Acute Treatment with the M-Channel (Kv7, KCNQ) Opener Retigabine Reduces the Long-Term Effects of Repetitive Blast Traumatic Brain Injuries.

We investigated whether pharmacological increase of "M-type" (KCNQ, Kv7) K + channel currents by the M-channel opener, retigabine (RTG), acutely after repetitive traumatic brain injuries (rTBIs), prevents or reduces their long-term detrimental effects. rTBIs were studied using a blast shock air wave mouse model. Animals were monitored by video and electroencephalogram (EEG) records for nine months after the last injury to assess the occurrence of post-traumatic seizures (PTS), post-traumatic epilepsy (PTE), sleep-wake cycle architecture alterations, and the power of the EEG signals. We evaluat

Effect of resolution enhancement using metal ion assisted strategy based on electrospray ionization-ion mobility spectrometry: A case study of carbendazim and thiabendazole in fruits.

A method for the rapid and simultaneous determination of carbendazim and thiabendazole residues by electrospray ionization-ion mobility spectrometry (ESI-IMS) combined with a metal ion-assisted technique was developed and validated in different fruit matrices. The metal ion assisted strategy was performed instead of tedious pre-separation procedures to overcome the limitation of low resolution of IMS. Four transition metal cations, Co(II), Ni(II), Cu(II), and Zn(II), were screened and their interactions with carbendazim and thiabendazole were investigated. The injection flow rate and metal ion

Comparison of Solriamfetol and Modafinil on Arousal and Anxiety-Related Behaviors in Narcoleptic Mice.

Wake-promoting agents are used for the management of excessive daytime sleepiness caused by narcolepsy. Clinical and preclinical data suggests that solriamfetol, a novel dopamine and norepinephrine reuptake inhibitor, is a promising therapeutic option for excessive daytime sleepiness. We provide the first head-to-head comparison of in vivo efficacy between modafinil and solriamfetol in narcoleptic mice. Both compounds induced potent wake-promoting effects in littermate wild-type and orexin-tTA; TetO-DTA mice when dosed at active and resting phases. However, neither modafinil nor solriamfetol a

Successful encorafenib desensitization in a patient with recurrent metastatic melanoma.

Type I hypersensitivity reactions (HSR) to dabrafenib are rare but have been previously described. We present a case where a 72-year-old woman with recurrent, metastatic BRAF-mutated melanoma developed a type I HSR to dabrafenib. We, therefore, developed a desensitization protocol with encorafenib, a similar class agent, to allow the patient to continue with treatment. Patients with a history of HSR to dabrafenib may be considered for encorafenib desensitization when other therapeutic options are limited.

Molecular and Biochemical Characterization of Carbendazim-Resistant Botryodiplodia theobromae Field Isolates.

Stem-end rot caused by Botryodiplodia theobromae is a destructive disease of mango. B. theobromae field isolates resistant to carbendazim (MBC) were collected in Hainan Province, China. In this study, the characteristics of these field isolates with resistance to MBC were investigated. The resistance of B. theobromae isolates to MBC was stably inherited. Both the MBC-resistant and MBC-sensitive isolates had similar mycelial growth rates, pathogenicity, sensitivity to high glucose, glycerol content, and peroxidase activity. Compared with MBC-sensitive isolates, MBC-resistant isolates were more

Evaluation of Inhibition Efficiency for the Detection of Captan, 2,3,7,8-Tetrachlorodibenzodioxin, Pentachlorophenol and Carbosulfan in Water: An Electrochemical Approach.

A novel bio-analytical method has been devised based on the change in catalytic activity of acetylcholinesterase (AChE) enzyme induced by captan, carbosulfan, 2,3,7,8-tetrachlorodibenzodioxin (TCDD) and pentachlorophenol (PCP) for the investigation of inhibition efficiency and sensitivity using Pt/ZnO/AChE/Chitosan bioelectrode. The inhibition curves of captan, carbosulfan, TCDD and PCP were similar to Michaelis-Menten curve. TCDD held the minimum inhibitor Michaelis-Menten constant ([Formula: see text]) value (10.2 nM) in comparison with PCP (10.9 nM), carbosulfan (14.5 nM) and captan (7.9 ×

Encapsulation of 3-iodo-2-propynyl N-butylcarbamate (IPBC) in polystyrene-polycaprolactone (PS/PCL) blends.

