and methamphetamine would be safe and well tolerated. Ten subjects completed th

Survival Tactics and Strategies of Methamphetamine-Using HIV-Positive Men Who Have Sex with Men in San Diego.

In this article, two ways that HIV-positive drug users survive under the supervision of law enforcement agencies, community health organizations, and social welfare offices are differentiated. First, strategies are long-ranging and often carefully planned, and they involve conscious utilization and manipulation of bureaucratic processes. Second, tactics are short-ranging and often haphazard, and they are used to survive on daily or weekly bases, with entrenched problems and structural solutions avoided or ignored. Data from three years of ethnographic fieldwork with 14 methamphetamine-using HI
