Global epidemic studies have suggested that coffee consumption is reversely correlated with the incidence of type 2 diabetes mellitus (T2DM), a metabolic disease. The misfolding of human islet amyloid polypeptide (hIAPP) is regarded as one of the causative factors of T2DM. Coffee extracts have three major active components: caffeine, caffeic acid (CA), and chlorogenic acid (CGA). In this study, the effects of these major coffee components, as well as dihydrocaffeic acid (DHCA) (a major metabolite of CGA and CA), on the amyloidogenicity of hIAPP were investigated by thioflavin-T based fluoresce…

Alzheimer disease (AD) is a devastating neurodegenerative disease with complex and strong genetic inheritance. Four genes have been establis … ounting evidence suggests that the excessive accumulation of Abeta, the proteolytic product of beta-amyloid precursor protein (APP), is the primary AD pathological event. In this study, we ask whether ATXN1 may lead to AD pathogenesis by affecting Abeta and APP processing utilizing RNA interference in a human neuronal cell model and mouse primary cortical …

Cerebral microvascular amyloid beta protein (Abeta) deposition and associated neuroinflammation are increasingly recognized as an important component leading to cognitive impairment in Alzheimer's disease and related cerebral amyloid angiopathy (CAA) disorders. Transgenic mice expressing the vasculotropic Dutch/Iowa (E693Q/D694N) mutant human Abeta precursor protein in brain (Tg-SwDI) accumulate abundant cerebral microvascular fibrillar amyloid deposits exhibiting robust neuroinflammation. In the present study, we sought to determine if the unique amyloid pathology of Tg-SwDI mice was associat…

Transgenic mice expressing the amyloidogenic human islet amyloid polypeptide (hIAPP) in their islet beta-cells are a model of islet amyloid formation as it occurs in type 2 diabetes. Our hIAPP transgenic mice developed islet amyloid when fed a breeder chow but not regular chow. Because the breeder chow contained increased amounts of fat, we hypothesized that increased dietary fat enhances islet amyloid formation. To test this hypothesis, we fed male hIAPP transgenic and nontransgenic control mice diets containing 15% (low fat), 30% (medium fat), or 45% (high fat) of calories derived from fat f…

We have created early-onset transgenic (Tg) models by exploiting the synergistic effects of familial Alzheimer's disease mutations on amyloid beta-peptide (Abeta) biogenesis. TgCRND8 mice encode a double mutant form of amyloid precursor protein 695 (KM670/671NL+V717F) under the control of the PrP gene promoter. Thioflavine S-positive Abeta amyloid deposits are present at 3 months, with dense-cored plaques and neuritic pathology evident from 5 months of age. TgCRND8 mice exhibit 3,200-4,600 pmol of Abeta42 per g brain at age 6 months, with an excess of Abeta42 over Abeta40. High level productio…

Type 2 diabetes is characterized by islet amyloid deposits, which are primarily composed of the amyloidogenic human form of islet amyloid polypeptide (IAPP, amylin). The mechanism of islet amyloido-genesis is not known, but other products (e.g., apolipoprotein E and perlecan) contained within islet amyloid may be necessary. Because rodent IAPP does not form islet amyloid, the currently available beta-cell lines are not useful for studying processes involved in amyloid formation. To develop a suitable in vitro cell system for the study of islet amyloid formation, we generated two new beta-cell …

An insulin-deficient 51-year-old man was put on dietary therapy and sulfonylurea (SU). Although there was good glycemic control for 2 years, … e pancreas showed marked atrophy (32 g) with extensive fatty degeneration. Islets replaced by islet amyloid polypeptide (IAPP)-positive amyloid (IAPP-AM) amounted to 77% in the tail, 74% in the body, and 73% in the head of the pancreas. All islets were positive for IAPP-AM throughout the pancreas, except for a pancreatic polypeptide-rich lobe, where none …

