Leu-M1 antigen expression in advanced (tumor) stage mycosis fungoides.

The authors describe two patients with clinically and histopathologically documented advanced (tumor) stage mycosis fungoides. In each case the large, pleomorphic neoplastic cells lacked the monoclonal antibody-defined cell surface antigens commonly associated with immature and mature T-cells, i.e., T11, Leu-1, T3, T4, T6, T8, and T10, but expressed various T-cell-associated activation antigens, such as HLA-DR, Tac, and T21. Leu-M1, a monocyte-associated antigen, was not expressed by the small, cerebriform neoplastic cells in the plaque stage lesions of either patient. However, Leu-M1 was expr

A simple suspension culture method for generating human iPSC-derived liver organoids.

Human liver in vitro models are indispensable for toxicity studies requiring metabolic competence. However, widely used two-dimensional hepatocyte-derived cell lines exhibit limited drug-metabolizing capacity, whereas primary human hepatocytes are constrained by limited availability, donor variability, dedifferentiation, and poor long-term functional stability. Although three-dimensional liver models offer greater physiological relevance, many existing methods depend on exogenous extracellular matrix (ECM) scaffolds or specialized equipment, thereby limiting standardization, scalability, and b

Epithelial cell TGFβ signaling induces acute tubular injury and interstitial inflammation.

TGFβ signaling plays a central role in the development of acute and chronic kidney diseases. Previous in vivo studies involved systemic alteration of TGFβ signaling, however, limiting conclusions about the direct role of TGFβ in tubular cell injury. Here, we generated a double transgenic mouse that inducibly expresses a ligand-independent constitutively active TGFβ receptor type 1 (TβR1) kinase specifically in tubular epithelial cells, with expression restricted by the Pax8 promoter. In this model, activation of TGFβ signaling in the tubular epithelium alone was sufficient to cause AKI charact

Advances in Astrocyte Research on Neuronal Death and Regeneration.

The onset and progression of central nervous system (CNS) disorders are frequently associated with aberrant neuronal death. In addition to the classical forms of cell death such as apoptosis and necrosis, neurons can also undergo alternative modes of death, including ferroptosis, cuproptosis, and ammonia-induced cell death, all of which may involve the participation of astrocytes. Neuronal death is an irreversible process and plays a central role in the pathogenesis of numerous CNS diseases. We found that astrocytes exhibit the capacity to regenerate into neurons, a characteristic that may pav

In vitro differentiation of pluripotent stem cells into hepatocyte like cells - Basic principles and current progress.

Research in the field of hepatology is limited by the incomplete recapitulation of all major aspects of human hepatic metabolism in most established models. This restricts our ability to study the molecular mechanisms underlying hepatic diseases, and it leads to inadequate assessment of toxicology during drug development, resulting in tremendous unnecessary costs for the pharma industry. Animal models differ in their metabolism compared to the human system, while primary human cells dedifferentiate rapidly and are not suitable for long-term culture and studies. To overcome these obstacles, sev

Characterization of the genomic and transcriptomic landscape of invasive non-mucinous lung adenocarcinoma based on IASLC grading.

BACKGROUND: The IASLC grading system has prognostic utility and potential therapeutic implications in invasive non-mucinous lung adenocarcinoma (LUAD), but the molecular basis underlying the grading spectrum remains unclear. METHODS: We performed whole-genome sequencing in 138 Chinese patients with invasive non-mucinous LUAD and RNA sequencing of 96 matched tumor-normal tissue pairs to systematically characterize the molecular features across grades, including coding driver events, mutational signatures, non-coding regulatory disruptions, and transcriptional programs. RESULTS: Compared with

Superficial dedifferentiated liposarcoma: A clinicopathologic study.

