Biocatalytic synthesis of S-adenosyl-l-methionine analogues for allosteric activation of cystathionine-β-synthase.

Cystathionine-β-synthase (CBS) is the key regulatory enzyme of the transsulfuration pathway and plays a crucial role in eliminating homocyst … eric activator of CBS as a template, we rationally designed 15 analogues and synthesized them using biocatalysis. We characterized their action on isolated recombinant human CBS and cell lysate and their metabolic breakdown in DNA methylation. Besides SAM, three analogues enhanced the catalytic activity of CBS and stabilized CBS during thermal denaturation, su …

Stereoconvergent Photo-Biocatalytic Cascade to Optically Enriched Amines from Racemic Carboxylic Acids.

Herein, we report a synergistic synthetic strategy to integrate photoredox catalysis and biocatalysis in a one-pot/two-step linear cascade synthesis of enantiomerically pure amines starting from racemic carboxylic acids utilizing visible light and transaminases. This transformation was achieved via quantitative decarboxylative oxidation (>99% conv.) of racemic aryl-alkyl carboxylic acids followed by stereoselective reductive amination of the corresponding dehomologated carbonyl intermediates. The initial photocatalytic step employs blue LED irradiation (427 nm) and molecular oxygen (O2) as the…

Deciphering the Unconventional Reduction of C=N Bonds by Old Yellow Enzymes Using QM/MM.

The reduction of C=X (X = N, O) bonds is a cornerstone in both synthetic organic chemistry and biocatalysis. Conventional reduction mechanisms usually involve a hydride ion targeting the less electronegative carbon atom. In a departure from this paradigm, our investigation into Old Yellow Enzymes (OYEs) reveals a mechanism involving transfer of hydride to the formally more electronegative nitrogen atom within a C=N bond. Beyond their known ability to reduce electronically activated C=C double bonds, e.g., in α, β-unsaturated ketones, these enzymes have recently been shown to reduce α-oximo-β-k…

Catalytic and structural insights into a stereospecific and thermostable Class II aldolase HpaI from Acinetobacter baumannii.

Aldolases catalyze the reversible reactions of aldol condensation and cleavage and have strong potential for the synthesis of chiral compounds, widely used in pharmaceuticals. Here, we investigated a new Class II metal aldolase from the p-hydroxyphenylacetate degradation pathway in Acinetobacter baumannii, 4-hydroxy-2-keto-heptane-1,7-dioate aldolase (AbHpaI), which has various properties suitable for biocatalysis, including stereoselectivity/stereospecificity, broad aldehyde utilization, thermostability, and solvent tolerance. Notably, the use of Zn2+ by AbHpaI as a native cofactor is distinc…

Local Flexible-Region Engineering Improves the Thermostability and Catalytic Activity of 2-Deoxy-D-Ribose-5-Phosphate Aldolase.

2-Deoxy-D-ribose-5-phosphate aldolase (DERA) is a useful biocatalyst, but its application is limited by poor thermostability. Here, a flexibility-guided engineering strategy was applied to Escherichia coli DERA (EcDERA) to improve thermostability while retaining catalytic performance. Molecular dynamics simulations were used to identify flexible regions for mutation design. Among the variants tested, D26P/Y233F showed the best overall performance. Its half-life at 60 °C increased from 103 to 363 min (3.5-fold), and its specific activity increased by 60.9% (17.28 vs 10.74 U·mg-1) compared with

Identification of a Novel Glycosyltransferase CaUGT Specifically Targeting the C-30 Position of Glycyrrhetinic Acid and Development of an Efficient Enzymatic Biotransformation System.

Glycyrrhetinic acid (GA) is a major bioactive ingredient of licorice with extensive pharmacological functions, while its strong hydrophobicity and low permeability severely restrict practical application. Enzymatic glycosylation serves as a mild method to enhance GA solubility and bioactivity, yet few reported glycosyltransferases can specifically modify the C30 carboxyl group of GA. In this study, a novel glycosyltransferase gene CaUGT was identified and cloned from Cardamine amara subsp. amara. The recombinant GST-CaUGT achieved optimal expression under 2% dextrin, 0.2 mM IPTG and 25 °C indu

Facile immobilization of an unstable recombinant α-L-rhamnosidase on magnetite nanoparticles for efficient naringin biotransformation.

α-L-Rhamnosidase is a significant biocatalyst that specifically cleaves terminal α-L-rhamnose group from natural flavonoid diglycosides, enabling the biocatalytic production of high-value flavonoid glucosides like prunin from naringin. Compared to its precursor naringin, prunin exhibits superior solubility and bioavailability, making this enzymatic conversion commercially valuable for the food and pharmaceutical industry. However, the industrial application of free α-L-rhamnosidases faces significant challenges, including enzyme instability, difficulty in recovery, and unsatisfactory reusabili

Multidimensional Computational Engineering of Pectate Lyase for Enhanced Thermostability and Moderately Improved Catalytic Efficiency.

