A 33-year-old man developed seropositive rheumatoid arthritis (RA) followed soon after by a monoclonal lymphoproliferative disorder characterized by CD4 positive T lymphocytes in his peripheral blood, bone marrow and lymph nodes. Previous reports of polyarthritis in association with a T cell lymphoproliferative disorder have all involved the CD8 phenotype.

BACKGROUND: The immunological background of primary biliary cirrhosis (PBC) remains largely obscure.
METHODS: Using double colour flow cytometry, we estimated the distribution of functionally different lymphocyte subpopulations in the peripheral blood of 25 PBC patients and 18 controls. We examined: 1) the expression of CD3, CD4, CD8, CD19 and CD56 surface receptors, 2) the distribution of lymphocyte subsets bearing 'naive' (CD45RA+) and 'memory' (CD45RO+) phenotypes in both CD4+ and CD8+ cell populations, 3) the expression of an early activation marker (CD69), 4) the distribution of C1.7 mAb

Infections elicit immune adaptations to enable pathogen resistance and/or tolerance and are associated with compositional shifts of the intestinal microbiome. However, a comprehensive understanding of how infections with pathogens that exhibit distinct capability to spread and/or persist differentially change the microbiome, the underlying mechanisms, and the relative contribution of individual commensal species to immune cell adaptations is still lacking. Here, we discovered that mouse infection with a fast-spreading and persistent (but not a slow-spreading acute) isolate of lymphocytic chori

OBJECTIVES: Rheumatoid arthritis (RA) is attributed to inflammation and infiltration of immune cells into the inflamed joints. In the synovial fluid (SF), accumulated neutrophils can transdifferentiate into neutrophil-dendritic cell hybrids (N-DCs) and acquire antigen presenting properties. This study aimed to establish the crosstalk, if any, between N-DCs and T lymphocytes in the SF as also assess their potential role in disease sustenance.
METHODS: In the SF of patients with RA (n=15), the frequency and activation status of CD4+ and CD8+ T lymphocyte subsets (i.e. memory, naive, central mem

A major challenge in advancing neoantigen-targeted therapies lies in the limited capacity of predicted neoantigens to robustly activate CD8+ T cells, emphasizing the critical need for comprehensive functional validation before patient vaccination. Here, we provide a direct comparison of neoantigen-specific T cell responses between vaccinated patients with cancer and HLA-matched healthy donors. Despite vaccination, patient-derived T cells recognized only a small fraction of predicted neoantigens, whereas healthy donor T cells consistently exhibited broader and more robust neoantigen reactivity.

Combination therapies are becoming a focal point in cancer immunotherapy. In this issue of Cancer Cell, Johnston and colleagues identify the TIGIT/CD226 pathway, which provides significant interest for combination with PD-1 pathway blockade to improve anticancer CD8(+) T cell responses, because it acts by a novel mechanism to regulate CD8(+) T cell functions within the tumor microenvironment.

A variety of adhesion molecules regulate the traffic and tissue localization of lymphocytes in vivo by mediating their binding to vascular endothelial cells. The homing receptor gp90MEL-14 (gp90), also known as LECAM-1 or L-selectin, mediates the adhesion of lymphocytes to specialized high endothelial venules in lymph nodes (LN) and is the primary molecule regulating lymphocyte recirculation and homing to LN, whereas other adhesion molecules have a major role in the localization of lymphocytes in inflammatory sites. We used four-color flow cytometric analysis to examine the regulation of adhes

This study characterizes changes in peripheral blood lymphocyte subsets among pediatric hemophagocytic lymphohistiocytosis (HLH) patients. Compared to healthy controls, HLH patients exhibited significantly elevated levels of CD8⁺ T cells, with concomitant reductions in B cells, CD4⁺ T cells, DPT cells, DNT cells, and the CD4⁺/CD8⁺ T cell ratio as determined by multiparametric flow cytometry. Organ dysfunction was further correlated with distinct immunophenotypic alterations. Specifically, respiratory dysfunction was associated with decreased CD4⁺ CM T cells, naïve B cells, plasmablasts, and tr

INTRODUCTION: CD4/CD8 ratio is a marker of immune activation in HIV infection and has been associated with neurocognitive performance during chronic infection, but little is known about the early phases. The aim of this study was to examine the relationship between blood CD4/CD8 ratio and central nervous system endpoints in primary HIV infection (PHI) before and after antiretroviral treatment (ART).
METHODS: This was a retrospective analysis of the Primary Infection Stage CNS Events Study (PISCES) cohort. We longitudinally assessed blood and cerebrospinal fluid (CSF) markers of inflammation,