Amyloid myopathy is an etiologically heterogeneous group of disorders associated with amyloid deposition in skeletal muscle. The great majority of cases reflect muscle involvement in systemic amyloidosis, particularly immunoglobulin light-chain amyloidosis and hereditary amyloidosis. Occasionally, amyloid myopathy presents as an isolated myopathy associated with muscle proteinopathies. Clinical manifestations are generally nonspecific. Muscle weakness is the most common presentation; however, many patients with biopsy-confirmed amyloid deposition lack objective weakness, as amyloid within musc…

Amyloid-β (Aβ) aggregation is considered a central hallmark in the pathophysiology of Alzheimer's disease (AD). Aβ protein aggregates disrupt synaptic architecture, calcium homeostasis, and mitochondrial function, leading to excitotoxicity and synaptic plasticity deficits. Animal models and human studies reveal that Aβ-induced alterations in synaptic activity and neuronal circuit function appear before irreversible damage and the onset of cognitive impairment. This review examines the multifaceted effects of Aβ on synaptic and neuronal circuits across its distinct aggregation states, including…

Cerebral amyloid angiopathy (CAA) is a cerebrovascular disorder characterized by the deposition of amyloid-β (Aβ) in the walls of leptomeningeal and cortical blood vessels that increases risk of intracerebral hemorrhages and progressive cognitive decline. More than 90% of individuals with Alzheimer's disease (AD) exhibit some level of CAA. Notably, in the new era of disease-modifying treatments for AD, CAA is a significant risk factor for amyloid-related imaging abnormalities (ARIA), an adverse event associated with anti-amyloid treatments. Therefore, there is great need for accessible, reliab…

Given the multi-organ involvement of transthyretin (ATTR) amyloidosis, associated symptoms such as peripheral neuropathy, biceps tendon rupture, lumbar stenosis, and bilateral carpal tunnel syndrome can develop concurrently with cardiac disease or even precede the cardiac diagnosis by up to a decade. Screening of tissue obtained opportunistically during an otherwise indicated procedure or diagnostic test has shown promise for the early identification of nascent cardiac amyloidosis (CA) in patients who might otherwise remain undiagnosed for years. This review aims to: (1) briefly summarize the …

Alzheimer's disease (AD) is a complex and multifactorial disorder that affects all races and genders. Genetic traits influenced by lifestyle and environment lead to a tremendous amount of heterogeneity in Alzheimer's disease onset and severity. Regardless of these unique contributing factors, Alzheimer's disease is traditionally met with amyloid-beta plaque formation in the central nervous system. In this commentary, we shed light on the growing literature surrounding amyloid-beta's ability to act as an antimicrobial peptide in the central nervous system's innate immune response to pathogenic …

The amyloid precursor protein (APP) is a transmembrane protein widely expressed throughout the brain, where it plays critical roles in both physiological and pathological states. APP undergoes complex post-translational processing by various secretases, a process that can lead to amyloid plaque formation via its amyloidogenic pathway. Consequently, APP has been extensively studied in the context of Alzheimer's disease (AD). However, emerging evidence highlights its physiological functions and the diverse roles of its cleavage fragments. This review explores the dual role of APP and its fragmen…

Mammary tumor-associated amyloidosis in dogs has been reported in several studies, but its incidence and predisposing conditions have not been investigated. We examined 252 mammary masses from 183 dogs to determine the prevalence of amyloid deposition and elucidate etiologic factors. Histologically, amyloid deposition was observed in 88 of 252 (35%) mammary masses. Amyloid deposition patterns were categorized into 3 types: stromal deposition (ST), deposition in corpora amylacea (CA), and precipitation within luminal secretions (PR). Of the 88 amyloid deposition cases, 49 were intra-tumoral, es…

Transthyretin amyloid cardiomyopathy (ATTR-CM) is an infiltrative cardiomyopathy that results from myocardial deposition of misfolded transthyretin (TTR) protein. The biology of amyloid formation has been elucidated resulting in several effective therapeutic strategies. Accordingly, the therapeutic landscape for ATTR-CM is rapidly evolving, with multiple disease-modifying therapies (DMTs) approved and others anticipated to be imminently available. Currently, DMT strategies involve either stabilization of TTR, thereby inhibiting misfolding, or reduction of hepatic TTR production, and antibodies…