Pectate lyases are promising industrial biocatalysts, but limited thermostability restricts their practical use. Here, we developed a multidimensional consensus computational framework integrating sequence conservation, structural dynamics, and thermodynamic prediction to identify functional mutation hotspots in PcPel1834. Iterative saturation mutagenesis yielded M5 (S212E/Q225L/F241L/L260A/G296T), which showed markedly enhanced thermostability and moderately improved apparent catalytic efficiency. M5 increased the melting temperature from 52.1 to 65.0 °C and retained nearly full activity afte

Rational Design of a Dual Mutant Degrading Enzyme with Improved Catalytic Efficiency and Acid Tolerance for Four Aflatoxins.

Aflatoxins are major contaminants in raw food materials and their products. Developing enzymes that efficiently degrade multiple aflatoxins under acidic conditions has great practical value. In this study, the aflatoxin-degrading enzyme ADPPIII from Aspergillus terreus was systematically engineered. Substrate tunnel optimization obtained mutant F375L, which showed about twice the degradation activity for four aflatoxins versus the wild-type. Surface charge neutralization generated dual mutant F375L-K570Q with significantly improved acid tolerance. At pH 5.0, its degradation rates of aflatoxins

Can ferric-oxyl excited states explain elongated iron-oxygen bonds in heme peroxidase catalytic intermediates?

The use of X-ray structures to determine and interpret the ferryl iron-oxygen bond order in molecular oxygen-activating heme enzymes has, in the past, been controversial. This has mainly stemmed from the susceptibility of ferryl species to X-ray-induced electronic state changes. In this work we establishe using time-resolved serial femtosecond X-ray crystallography (tr-SFX) on a dye-decolourising peroxidase that the ferryl intermediate species (Compounds I and II) captured following in situ mixing of microcrystals with H2O2 have single, rather than the double bond character expected. X-ray emi

Insilico engineering of transaminase variants for enhanced biocatalytic conversion of an ACE inhibitor precursor.

Angiotensin-converting enzyme (ACE) inhibitors are widely prescribed for cardiovascular disorders, yet their conventional chemical synthesis involves multiple steps, high energy consumption, and poor stereoselectivity. In this work, we present a fully insilico framework for the computational design and prediction of transaminase (TA) variants capable of catalyzing the asymmetric amination of ethyl 2-oxo-4-phenylbutanoate (OPBE) to yield L-homophenylalanine ethyl ester (L-HPE), a key chiral intermediate in ACE inhibitor synthesis. Co-catalysis of modified metal-organic framework immobilized enzyme with nonionic surfactant for biodiesel preparation.

With the increasing demand for sustainable energy, enzyme-catalyzed biodiesel preparation has garnered significant interest owing to its green and efficient characteristics. However, conventional immobilized lipase (LIP) is susceptible to desorption and inactivation and fails to balance the catalytic efficiency and stability, limitations that seriously restrict the industrial application. To address this, we constructed a carrier through hydrophobically modifying University of Oslo-66-NH2 (UiO-66-NH2), an amino-functionalized metal-organic framework (MOF), with octadecylphosphonic acid (OPA).

Substrate Epoxidation Catalyzed by the Nonheme Iron Dioxygenase Dapdiamide Biosynthesis Enzyme C. Why Is the Substrate Tethered?

Enzymes usually react with a free substrate, although some examples have appeared in the literature of enzymes that utilize a substrate tethered to a protein carrier. However, it is not clear what advantage the tethering has, and therefore a computational study was performed. In particular, we report here the first computational study on the nonheme iron dioxygenase involved in the epoxidation reaction during the dapdiamide biosynthesis (DdaC) and investigate tethered and nontethered substrates. Molecular dynamics (MD) simulations show that the protein carrier applies pressure onto the surface

Humic acid aggregates with laccase and decreases the performance of the enzyme catalytic systems through various mechanisms.

Laccases are among the best-rated enzymes for industrial and environmental applications, yet their use in bioremediation is limited by interference from environmental components like humic acid (HA). This study evaluated HA impact on the oxidation of 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulphonate (ABTS) and two model pollutants - anthracene and methyl orange - by laccase(-mediator) systems. HA consistently diminished conversion rates, with EC50 values between 5 and 51 mg/L suggesting diverse inhibitory mechanisms. We investigated potential mechanisms including substrate sequestration, rad

Switching engineered Vitreoscilla hemoglobin into carbene transferase for enantioselective SH insertion.

An attractive strategy for efficiently forming CS bonds is through the use of diazo compounds SH insertion. However, achieving good enantioselective control in this reaction within a biocatalytic system has proven to be challenging. This study aimed to enhance the activity and enantioselectivity of to enable asymmetric SH insertion. The researchers conducted site-saturation mutagenesis (SSM) on 5 amino acid residues located around the iron carbenoid intermediate within a distance of 5 Å, followed by iterative saturation mutagenesis (ISM) of beneficial mutants. Through this process, the benefic

Characterization of the flavin monooxygenase involved in biosynthesis of the antimalarial FR-900098.

The latter steps in this biosynthetic pathway for the antimalarial phosphonic acid FR-900098 include the installation of a hydroxamate onto 3-aminopropylphosphonate, which is catalyzed by the consecutive actions of an acetyltransferase and an amine hydroxylase. Here, we present the 1.6 Å resolution co-crystal structure and accompanying biochemical characterization of FrbG, which catalyzes the hydroxylation of aminopropylphosphonate. We show that FrbG is a flavin-dependent N-hydroxylating monooxygenase (NMO), which shares a similar overall structure with flavin-containing monooxygenases (FMOs).