The proprotein convertase subtilisin/kexins (PCSKs) regulate biological actions by cleaving immature substrate proteins. The archetype PCSK, FURIN, promotes the pathogenicity of viruses by proteolytically processing viral proteins. FURIN has also important regulatory functions in both innate and adaptive immune responses but its role in the CD8+ CTLs remains enigmatic. We used a T-cell-specific FURIN deletion in vivo to demonstrate that FURIN promotes host response against the CTL-dependent lymphocytic choriomeningitis virus by virtue of restricting viral burden and augmenting interferon gamma

Over 14 weeks, peripheral blood lymphocytes (PBL) were isolated from eight adult cattle which had been orally infected with Fasciola hepatica via trickle infection over a 10-day period. Two age, breed and sex-matched cattle served as controls. CD4+, CD8+ and gammadelta+ T cells were depleted from whole PBL populations by magnetic bead depletion. Lymphocyte proliferation assays demonstrated a transient, but marked elevation in responsiveness to Concanavalin A (Con A) between weeks 2 and 4 post-infection in PBL from infected animals. Proliferative responses to Con A were significantly greater in

Aging is associated with impaired T cell immune responses, raising the susceptibility of the elderly to infections and cancers. Aged T cells exhibit impaired T cell responses to T cell receptor (TCR) stimulation accompanied by reduced glycolytic activity, but the molecular basis of these defects is largely elusive. Using data-independent acquisition (DIA) mass spectrometry-based proteomic analysis, we identified pyruvate dehydrogenase kinase 1 (PDHK1), a key glycolytic enzyme, as significantly downregulated in aged murine T cells. This loss of PDHK1 expression was confirmed in both CD4+ and CD

Immunization with vaccinia virus resulted in long-lasting protection against smallpox and was the approach used to eliminate natural smallpox infections worldwide. Due to the concern about the potential use of smallpox virus as a bioweapon, smallpox vaccination is currently being reintroduced. Severe complications from vaccination were associated with congenital or acquired T cell deficiencies, but not with congenital agammaglobulinemia, suggesting the importance of T cell immunity in recovery from infection. In this report, we identified two CD8+ T cell epitopes restricted by the most common

Leishmaniasis, caused by infection with the protozoan parasite Leishmania, affects millions of individuals worldwide, causing serious morbidity and mortality. This study directly determined the frequency of cells producing key immunoregulatory cytokines in response to the recombinant antigen Leishmania homolog of receptors for activated kinase C (LACK) and soluble leishmania antigen (SLA), and it determined relative contributions of these antigens to the overall cytokine profile in individuals infected for the first time with Leishmania braziliensis. All individuals presented with the cutaneou

BACKGROUND: A possible interaction between a specific HLA type and Adenovirus has been postulated as a promoter in leukemia clonal evolution. The HLA-DRB1*14, specifically DRB1*14:21, 14:22, 14:45, 14:26, 14:33, 14:51, 14:35 subtypes was the most frequent in CML Venezuelan patients.
OBJECTIVES: It is interesting to evaluate the molecular mimicry between the Adenovirus and the DRB1*14 subtypes which exhibit the same change in the amino acid position of the DR53 epitope. This mimicked segment has been identified as a LLERRRA polypeptide.

Although fetal growth restriction (FGR) is linked to dysregulated immune responses, the immunological profiles of maternal and umbilical cord blood remain poorly understood. The purpose of this study was to compare immune cell subsets in maternal blood (MB) and umbilical cord blood (UCB) between pregnancies that are normal and those that are complicated by FGR. Flow cytometry was used to analyze MB and UCB samples from 33 pregnant women (8 in the FGR group and 25 in the normal control group). In contrast to UCB, which was enriched in naïve T cells, MB from normal pregnancies contained greater

We developed a carboxyfluorescein succinimidyl ester (CFSE)-based flow-cytometric assay that can detect different subsets of vaccinia-specific T cells capable of both antigen-specific expansion and protective effector functions. Proliferation and effector functions were detected by CFSE dilution and intracellular staining, respectively. Absolute numbers of CD4(+)/CFSE(lo)/interferon (IFN)- gamma (+), CD8(+)/CFSE(lo)/IFN- gamma (+), CD8(+)/CFSE(lo)/granzyme A(+), and CD8(+)/CFSE(lo)/CD107a(+) T cells present after in vitro stimulation with live vaccinia were significantly higher in immunized in

Two adult cases of extranodal malignant lymphoma diffusely involving the intestinal wall are reported. Lymphoblastic lymphoma in case 1 (68-year-old male) and small lymphocytic lymphoma in case 2 (48-year-old male) were initially manifested as protein-losing enteropathy or malabsorption syndrome. In case 1, the patient died of massive ascites and intestinal bleeding 10 months after presentation. Autopsy revealed diffuse and extensive lymphomatous involvement of the small and large intestine, peritoneum and liver. The patient in case 2, showing low-grade diffuse lymphomatous invasion throughout
