Environmental and human influence on the ecology of Trichinella spiralis and Trichinella britovi in Western Europe.

Surveys on Trichinella parasites in domestic and sylvatic animals collected in France, Italy, and in the Extremadura region of Spain showed that the distribution of Trichinella spiralis and Trichinella britovi is influenced by both environmental and human behaviour factors. In France, both Trichinella species are prevalent in the fox population from mountain areas and natural parks but are infrequent in wild boars (< 0.001%). In Italy, only T. britovi is present in sylvatic animals (foxes, wolves, and mustelids) living 500 m above sea level. This species is rare in wild boars (< 0.001%) in tha

The skin inflammatory response of the badger (Meles meles).

Twenty-five badgers, captured in an area where they had been implicated in outbreaks of bovine tuberculosis, received intradermal inoculations of control medium, 150 micrograms phytohaemagglutinin (PHA), 40 units streptokinase/10 units streptodornase (SK/SD), 200 micrograms purified protein derivative of Mycobacterium bovis (PPD), Freund's incomplete adjuvant (IFA), and Freund's complete adjuvant (CFA), each in 0.1 ml of inoculum. The reactions were assessed by skinfold thickness and skin histology 30 min--7 days after inoculation. Control medium caused slight cellular reaction, mostly polymor

Experimental babesiosis in coyotes and coydogs.

One splenectomized and 6 intact coyotes (Canis latrans), and 2 coydogs were experimentally inoculated with a recent isolate of Babesia gibsoni. The disease was mild in intact animals, was fatal in the splenectomized coyote, and was characterized by a regenerative hemolytic anemia with the PCV decreasing to 16% in intact animals and to 6% in the splenectomized coyote. Peak parasitemia ranged from 3% to 21% of erythrocytes infected and was inversely correlated to PCV. Serum lactate dehydrogenase, bilirubin, and globulin concentrations were increased in all infected animals. Three weeks after ino

Interspecies transmission of Cytauxzoon felis.

The causative agent of feline cytauxzoonosis was experimentally inoculated into 4 species of domestic farm animals, 9 species of laboratory animals, and 17 wildlife species. The inoculum consisted of freshly collected or deep-frozen blood and/or tissue homogenates from domestic cats euthanatized in extremis with experimentally transmitted feline cytauxzoonosis. A bobcat, Lynx rufus floridanus (Florida bobcat), developed cytauxzoonosis typical of the disease observed in domestic cats and died of the disease 2 weeks after inoculation. A persistent parasitemia, but no overt signs of illness, deve

Evaluation of the hemagglutination test for epidemiologic studies of leptospiral antibodies in wild mammals.

Sera from 153 wild animals of 18 species were tested for antibodies against 12 serovars of Leptospira by the microscopic agglutination (MA) test. Seventy-five percent of the animals tested were seropositive against one or more of the 12 serovars used. The most commonly found serovars were pomona, autumnalis, pyrogenes, icterohaemorrhagiae, australis, and canicola. Of 62 carnivores representing 7 species, 55 (89%) were seropositive, as were 46 (60%) of 77 rodents from 9 species. Leptospira of the serovar copenhageni serogroup icterohaemorrhagiae were recovered from kidney tissues of a Norway ra

First report on detection and molecular characterization of alphacoronaviruses in the small Indian mongoose (Urva auropunctata).

BACKGROUND: Members of the species Alphacoronavirus suis/Alphacoronavirus-1 (αCoV-1) are important viral pathogens of canids/felids that exhibit complex evolutionary patterns and have been associated with interspecies transmission events including zoonoses. Mongooses (family Herpestidae, order Carnivora) are scavengers that pose a risk as potential carrier of viral pathogens. To date, there are no reports on the genetic make-up/diversity of CoVs circulating in mongoose populations. METHODS: Fecal samples obtained from 53 small Indian mongooses (Urva auropunctata) on the Caribbean island of St

Morphological and molecular identification of cardiopulmonary nematodes in the European wildcat (Felis silvestris) and nine other carnivores in Spain.

Cardiopulmonary nematodes (CPN) are among the most pathogenic helminths, with wild carnivores serving as hosts for a variety of species. Knowledge of their presence in the Iberian Peninsula is limited, and for certain hosts, such as the European wildcat (Felis silvestris), no data are currently available. Lung and heart samples from 112 road-killed individuals belonging to 10 different species, primarily wildcats (n = 33), collected in Navarra (northern Spain) were collected. Morphological identification of nematodes was done and confirmed, when possible, by sequencing fragments of the cytochr

Species delimitation based on mtDNA genes suggests the occurrence of new species of Mesocestoides in the Mediterranean region.

BACKGROUND: This study is the first contribution to the molecular taxonomy of Mesocestoides spp. from domestic and wild carnivores in the Mediterranean area. A total of 13 adult worms and 13 larval stages of Mesocestoides spp. were collected from domestic and wild carnivore hosts in Italy and Tunisia. Samples collected in the Slovak Republic were used as comparative samples from outside the Mediterranean. The genes cytochrome c oxidase subunit 1 (cox1) and NADH dehydrogenase subunit 1 (nad1) of the mitochondrial genome were used as molecular markers to investigate the presence of cryptic Mesoc

A flotation/sieving method to detect Echinococcus multilocularis and Toxocara spp. eggs in soil by real-time PCR.

