The Flavohemoglobin Hmp and Nitric Oxide Reductase Restrict Initial nir Expression in the Bet-Hedging Denitrifier Paracoccus denitrificans by Curtailing Hypoxic NO Signalling.

In denitrifying bacteria, nitric oxide (NO) is an electron acceptor and a free intermediate produced during anaerobic respiration. NO is also a signal for transcriptional regulation of the genes encoding nitrite (Nir), nitric oxide (Nor) and nitrous oxide reductases (N2OR). We hypothesise that the timing and strength of the NO signal necessary for full nir expression are key factors in the bet-hedging strategy of Paracoccus denitrificans, and that systems scavenging NO under hypoxia reduce the probability of nir induction. We show that the flavohemoglobin Hmp scavenges NO in aerobic cultures a

The Human Gut Colonizer Blastocystis Respires Using Complex II and Alternative Oxidase to Buffer Transient Oxygen Fluctuations in the Gut.

Blastocystis is the most common eukaryotic microbe in the human gut. It is linked to irritable bowel syndrome (IBS), but its role in disease has been contested considering its widespread nature. This organism is well-adapted to its anoxic niche and lacks typical eukaryotic features, such as a cytochrome-driven mitochondrial electron transport. Although generally considered a strict or obligate anaerobe, its genome encodes an alternative oxidase. Alternative oxidases are energetically wasteful enzymes as they are non-protonmotive and energy is liberated in heat, but they are considered to be in

Osteopontin deficiency ameliorates Alport pathology by preventing tubular metabolic deficits.

Alport syndrome is a rare hereditary renal disorder with no etiologic therapy. We found that osteopontin (OPN) is highly expressed in the renal tubules of the Alport mouse and plays a causative pathological role. OPN genetic deletion ameliorated albuminuria, hypertension, tubulointerstitial proliferation, renal apoptosis, and hearing and visual deficits in the Alport mouse. In Alport renal tubules we found extensive cholesterol accumulation and increased protein expression of dynamin-3 (DNM3) and LDL receptor (LDLR) in addition to dysmorphic mitochondria with defective bioenergetics. Increased

Vertebrate endoderm development.

The endoderm forms the innermost layer of the embryonic gut tube. It gives rise to the epithelial and glandular components of many internal organs, including those of the digestive and respiratory systems. Endodermal organs have essential physiological processes such as nutrient absorption, respiration, detoxification and endocrine regulation. Over the past two decades, work in vertebrate embryos and human pluripotent stem cell models has defined the mechanisms that govern endoderm formation, regionalization and organogenesis. These processes are driven by conserved cell signaling pathways act

Metformin dual-targets metabolism and survival pathways in BPDCN.

Blastic plasmacytoid dendritic cell neoplasm (BPDCN) is a rare and aggressive hematologic malignancy with limited therapeutic options. Metformin, a commonly prescribed antidiabetic drug, has recently gained attention for its anticancer potential, but its effects on BPDCN remain unknown. Here, we show that metformin reduces cell viability and induces caspase-dependent apoptosis in both established (CAL-1, GEN2.2) and primary BPDCN cells, partly through activation of the intrinsic apoptotic pathway. Mechanistically, metformin activates AMPK and disrupts mitochondrial respiration and glycolysis,

Novel SLC5A6 mutations lead to B lymphocyte maturation defects with metabolic abnormality rescuable by biotin replenishment.

We characterized a family diagnosed with immunodeficiency disease presenting with low immunoglobulin levels and skin dyskeratosis. Exome sequencing revealed compound heterozygous missense variants in SLC5A6, the gene encoding a cellular sodium-dependent multivitamin transporter (SMVT) responsible for transporting vitamins, including biotin (vitamin B7). We showed that the biotin deficiency was caused by the SLC5A6 variants resulting in defective B cell differentiation and antibody deficiency. Altered cellular metabolic profiles, including aberrant mitochondrial respiration and reliance on glyc

[Level of cyclic AMP during induction of alternative oxidase in Yarrowia lipolytica yeast cells].

The effect of cyanide, antimycin A, ethanol, and acetate on the induction of alternative oxidase in the yeast Yarrowia lipolytica VKM Y-155 sensitive to cyanide, in the presence of the aforementioned compounds led to the development of cyanide-resistant respiration, which could be suppressed by benzohydroxamic acid, an inhibitor of alternative oxidases. The incubation of cells with cyanide, ethanol, or acetate raised the intracellular pool of cAMP, which attained maximal values after a 2- to 3-min incubation, then rapidly decreased to the initial value and did not change over the next three ho

Mitochondrial Function and Resistance to Oxidative Stress in the Kidney during Pregnancy.

We demonstrated that the serum of pregnant rats increases viability of kidney epithelial cells and promotes their proliferation. The intensity of oxidative stress in the kidneys was also reduced during pregnancy, but only in rats that were not exposed to acute ischemic kidney injury. This decrease in oxidative stress was not associated with changes in transmembrane mitochondrial potential, the size of mitochondria, time of opening of mitochondrial permeability transition pore (mPTP), mitochondrial respiration rate, antioxidant activity, or nitric oxide level.

Betaine delays age-related muscle loss by mitigating Mss51-induced impairment in mitochondrial respiration via Yin Yang1.

