Digital Twin Control Framework Uncovers Individualized Cross-Scale Links Between Functional Connectivity Deviations, Communication Impairment, and Molecular Mechanisms in Autism.

BACKGROUND: Autism spectrum disorder (ASD) is hallmarked by pronounced inter-individual heterogeneity in brain function and clinical manifestations. Conventional neuroimaging studies yield highly inconsistent findings, as extrinsic confounding factors and intrinsic disease heterogeneity mask ASD-specific functional connectivity (FC) signatures, hampering the identification of individualized neural mechanisms linked to symptom severity. METHODS: We developed a digital twin control analytical framework based on deep contrastive learning. By comparing each ASD participant with their matched digi

Predicting susceptibility to inflammaging using a genetic inflammatory index through Prakriti-based phenotypic stratification.

Inflammaging is chronic low-grade inflammation arising from a progressive imbalance between pro-inflammatory and anti-inflammatory immune networks. The rate of inflammaging exhibits marked interindividual variation owing to differences in the pace of biological aging. Within the two-hit theory of inflammaging, such variability involves both genetic and epigenetic components. Stratification models derived from quantification of the genetic component allow classification of individuals according to their baseline genetic inflammatory tone. Such phenotypic stratification could facilitate informed

Early renal and vascular damage within the normoalbuminuria condition.

OBJECTIVE: A continuous association between albuminuria and cardiorenal risk exists further below moderately increased albuminuria ranges. If only based in albumin to creatinine ratio (ACR) higher than 30 mg/g, a significant percentage of individuals may be out of the scope for therapeutic management. Despite epidemiological outcomes, the identification of biochemical changes linked to early albuminuria is underexplored, and normoalbuminuric individuals are usually considered at no risk in clinical practice. Here, we aimed to identify early molecular alterations behind albuminuria development.

Individual differences in stress coping are linked to working memory performance in male and female F344 rats.

Executive dysfunction and altered stress reactivity are core features of many neuropsychiatric disorders, contributing substantially to global disability and frequently exhibiting sex-specific prevalence and symptom profiles. Understanding how stress responses normally relate to executive function, and whether these relationships differ by sex, is critical for identifying mechanisms of vulnerability and resilience. Although sex differences in stress and cognition have been reported, preclinical findings remain mixed, and few studies assess behavioral coping, endocrine responses, and working me

A clinical algorithm to determine target blood pressure in the elderly: evidence and limitations from a clinical perspective.

As the elderly population is growing rapidly, management of hypertension in South Korea faces major challenges because the proportion of elderly hypertension patients is also increasing. The characteristics of this population are also much more complex than younger patients. Elderly hypertension is characterized by wide variations in (1) fitness or biological age, (2) white-coat effect, (3) poor functional status or frailty, (4) dependency in activities of daily living or institutionalization, (5) orthostatic hypotension, and (6) multiple comorbidities. All of these should be considered when c

The power of genetic diversity in genome-wide association studies of lipids.

Increased blood lipid levels are heritable risk factors of cardiovascular disease with varied prevalence worldwide owing to different dietary patterns and medication use1. Despite advances in prevention and treatment, in particular through reducing low-density lipoprotein cholesterol levels2, heart disease remains the leading cause of death worldwide3. Genome-wideassociation studies (GWAS) of blood lipid levels have led to important biological and clinical insights, as well as new drug targets, for cardiovascular disease. However, most previous GWAS4-23 have been conducted in European ancestry

Diabetes Genetic Clusters and Clinical Outcomes in American Indians.

UNLABELLED: Diabetes has a large medical and public health impact in American Indians. Studies have used genetic data to distinguish type 1 diabetes (T1D) and type 2 diabetes (T2D) and uncover biologic mechanisms underlying T2D clinical heterogeneity. We applied a T1D polygenic score (PS) to 3,084 American Indians (mean age 56 years, 58% women, 39% diabetes). We also calculated partitioned PS for eight clusters of T2D-associated variants and evaluated their association with 20 cardiometabolic traits and five clinical outcomes. The profile of T1D PS for individuals with diabetes was consistent

Comorbidities in Ehlers-Danlos syndromes and hypermobile spectrum disorders.

INTRODUCTION: hypermobile spectrum disorders (HSD) and Ehlers-Danlos syndromes (EDS) are connective tissue disorders often associated with systemic manifestations such as mast cell activation syndrome (MCAS), postural orthostatic tachycardia syndrome (POTS), irritable bowel syndrome (IBS), and autoimmune conditions, including spondyloarthritis (SpA). The overlap of these conditions complicates diagnosis and treatment. This study investigates the prevalence of MCAS and other comorbidities, as well as patterns of medication use, in individuals with HSD/EDS. MATERIAL AND METHODS: this cross-sect

The impact of interferon-γ pathway on trained immunity induction by vaccination with Bacille Calmette-Guérin.

