Disposition kinetics of ketamine in the domestic cat.

The disposition kinetics and systemic availability of ketamine, a dissociative anaesthetic agent, was studied in normal domestic cats. A similar dose (25 mg/kg) of ketamine hydrochloride was administered by the i.v. and i.m. routes; drug concentrations in plasma were measured by a gas-liquid chromatographic procedure. A rapid distribution phase (t1/2 (alpha) = 3 min) was followed by a slower first-order elimination phase. The half-life of the drug (66.9 +/- 24.1 min) was independent of the route of parenteral administration. Absorption from i.m. site of administration was rapid, with peak plas

Hyaluronic acid in functional foods and oral supplements: linking molecular weight, bioavailability, and food matrix effects to clinical translation.

Hyaluronic acid (HA) has attracted growing interest as a multifunctional ingredient in functional foods and oral supplements because of its techno-functional properties and proposed benefits for skin and joint health. However, the physiological relevance of orally administered HA remains difficult to interpret because its gastrointestinal fate, exposure, and clinical outcomes may depend on molecular weight distribution, dose, formulation, processing history, food-matrix interactions, and host-related factors. This review integrates evidence on HA molecular characteristics, gastrointestinal tra

Oleogel-based emulsions as proanthocyanidin delivery systems: Impact of fatty acid saturation and oil/water ratio on molecular packing, rheology, and bioaccessibility.

This study investigated oil-type-dependent variations in molecular packing, crystallization behavior, interfacial properties, rheology, and oxidative stability of oleogel-based emulsions (OGEs), with a focus on correlated gradients of fatty acid saturation, and developed a delivery system to encapsulate the hydrophilic bioactive substance (proanthocyanidins, PAs). Results showed that the fatty acid saturation was associated with molecular stacking and crystal arrangement, further modulating gel strength and thermal stability. Among the tested oils, medium-chain triglyceride (MCT) based OGEs ex

An overview of international reference values for vitamin E intake - similarities and differences in derivation.

PURPOSE: The scientific knowledge for the essential fat-soluble vitamin E (VitE) remains inconclusive. There is still a lack of valid status parameters and deficiency symptoms for insufficient intake. Therefore, the derivation of a dietary reference value (DRV) for VitE is challenging and differs between the different organizations and countries. The aim of the current paper is to provide an overview of selected international DRVs and discuss the different derivation procedures. METHODS: DRVs from sixteen countries and international organizations were retrieved, compared and evaluated. RESUL

Evaluation of absorption kinetics and bioavailability of palmitic acid and 2-palmitoylglycerol, a metabolite of interesterified fat, radiolabeled with 99m-technetium.

Interesterified fat (IF) has replaced partially hydrogenated fats because it does not generate trans isomers, but it may affect lipid metabolism due to structural changes in triacylglycerols (TAGs). Since chemical interesterification alters TAG configuration and influences digestion and absorption, methodological approaches capable of tracking lipid metabolites are needed. In this study, we investigated the absorption kinetics and systemic distribution of IF metabolites using a radiolabeling strategy. Eight-week-old Swiss mice received via gavage: PA group-3.7 MBq of 99mTc-palmitic acid (99mTc

Antibacterial potential of rutin nanoformulations: Current insights and future directions.

Rutin, a plant-derived flavonoid, exhibits broad pharmacological activities, including antibacterial effects against Gram-positive and Gram-negative bacteria. However, its clinical and industrial applications are restricted by poor aqueous solubility, low bioavailability, and instability under physiological conditions. To address these limitations, several nanoformulation strategies have been developed, including polymeric nanoparticles, lipid-based carriers, nanoemulsions, and micro- and nano-encapsulation techniques. These approaches have been shown to improve rutin's solubility, dispersibil

A chitosan-based cinnamaldehyde-raspberry ketone integrated carrier architecture for oral delivery toward mild metabolic regulation.

Obesity-related metabolic imbalance represents a growing health concern, and increasing attention has been directed toward mild metabolic regulatory interventions based on plant-derived phytochemicals before progression to more severe metabolic disorders. Raspberry ketone (RK), a naturally occurring phytochemical with reported metabolic regulatory activity, exhibits limited gastrointestinal accessibility due to its high crystallinity and poor aqueous solubility. In this study, a chitosan-based cinnamaldehyde-raspberry ketone integrated carrier system (CS-CA/RK) was developed as a surfactant-fr

OralAbsPredict: A data-driven framework to predict human intestinal absorption (HIA) and human oral bioavailability (HOB) from chemical structures.

Oral absorption is a key pharmacokinetic process for orally administered drugs, determining therapeutic efficacy and influencing distribution, metabolism, and excretion. One of the major reasons a drug fails in early development is poor oral absorption. Therefore, it is important to determine the oral absorption properties of drugs during early development, even before synthesis. Human intestinal absorption (HIA) and human oral bioavailability (HOB) are two important pharmacokinetic properties of orally administered drugs that provide a good measure of drug absorption through the oral route. Age-dependent bioaccessibility of citrinin, ochratoxin A, and zearalenone in boiled white Rice: A comparative INFOGEST in vitro study.