BACKGROUND: Mitochondrial dysfunction is one of the hallmarks of aging and a leading contributor to sarcopenia. Nutrients are essential for improving mitochondrial function and skeletal muscle health during the aging process. Betaine is a nutrient with potential muscle-preserving properties. However, whether and how betaine could regulate the mitochondria function in aging muscle are poorly understood. We aimed to explore the molecular target and underlying mechanism of betaine in attenuating the age-related mitochondrial dysfunction in skeletal muscle. METHODS: Young mice (YOU, 2 months), ol

Microbes as Key Players During Evolution: I From the Beginnings to the Appearance of Eukaryotes.

Life on Earth originated in the Precambrian, around 4 billion years ago. This review focuses on microbes (viruses, bacteria, archaea) which developed in these prehistoric times and their roles during the later evolution of protists (eukaryotes). Among the different theories of life evolution, the author favors the theory of the RNA world as having been first. Over the past few decades, new molecular biological technologies and findings have changed the paradigm of mutation and selection as being the main driving force of biological evolution. Instead, microbes and microbe-derived genetic eleme

Secreted phosphoprotein 1 slows neurodegeneration and rescues visual function in mouse models of aging and glaucoma.

Aging causes an irreversible, cumulative decline in neuronal function. Using the visual system as a model, we show that astrocytes play a critical role in maintaining retinal ganglion cell health and that deletion of SPP1 (secreted phosphoprotein 1, or osteopontin) from astrocytes leads to increased vulnerability of ganglion cells to age, elevated intraocular pressure, and traumatic optic nerve damage. Overexpression of SPP1 slows the age-related decline in ganglion cell numbers and is highly protective of visual function in a mouse model of glaucoma. SPP1 acts by promoting phagocytosis and se

Fludarabine disrupts the STAT1/AhR interaction to attenuate type 1 diabetes by inducing tolerogenic dendritic cells along with targeting effector T cells.

BACKGROUND: Loss of immune homeostasis in Type 1 diabetes (T1D) leads to a dysregulated and autoreactive immune response that destroys pancreatic β cells, causing absolute insulin deficiency. Nevertheless, current strategies for restoring immune homeostasis remain limited. METHOD: Inspired by our earlier research, we leveraged the single-cell RNA sequencing data from T1D patients, and unexpectedly found that STAT1 overrepresentation is much more prominent in dendritic cells (DCs) rather in CD4 T cells. Subsequently, we repurposed the clinically-applied STAT1 inhibitor fludarabine in T1D settin

Low Temperature Suppress Cell Wall Degradation by Modulating Antioxidant Metabolism in Figs.

The effects of low (4 °C) versus ambient (25 °C) temperature on cell wall turnover, and redox homeostasis in 'Jin'Aofen' figs were investigated. Compared with 25 °C, storage at 4 °C reduced decay and respiration rates, delayed firmness loss and color deterioration. It also suppressed PG, PME and CEL activities, maintained higher contents of protopectin and cellulose; and slowed soluble pectin accumulation. Storage at 4 °C maintained higher activities of SOD, CAT, POD, APX and GR. It also preserved the levels of non-enzymatic antioxidants, including TPC, TFC, ASA GSH. Meanwhile, it reduced H2O2

From copper homeostasis to cuproptosis: a new perspective on CNS immune regulation and neurodegenerative diseases.

Copper, an essential trace element for the human body, plays a key role in energy metabolism, mitochondrial respiration, redox reactions, and neural signal transmission. The recently proposed concept of "cuproptosis" has further revealed the unique status of copper in cellular regulation: when copper abnormally accumulates within cells, it can directly bind to the lipoylated proteins of the mitochondrial TCA cycle, triggering protein aggregation and metabolic disorders, ultimately leading to cell death. This form of cell death plays an important role in various neurodegenerative diseases of th

Acidosis orchestrates adaptations of energy metabolism in tumors.

Malignant tumors are characterized by diverse metabolic stresses, including nutrient shortages, hypoxia, and buildup of metabolic by-products. To understand how cancer cells adapt to such challenges, we conducted sequential CRISPR screens to identify genes that affect cellular fitness under specific metabolic stress conditions in cell culture and to then probe their relevance in pancreatic tumors. Comparative analyses of hundreds of fitness genes revealed that cancer metabolism in vivo was shaped by bioenergetic adaptations to tumor acidosis. Mechanistically, acidosis suppressed cytoplasmic ac

Glucose sensitizes the stationary and persistent population of Vibrio cholerae to ciprofloxacin.

The subject of analysis in this report was the antibiotic susceptibility of V. cholerae under glucose supplementation since the metabolites can significantly alter the antibiotic sensitivity of bacteria. Glucose could change the antibiotic susceptibility in a growth phase-dependent manner, however, the antibiotic susceptibility of exponentially growing cells was not affected in the presence of glucose. What has been shown is that the stationary phase cells which show higher antibiotic tolerance, could be sensitized to ciprofloxacin and ampicillin by glucose supplementation (tenfold sensitive).

[Oxidative phosphorylation capacity of Rhodopseudomonas palustris during growth in light and darkness].

Assimilation of oxygen by the cells of Rhodopseudomonas palustris grown in the light and in the darkness is stimulated by p-chlorocarbonylcyanidephanylhydrazone, suggesting respiration coupled to phosphorylatin. Membranes of the cells grown in the light are capable of oxidative phosphorylation in the course of electron transport from NADH and succinate to O2 with P/O being 0.03 and 0.20, respectively. Membranes of the cells grown in the darkness in aerobic conditions are capable of oxidative phosphorylation in the presence of NADH, succinate and ascorbate + phenazinemetasulphate with P/O being