INTRODUCTION: Dedifferentiation occurs in approximately 10% of atypical lipomatous tumors/well-differentiated liposarcomas (ALT/WDLPS), primarily in retroperitoneal or deep-seated tumors, conferring metastatic potential. Superficial dedifferentiated liposarcoma (sDDLPS) is rare, and its progression and natural history are poorly documented. METHODS: We performed a 15-year retrospective review of our pathology database to identify cases of DDLPS in the skin or subcutaneous tissue. Diagnosis of primary sDDLPS required evidence of non-lipogenic sarcoma in the skin or subcutis, with concurrent AL

Heterogeneity of Sox2-expressing cells in mouse pituitary and their roles in postnatal gonadotroph differentiation.

Postnatal differentiation of gonadotrophs from Sox2-expressing stem cells is essential for maturation of the hypothalamic-pituitary-gonadal axis, puberty, and reproduction. Here, we examined the differentiation and maintenance of gonadotrophs in developing and adult mice. Gonadotrophs and Sox2-expressing cells were identified by immunostaining, and gonadotrophs were also visualized by specific expression of the fluorescent protein tdTomato during embryonic and postnatal differentiation. Sox2-expressing cells are localized in the anterior parenchyma, marginal zone, and posterior pituitary, rega

Ectopic Phosphorylated Creb Marks Dedifferentiated Proximal Tubules in Cystic Kidney Disease.

Ectopic cAMP signaling is pathologic in polycystic kidney disease; however, its spatiotemporal actions are unclear. We characterized the expression of phosphorylated Creb (p-Creb), a target and mediator of cAMP signaling, in developing and cystic kidney models. We also examined tubule-specific effects of cAMP analogs in cystogenesis in embryonic kidney explants. In wild-type mice, p-Creb marked nephron progenitors (NP), early epithelial NP derivatives, ureteric bud, and cortical stroma; p-Creb was present in differentiated thick ascending limb of Henle, collecting duct, and stroma; however, it

Multi-omics analysis unraveling stemness features associated with oncogenic dedifferentiation in 12 cancers.

Tumorigenesis is typically accompanied by cellular dedifferentiation and the acquisition of stem cell-like attributes. However, few studies have comprehensively evaluated the putative relationships between these characteristics and various cancers. Here, we integrated gene expression and DNA methylation quantitative trait loci (cis-eQTL and cis-mQTL) data from the blood to perform multi-omics Mendelian randomization analysis. Our analyses revealed 967 stem cell-associated genes (P < 0.05) and 11,262 methylation sites (P < 0.01) significantly related to 12 cancers. SMAD7 (cg14321542) in colon c

Epigenetic memory as crucial contributing factor in directing the differentiation of human iPSC into pancreatic β-cells in vitro.

Impaired insulin secretion contributes to the pathogenesis of type 1 diabetes mellitus through autoimmune destruction of pancreatic β-cells and the pathogenesis of severe forms of type 2 diabetes mellitus through β-cell dedifferentiation and other mechanisms. Replenishment of malfunctioning β-cells via islet transplantation has the potential to induce long-term glycemic control in the body. However, this treatment option cannot widely be implemented in clinical due to healthy islet donor shortage. Emerging β-cell replacement with human-induced pluripotent stem cell (iPSC) provides high remedia…

Advances in salivary gland pathology.

This review summarizes the new findings on salivary gland pathology under the following categories: immunohistochemistry; molecular genetics; newly recognized tumour types; known tumour entities with new findings; and progression of salivary gland tumours. In the application of immunohistochemistry, CD117 can aid in highlighting the luminal cell component of various salivary gland tumours, whereas p63 or maspin can aid in highlighting the abluminal cell component. A high Ki67 index remains the most useful marker to predict adverse outcome in salivary gland carcinoma. Specific chromosomal trans

Anti-tumor effects of tirbanibulin in squamous cell carcinoma cells are mediated via disruption of tubulin-polymerization.