Mycotoxins frequently co-occur in staple foods such as rice, yet little is known about how age-related gastrointestinal changes influence their release during digestion. This study evaluated the bioaccessibility of citrinin (CIT), ochratoxin A (OTA), and zearalenone (ZEN) in cooked white rice using the standard INFOGEST protocols simulating adults and older adults digestion under single and mixed exposure scenarios. Gastric and intestinal bioaccessible fractions were quantified using a validated LC-MS/MS method. Significant differences in bioaccessibility were evaluated across age groups and e

Research progress and prospects of nutrient delivery systems in food design for the elderly.

The global aging population has led to age-related digestive dysfunction, which has become a key driver of malnutrition and declining health. Physiological degeneration in older adults severely reduces nutrient bioavailability, making conventional single nutrient supplementation ineffective. Nutrient delivery systems tailored to the physiological constraints of the elderly show great value in alleviating geriatric malnutrition. Micro/nanoencapsulation, emulsions, and gels are particularly promising platforms for elderly-targeted food innovation due to their tunable structure, controlled target

Designing liposomal oral formulations aligned with physiology, payload properties, and scalable manufacturing.

Liposomes have long been established as versatile and biocompatible carriers for biologically active molecules. Advances in manufacturing technology have dramatically broadened their application landscape, positioning them today as effective platforms for the oral delivery of pharmacologically active compounds, nutrients, and dietary supplements. Developing effective oral liposomal formulations, however, demands more than empirical optimization. It requires a strategy that simultaneously accounts for the complex physiological environment of the gastrointestinal (GI) tract, the physicochemical

Mercury in Antarctic vegetation as an indicator of uptake routes and bioavailable forms.

Mercury (Hg) is a global contaminant of concern due to its persistence, toxicity, and capacity for long-range transport. This study investigated the Hg concentration, chemical speciation, and δ13C/δ15N stable isotope signatures in Antarctic coastal flora, comprising lichens, mosses, vascular plants, and algal mats (Prasiola crispa) to evaluate the relative importance of atmospheric, edaphic, and biogenic inputs in Hg accumulation. Samples were collected from King George Island during the austral summer of 2018/19. Lichens showed the highest Hg concentration (median THg = 116.6 ng g⁻¹) and an e

Enhancing Bioavailability of Ultra-High Concentration Antibody Formulations Using Ionic Liquids.

Subcutaneous (SC) delivery of ultra-high concentration antibody formulations (uHCAFs, > 150 mg/mL) is limited by excessive viscosity and poor bioavailability. Here, we present an ionic liquid made with choline and tryptophan (CHAT) as a multifunctional excipient that enables SC injection of antibody formulations at a concentration > 200 mg/mL by simultaneously reducing viscosity, enhancing tissue permeation, and stabilizing antibodies. Notably, CHAT-formulated IgG exhibited viscosities below the injectability threshold (∼20 cP). In murine studies, SC delivery of CHAT-formulated IgG achieved si

Dissolved Organic Matter Chemistry Modulates Biochar Effects on Phenanthrene Biodegradation in Agricultural Soil: Bioavailability and Microbial Responses.

Biochar is increasingly applied to cropland, yet co-occurring dissolved organic matter (DOM) jointly regulates polycyclic aromatic hydrocarbons (PAHs) fate and food-safety risks. We amended phenanthrene-spiked agricultural soil with wheat-straw biochar and three DOM types (citric acid, oxalic acid and humic acid) and linked biodegradation to bioavailable pools and microbial responses. Citric and oxalic acids increased bioavailable fractions and raised microbial removal from 43.98 ± 0.72% (control) to 55.84 ± 0.87% and 54.74 ± 2.29%, whereas humic acid lowered effective bioavailability and redu

Optimization of Curcumin-Ascorbic Acid Cocrystal for Enhanced Solubility, Tablettability, and Oral Bioavailability: Structural, Functional, and Pharmacokinetic Insights.

Curcumin (CUR) exhibits broad therapeutic potential across multiple disease conditions. However, it's extremely low aqueous solubility, poor dissolution, and limited oral bioavailability constrain its clinical utility. Therefore, the present study aimed to address the aforesaid limitations through design, optimization, and evaluation of a pharmaceutical cocrystal of CUR with ascorbic acid (CUR-AA-C). Initially, in silico modelling was employed to predict drug-coformer interactions, followed by optimization of the CUR-AA stoichiometry using a solvent-evaporation technique. The resulting solid f

Phosphatidylethanolamine, oleic acid and gondoic acid enhance lutein bioavailability: A hybrid static-dynamic-in vivo study.

Lutein has low stability and limited bioaccessibility in infant formulas. By integrating in vitro (static and dynamic) and in vivo digestion models, oleic acid (OA), gondoic acid (GA) and phosphatidylethanolamine (PE) were identified as agents that raise lutein bioaccessibility by 6-13% under static in vitro conditions. During digestion the additives reduced emulsion droplet size (0.3% PE most effective), inhibited lipid-droplet aggregation and enhanced micellar stability. PE additionally accelerated intestinal free fatty acid release. Dynamic digestion gave higher absolute values but identica