Polystyrene (PS, 1), polycaprolactone homopolymers (PCL, 2) and 3-Iodo-2-propynyl n-butylcarbamate (IPBC, 3) were physically mixed in dichloromethane (DCM) and processed into solid microspheres by using emulsion solvent evaporation method. Five different compositions with varying PS/PCL ratio were tested. The phase morphology of the microspheres was studied using Phase imaging atomic force microscopy (AFM) of polished cross-sections. Scanning electron microscopy was utilized to assess the distribution of IPBC in the polymer microspheres. The phase separation of the PS and PCL polymers in solve

Physiological modes of action of toxic chemicals in the nematode Acrobeloides nanus.

To gain a better understanding of the mechanisms through which a chemical exerts toxicity, a deeper insight is needed regarding the physiological processes that take place during a toxic stress. This issue can have important benefits for risk assessment, because it can contribute to a better interpretation of toxicity data. Here, we study the physiological mode of action of three different compounds (cadmium, carbendazim, and pentachlorobenzene) with an experimental data-based approach using whole life-cycle toxicity data from the nematode Acrobeloides nanus. We use a process-based model, base

Iodopropynyl butylcarbamate.

Iodopropynyl butylcarbamate (IPBC) is an internationally recognized chemical that has been used for years because of its wide field of application. Initially used as a water-based paint and wood preservative and then in metalworking fluids, its role has expanded into the more recent uses in cosmetic products. The need for a potent, broad-spectrum, and safe preservative system in cosmetics allowed for the discovery of several combinations of IPBC effective against a wide variety of organisms. Although IPBC has claimed to be safe when used at concentrations less than 0.1%, the introduction of IP

[Factors affecting the efficacy of 2 growth regulator compounds, OMS 3010 (phenoxycarb) and OMS 3019 (ethoxypyridine), on larvae of the Simulium damnosum complex].

Two insect growth regulators (IGR'S) acting as juvenile hormone analogs (JHA'S), OMS 3010 (phenoxycarb) and OMS 3019 (ethoxypyridine) were selected after a screening of several compounds proposed by industry on black fly larvae of the Simulium damnosum complex (Diptera: Simuliidae) the vector of onchocerciasis in West Africa. Bioassays were designed to study two parameters: larval age and exposure time. The results showed that these compounds are more efficient on old larvae (6th and 7th instars) than young larvae (3rd, 4th and 5th instars). However, exposure time seems essential. The black fl

Safety and efficacy of venglustat in GBA1-associated Parkinson's disease: an international, multicentre, double-blind, randomised, placebo-controlled, phase 2 trial.

BACKGROUND: Variants in the GBA1 gene, which encodes lysosomal acid glucocerebrosidase, are among the most common genetic risk factors for Parkinson's disease and are associated with faster disease progression. The mechanisms involved are unresolved but might include accumulation of glucosylceramide. Venglustat is a brain-penetrant glucosylceramide synthase inhibitor that, in previous studies, reduced amounts of the glycosphingolipid. We aimed to assess the safety, efficacy, and target engagement of venglustat in people with early-stage Parkinson's disease carrying pathogenic GBA1 variants. M

Carbendazim toxicity in different cell lines and mammalian tissues.

The extensive production and use of harmful pesticides in agriculture to improve crop yield has raised concerns about their potential threat to living components of the environment. Pesticides cause serious environmental and health problems both to humans and animals. Carbendazim (CBZ) is a broad spectrum fungicide that is used to control or effectively kill pathogenic microorganisms. CBZ is a significant contaminant found in food, soil and water. It exerts immediate and delayed harmful effects on humans, invertebrates, aquatic animals and soil microbes when used extensively and repeatedly. CB