Soil can be a source of human infection by many zoonotic helminth species including Echinococcus multilocularis and Toxocara spp. The prevention of alveolar echinococcosis could be greatly improved through the identification of at-risk areas. Yet very few data are available about the detection of E. multilocularis in soil, while more studies have been reported for Toxocara spp. Identification of soil contamination by E. multilocularis eggs requires the use of specific methods. This study describes the development of a method for the detection of E. multilocularis in soil samples with the conce

The opportunistic nature of Trichinella--exploitation of new geographies and habitats.

For more than 100 years, Trichinella spiralis (former Trichina spiralis) was considered a zoonotic parasite of the domestic habitat involving pigs, synanthropic rats and humans. In the last 70 years, there has been increasing evidence that the biomass of nematodes of the genus Trichinella is greater in wild animals than in domestic animals. Omnivores and carnivores (mammals, birds and some reptiles), mainly those with cannibalistic and scavenger behaviour, act primarily as reservoirs for the 12 Trichinella taxa recognized to date. The distribution areas of Trichinella spp. hosts can help to id

Detection of Echinococcus multilocularis in carnivores in Razavi Khorasan province, Iran using mitochondrial DNA.

BACKGROUND: Echinococcus multilocularis is the source of alveolar echinococcosis, a potentially fatal zoonotic disease. This investigation assessed the presence of E. multilocularis infection in definitive hosts in the Chenaran region of Razavi Khorasan Province, northeastern Iran. METHODOLOGY/PRINCIPAL FINDINGS: Fecal samples from 77 domestic and stray dogs and 14 wild carnivores were examined using the flotation/sieving method followed by multiplex PCR of mitochondrial genes. The intestinal scraping technique (IST) and the sedimentation and counting technique (SCT) revealed adult Echinococc

The important zoonoses in the protected areas of the Tatra National Park (TANAP).

The northern part of Slovakia constitutes an important tourist and recreational area of the country. Protected localities of the Tatra National Park (TANAP) are characterised by specific ecological conditions. The high numbers of animals inhabiting protected areas of the TANAP and their potential encounters with tourists contribute to a risk of transmission of important parasitic zoonoses. The aim of presently reported study was to perform a long-lasting and detailed survey on the occurrence of zoonotic agents (in particular Echinococcus multilocularis and Trichinella spp.) with particular emp

Beyond leopards: tooth marks and the contribution of multiple carnivore taxa to the accumulation of the Swartkrans Member 3 fossil assemblage.

The ca. 1.0 myr old fauna from Swartkrans Member 3 (South Africa) preserves abundant indication of carnivore activity in the form of tooth marks (including pits) on many bone surfaces. This direct paleontological evidence is used to test a recent suggestion that leopards, regardless of prey body size, may have been almost solely responsible for the accumulation of the majority of bones in multiple deposits (including Swartkrans Member 3) from various Sterkfontein Valley cave sites. Our results falsify that hypothesis and corroborate an earlier hypothesis that, while the carcasses of smaller an

Reconstructing the evolutionary history of microcephalin, a gene controlling human brain size.

The defining process in the evolution of primates and particularly humans is the dramatic expansion of the brain. While many types of genes could potentially contribute to this process, genes that specifically regulate brain size during development may be especially relevant. Here, we examine the evolution of the microcephalin gene, whose null mutation in humans causes primary microcephaly, a congenital defect characterized by severe reductions in brain size without other gross abnormalities. We show that the evolution of microcephalin's protein sequence is highly accelerated throughout the li

First record of Cryptosporidium infection in a raccoon dog (Nyctereutes procyonoides viverrinus).

Cryptosporidium species have been found in more than 150 species of mammals, but there has been no report in raccoon dogs. Here we found the Cryptosporidium organism in a raccoon dog, Nyctereutes procyonoides viverrinus, and identified this isolate using PCR-based diagnostic methods. Cryptosporidium diagnostic fragments of the 18S ribosomal RNA, Cryptosporidium oocyst wall protein and 70-kDa heat shock protein genes were amplified from the isolate and sequenced to reveal the phylogenetic relationships between it and other Cryptosporidium species or genotypes reported previously. The results sh

Sulfadimethoxine and rhodamine B as oral biomarkers for European badgers (Meles meles).

A field study was carried out on Little Island (County Waterford, Ireland) in June 2000 to evaluate the potential of a bait-marking system for use in European badgers (Meles meles). Two oral biomarkers, sulfadimethoxine (SDM) and rhodamine B, were incorporated into fishmeal baits and distributed by hand at main sets in five test territories for 3 consecutive days. In parallel, non-biomarked baits were distributed at a single control territory. The objectives of the study were to: (1) assess the effects of SDM and rhodamine B on palatability and thus bait acceptance, and (2) investigate the mar

Regular exposure to rabies virus and lack of symptomatic disease in Serengeti spotted hyenas.

We report a previously unrecognized complexity to the ecology of rabies in wildlife. Rabies-specific virus-neutralizing antibodies in spotted hyenas, the most numerous large carnivore in the Serengeti ecosystem (Tanzania, East Africa), revealed a high frequency of exposure of 37.0% to rabies virus, and reverse transcriptase (RT) PCR demonstrated rabies RNA in 13.0% of hyenas. Despite this high frequency, exposure neither caused symptomatic rabies nor decreased survival among members of hyena social groups monitored for 9 to 13 years. Repeated, intermittent presence of virus in saliva of 45.5%