Topical tirbanibulin is a highly effective and well tolerated novel treatment option for actinic keratoses (AKs). This study aimed to characterize the mode of action of tirbanibulin in keratinocytes (NHEK) and cutaneous squamous cell carcinoma (cSCC) cell lines (A431, SCC-12) in vitro. Tirbanibulin significantly reduced proliferation in a dose-dependent manner in all investigated cell lines, inhibited migration, and induced G2/M-cell cycle arrest only in the cSCC cell lines analyzed, and induced apoptosis solely in A431, which showed the highest sensitivity to tirbanibulin. In general, we dete

KCa3.1 Channel Modulators as Potential Therapeutic Compounds for Glioblastoma.

BACKGROUND: The intermediate-conductance Ca2+-activated K+ channel KCa3.1 is widely expressed in cells of the immune system such as T- and B-lymphocytes, mast cells, macrophages and microglia, but also found in dedifferentiated vascular smooth muscle cells, fibroblasts and many cancer cells including pancreatic, prostate, leukemia and glioblastoma. In all these cell types KCa3.1 plays an important role in cellular activation, migration and proliferation by regulating membrane potential and Ca2+ signaling. METHODS AND RESULTS: KCa3.1 therefore constitutes an attractive therapeutic target for d

Requirement of protein tyrosine phosphatase SHP2 for NO-stimulated vascular smooth muscle cell motility.

We have previously reported that nitric oxide (NO) increases the motility of differentiated cultured primary aortic smooth muscle cells from adult rats. There is little information on the role of protein tyrosine phosphatases in vascular biology. One such phosphatase, Src homology 2 phosphatase 2 (SHP2), is essential for motility. We tested the hypothesis that NO increases SHP2 levels via a cGMP-mediated mechanism and that this effect is necessary for NO-stimulated cell motility. Here we report that two different NO donors increased SHP2 protein levels and enzyme activity. This effect was mimi

Repair and healing of established gastric mucosal injury.

Superficial injury of the gastric mucosa, such as occurs after ethanol or aspirin ingestion, is usually resurfaced in 1 or 2 h. This is accomplished by rapid migration of surface mucous cells to cover the defect, followed by a later increase in cellular proliferation to restore the pits to their original length. This rapid restitution has been documented with electron microscopy, and is also reflected in the speedy return to baseline of the transmucosal potential difference and ionic barrier function after superficial injury. Deeper mucosal injury--which occurs focally after injury by aspirin,

Substrate stiffness and pressure alter retinal Müller glia response and extracellular matrix production.

BACKGROUND: The retina is highly influenced by its mechanical environment, with Müller glia (MG) acting as key mechanosensors and extracellular matrix (ECM) producers. This study examined MG responses to substrate stiffness and high pressure (HP), and whether TGF-β1 modulation could mitigate these effects. METHODS: Primary MG from adult rat retinas were cultured on glass (Young's modulus E'=∼1 gigapascal (GPa)) and polyacrylamide gels (10 kPa and 100 kPa). MG were exposed to atmospheric and 70 mmHg (HP) conditions, with TGF-β1 pharmacologically blocked. RESULTS: On glass and 100 kPa gels, MG

Diagnosing idiosyncratic drug-induced liver injury: lessons from monocyte-derived hepatocyte-like cells.

INTRODUCTION: Idiosyncratic drug-induced liver injury (iDILI) is an unpredictable adverse drug event that often involves an abnormal immune response. iDILI is a challenge in the development of new drugs and the diagnosis in clinical practice. Hepatocyte-like cells generated from monocytes of iDILI patients are being developed as an ex vivo assay to enhance understanding of underlying molecular mechanisms and to assist in drug causality assessment, especially in cases involving multiple agents. Brinker regulates reciprocal outcomes of BMP signal between stem cells and differentiating cells.

Drosophila male germline stem cells (GSCs) reside at the testis tip, surrounding a cluster of niche cells known as the hub. Bone Morphogenetic Protein (BMP) ligands secreted from the hub exert both contact-dependent and -independent effects. In close proximity to the niche, BMP signaling maintains stem cells by suppressing transcription of the key differentiation factor Bag of Marbles (Bam). In contrast, the diffusible fraction of BMP promotes differentiation of cells by activating bam. How a single signaling pathway produces such opposing outcomes has remained unclear. Here, we show that the
