Antimicrobial effect of chlorhexidine in a controlled release delivery system.

Chlorhexidine is widely used as a mouth rinse in the prevention and treatment of periodontal diseases and dental caries. The purpose of the present study was to evaluate the in vitro antimicrobial effect of chlorhexidine in a controlled release delivery system. The controlled release dispenser comprised a polymeric inner core matrix containing the medicament with an outer vinyl membrane controlling the drug release. The effect on the following bacteria was studied: Actinobacillus actinomycetemcomitans, Actinomyces viscosus, Streptococcus mutans, Wolinella recta, Bacteroides gingivalis, Bactero…

In vitro activity of CGP 31523A, a broad-spectrum cephalosporin, in comparison with those of other agents.

The activity against Bacteroides fragilis was modest (MIC90, 32 micrograms/ml). The susceptibility of known beta-lactamase-producing strains suggested that CGP 31523A was resistant to many beta-lactamases (but not those of Bacteroides fragilis). The serum protein binding of CGP 315 …

Premature labor. II. Bacterial sources of phospholipase.

Human term labor is thought to be initiated by amniotic and chorionic phospholipase A2, an enzyme that liberates arachidonic acid esters from the phospholipids of these membranes, leading to the synthesis of prostaglandins by the placental membranes. The striking association of premature labor with intrauterine infection or contamination, urinary tract infection, and early neonatal sepsis led us to study the microorganisms present in these infections for phospholipase A2 activity. Activity was found in Bacteroides fragilis, Peptostreptococcus, Fusobacterium necrophorum, Streptococcus viridans,…

The syndrome of inappropriate secretion of antidiuretic hormone associated with anaerobic thoracic empyema.

The syndrome of inappropriate secretion of antidiuretic hormone has been associated with many pulmonary diseases, including tuberculosis and bacterial and viral pneumonia: however, it has not been reported with anaerobic infections or empyema in the absence of pneumonia. We report a patient with empyema due to Bacteroides melaninogenicus, Bacteroides oralis, and Peptostreptococcus who developed the syndrome. Eight hours before the start of therapy, his serum sodium concentration was 127 mEq per liter; serum osmolality, 255 mOsm per kg; urine osmolality, 522 mOsm per kg; urinary sodium concentr…

[The ultrastructure of anaerobes isolated from the urogenital tract of women].

Electron microscopic (ultrathin sections) examination of culture strains of vaginal Gardnerella and bacteroids isolated in aerobic conditions from urogenital tract of women suffering from Gardnerella infection was carried out. Structural analysis of the bacteria is presented, i.e. their shape, size, morphology, and features of multiplication.

A gut microbial odd-chain fatty acid alleviates atherosclerosis in mice.

The gut microbiota plays a pivotal part in human health, yet the molecular mechanisms that underlie its effects are largely unexplored. Bacteroides, a dominant genus in the human gut microbiota, is depleted in patients with atherosclerosis1, but its causal relationship with disease remains unclear. Here, using a mouse model, we show that administration of Bacteroides uniformis alleviates atherosclerosis through the upregulation of hepatic low-density lipoprotein receptor expression. Bioactivity-guided screening revealed pentadecanoic acid (PA, C15:0), a saturated odd-chain fatty acid, as a pri…

Integrative metagenomics and structural bioinformatics identify explainable gut microbial variants associated with Crohn's disease.

Metagenomics has revealed disease-associated shifts in microbial taxa and functions in inflammatory bowel disease (IBD) patients. However, the role of genomic variation in gut commensals remains poorly understood. Here, we integrated metagenomic profiling, variant calling, and structural bioinformatics to identify disease-associated variants in the gut microbes. Crohn's disease (CD) and ulcerative colitis (UC) showed significant negative associations with Bacteroides uniformis, Bacteroides vulgatus, and Eubacterium rectale. These bacteria exhibited 190,712 single-nucleotide polymorphisms, incl…

Intratumoral microbiota and host genotype cooperatively shape neutrophil cytotoxic functions in colorectal cancer.

The role of tumor-associated neutrophils (TANs) infiltrating colorectal cancers (CRCs) is still debated. Here, we unravel that TAN recruitment and functions are modulated by intratumoral microbiota and only defined bacterial species can unleash neutrophil cytotoxic potential. Fusobacterium nucleatum promotes the production of neutrophil-recruiting chemokines by tumor cells and enhances neutrophil migration more efficiently than Bacteroides fragilis. Importantly, Fusobacterium nucleatum, but not Bacteroides fragilis, triggers neutrophils to release cytotoxic proteins showing tumoricidal activit…

Detection and isolation of protein susceptible indigenous bacteria affected by dietary milk-casein, albumen and soy-protein in the caecum of ICR mice.

In this study, we fed ICR mice with a high-sucrose diet containing 20% w/w of milk-casein (MC), egg-white (EW), or soy-protein (SP) for 14 days in order to detect the presence of protein-susceptible gut indigenous bacteria (P-SIB). The caecal microbiome was examined by 16S rDNA amplicon sequencing using a next-generation MiSeq system. Principal coordinate analysis of the operational taxonomic units (OTUs) revealed that the microbiomes differed among the three groups. Typical SIB found in the MC-fed group were Bacteroides acidifaciens-, Bacteroides sartorii-, Eisenbergiella sp., and Lachnospira…

The type VI secretion system governs strain maintenance in a wild mammalian gut microbiome.

Bacteria inhabiting the mammalian gut coexist in dense communities where contact-dependent antagonism mechanisms are widespread. The type VI secretion system (T6SS) is an interbacterial toxin delivery pathway prevalent among gut Bacteroidales, yet its function in naturally evolved microbiomes remains poorly defined. Here, we examine the role of the T6SS in Bacteroides within a physiologically relevant gut community derived from wild mice (the WildR microbiome). Using newly developed genetic tools and a strategy for functional replacement of strains within the WildR community, we demonstrate th…

Molecular basis of SAM-AMP synthesis and degradation in the type III-B CRISPR-Cas system.

Upon sensing nonself target RNA, the CorA-associated type III-B CRISPR-Cas system catalyzes S-adenosyl methionine (SAM) and ATP to synthesize SAM-AMP, which activates the effector CorA and triggers immune responses. SAM-AMP can be degraded by NrN and SAM lyase, potentially deactivating the system. Here we find that the type III-B effector complex from Bacteroides fragilis uses a specific mechanism to recognize nonself target RNA and synthesize SAM-AMP. The 3' anti-tag of nonself target RNA induces conformational changes in the Cmr2 subunit, triggering SAM-AMP synthesis independently of the sta…