Bacillus Calmette-Guérin (BCG) has multiple heterologous off-target effects which extend beyond tuberculosis (TB) prophylaxis, which include protection against other non-tuberculous infections, autoimmune diseases, and tumor development. These heterologous effects are at least partially mediated by induction of trained immunity. In this study, we aimed to investigate the impact of IFNγ production capacity on induction of trained immunity in human volunteers vaccinated with BCG. We evaluated inflammation and immune activation-specific cytokine responses (IFNγ, TNF, IL-1, and IL-6) in PBMCs isol

Assessment of the normal pediatric pineal gland - MR findings of a single centre; A retrospective study.

Retinopathy of prematurity (ROP) is a leading cause of childhood blindness that arises from disrupted retinal vascular development in premature infants. Oxygen exposure remains a central driver of treatment-warranted ROP, as higher saturation levels suppress early retinal vascular growth and later promote pathological neovascularization. Large, randomized trials of oxygen targeting show that lower oxygen saturation ranges reduce the incidence of treatment-requiring ROP, though with trade-offs in mortality. Observational cohorts, including the CRYO-ROP, ETROP, and e-ROP trials, consistently rep

Agent-based and continuous models of hopper bands for the Australian plague locust: How resource consumption mediates pulse formation and geometry.

Locusts are significant agricultural pests. Under favorable environmental conditions flightless juveniles may aggregate into coherent, aligned swarms referred to as hopper bands. These bands are often observed as a propagating wave having a dense front with rapidly decreasing density in the wake. A tantalizing and common observation is that these fronts slow and steepen in the presence of green vegetation. This suggests the collective motion of the band is mediated by resource consumption. Our goal is to model and quantify this effect. We focus on the Australian plague locust, for which excell

X-band EPR imaging as a tool for gradient dose reconstruction in irradiated bones.

PURPOSE: Various tools are currently available for dose reconstruction in individuals after accidental exposure to ionizing radiation. Among the available biological analyses, Monte Carlo simulations, and biophysical methods, such as electron paramagnetic resonance (EPR), the latter has proved its usefulness for retrospective dosimetry. Although EPR spectroscopy is probably the most sensitive technique, it does not provide spatial dosimetric data. This information is, however, highly desirable when steep dose gradient irradiations are involved. The purpose of this work was to explore the possi

A rapid direct fluorescent assay for cell-free DNA quantification in biological fluids.

BACKGROUND: Circulating cell-free DNA (CFD) levels may be elevated in trauma, stroke, sepsis, pre-eclampsia and cancer. Owing to the complex and expensive methodology, detection of CFD has hitherto been confined to research laboratories. This study presents a simple, inexpensive and accurate test for CFD. METHODS: Using the commercial fluorescent SYBR Gold stain, biological fluids were directly assayed for CFD without prior DNA extraction and amplification. Stain was added to the sample in 96-well plates (final stain dilution: 1:10,000) and fluorescence was read by a fluorometer (excitation w

Beyond a universal obesity microbiome signature: pre-intervention heterogeneity and a framework for baseline profiling.

Unlike previous reviews that primarily summarize obesity-associated microbial alterations, this review reframes the recurrent inconsistency of human obesity microbiome findings as an informative consequence of pre-intervention heterogeneity rather than merely failed replication or analytical noise. We integrate biological and contextual determinants of baseline microbiota variation with methodological and analytical sources of heterogeneity into a unified framework for interpreting why microbial diversity, taxonomic composition, and functional signals differ across individuals, populations, an

Cellular and Molecular Mechanisms of Hemorrhagic Shock: Biological Rationale for Individualized Fluid Resuscitation Strategies and Multimodal Monitoring.

Hemorrhagic shock is a leading cause of preventable death following multiple trauma, driven by a cascade of interacting cellular and molecular disturbances that extend well beyond simple volume loss. Acute blood loss initiates tissue hypoperfusion and cellular hypoxia, setting in motion the lethal triad of hypothermia, acidosis, and coagulopathy through several converging pathways: complement activation with excessive C3a and C5a production; neutrophil-mediated tissue injury; NADPH-oxidase-driven reactive oxygen species (ROS) overproduction that overwhelms superoxide dismutase defenses; mitoch
