Substance Use Disorder (SUD) involves emotional, cognitive, and motivational dysfunction. Long-lasting molecular and structural changes in brain regions functionally and anatomically linked to the cerebellum, such as the prefrontal cortex, amygdala, hippocampus, basal ganglia, and ventral tegmental area, are characteristic of SUD. Direct and indirect reciprocal connectivity between the cerebellum and these brain regions can explain cerebellar roles in Pavlovian and reinforcement learning, fear memory, and executive functions. It is increasingly clear that the cerebellum modulates brain functio

Peripheral inputs continuously shape brain function and can influence memory acquisition, but the underlying mechanisms have not been fully understood. Cannabinoid type-1 receptor (CB1R) is a well-recognized player in memory performance, and its systemic modulation significantly influences memory function. By assessing low arousal/non-emotional recognition memory in mice, we found a relevant role of peripheral CB1R in memory persistence. Indeed, the peripherally-restricted CB1R specific antagonist AM6545 showed significant mnemonic effects that were occluded in adrenalectomized mice, and after

Although cerebellar alterations have been implicated in stress symptoms, the exact contribution of the cerebellum to stress symptoms remains to be elucidated. Here, we demonstrated the crucial role of cerebellar neurons projecting to the ventral tegmental area (VTA) in the development of chronic stress-induced behavioral alterations in mice. Chronic chemogenetic activation of inhibitory Purkinje cells in crus I suppressed c-Fos expression in the DN and an increase in immobility in the tail suspension test or forced swimming test, which were triggered by chronic stress application. The combinat

Hypothalamic agouti-related peptide (AgRP) and neuropeptide Y-expressing neurons have a critical role in driving food intake, but also in modulating complex, non-feeding behaviours1. We interrogated whether AgRP neurons are relevant to the emergence of anorexia nervosa symptomatology in a mouse model. Here we show, using in vivo fibre photometry, a rapid inhibition of AgRP neuronal activity following voluntary cessation of running. All AgRP neuron-ablated, food-restricted mice die within 72 h of compulsive running, while daily activation of AgRP neurons using a chemogenetic tool increases volu

Our understanding of depression and its treatment has advanced with the advent of ketamine as a rapid-acting antidepressant and the development and refinement of tools capable of selectively altering the activity of populations of neuronal subtypes. This work has resulted in a paradigm shift away from dysregulation of single neurotransmitter systems in depression towards circuit level abnormalities impacting function across multiple brain regions and neurotransmitter systems. Studies on the features of circuit level abnormalities demonstrate structural changes within the prefrontal cortex (PFC

The dorsal medial prefrontal cortex (dmPFC) has been recognized as a key cortical area for nociceptive modulation. However, the underlying neural pathway and the function of specific cell types remain largely unclear. Here, we show that lesions in the dmPFC induced an algesic and anxious state. Using multiple tracing methods including a rabies-based transsynaptic tracing method, we outlined an excitatory descending neural pathway from the dmPFC to the ventrolateral periaqueductal gray (vlPAG). Specific activation of the dmPFC/vlPAG neural pathway by optogenetic manipulation produced analgesic

State-dependent memory describes a phenomenon that memory will be efficiently retrieved only when the brain state during retrieval matches the state during encoding. While a variety of psychoactive drugs, such as ethanol, cocaine, morphine and NMDA receptor antagonists, are able to induce state-dependent memory, the biological hallmark of brain state and neural mechanism of its regulation are still unknown. In this study, we found that MK-801 enhanced delta oscillations in awake mice, representing a drug-induced brain state, in which fear memory could only be successfully retrieved when the sa

Neurons containing melanin-concentrating hormone (MCH) in the posterior lateral hypothalamus play an integral role in rapid eye movement sleep (REMs) regulation. As MCH neurons also contain a variety of other neuropeptides [e.g., cocaine- and amphetamine-regulated transcript (CART) and nesfatin-1] and neurotransmitters (e.g., glutamate), the specific neurotransmitter responsible for REMs regulation is not known. We hypothesized that glutamate, the primary fast-acting neurotransmitter in MCH neurons, is necessary for REMs regulation. To test this hypothesis, we deleted vesicular glutamate trans

OBJECTIVES: To investigate the role of a neural pathway from oxytocin (OXT) neurons in the paraventricular hypothalamic nucleus (PVN) to γ-aminobutyric acid (GABA) neurons (GABAergic neurons) in the trigeminal nucleus caudalis (TNC) in regulating pain sensitization in a mouse model of chronic migraine and to explore the underlying mechanisms.
METHODS: A chronic migraine mouse model was established by intraperitoneal injection of nitroglycerin (NTG, 1 mg/mL, 10 mg/kg) on days 1, 3, 5, 7, and 9. The study consisted of four parts: PartⅠ: 24 male wild-type C57BL/6J mice were divided into four gro

Nociceptors - peripheral nervous system neurons that trigger the sensation of pain or itch in response to noxious stimuli - can communicate with leukocytes and modulate immune responses in a context-dependent manner 1,2 . Here, we demonstrate that nociceptors rapidly induce monocytes to adopt an immunosuppressive phenotype, reminiscent of myeloid-derived suppressor cells (MDSCs), which have been linked to impaired anti-tumor immunity. MDSCs are abundant in human bladder cancer, a malignancy where survival is negatively correlated with expression of pro-neurogenic cytokines. Accordingly, in an

Neuropathic pain after spinal cord injury reflects persistent hyperexcitability in the spinal cord dorsal horn, yet the molecular drivers sustaining this maladaptive state are unknown. Using an antibody microarray of dorsal horn tissue from mice six weeks after cervical contusion spinal cord injury, we found persistent upregulation of Eph-ephrin signaling, including increased EphB1, EphB2 and EphB3 expression and phosphorylation. Reversible chemogenetic inhibition of EphB kinase activity, using an EphB1/2/3 analog-sensitive knock-in mouse, selectively reversed established mechanical allodynia

Acquired cognitive impairments occur in diverse neurological and psychiatric conditions, yet the mechanisms linking neural circuit activity to neurovascular function remain poorly understood. Using a mouse model of nicotine withdrawal (WD), we identified an activity-dependent NEPAS-pentraxin-3 (PTX3) axis in mPFC neurons that couples circuit hypoactivity to cognitive deficits. In this model, NEPAS expression is significantly upregulated in mPFC neurons, accompanied by reduced myelin formation. Neuronal activity in mPFC and PVA is suppressed and chemogenetic activation of the PVA-mPFC neural ci

Postoperative cognitive dysfunction (POCD) negatively impacts prognosis; however, the underlying mechanisms remain unclear. We demonstrated that tibial fracture surgery led to cognitive dysfunction in 18-month-old mice, concomitant with a reduction in hippocampal levels of cholesterol and its key metabolite 24-hydroxycholesterol (24-OHC). Clinically, reduced blood 24-OHC levels were associated with cognitive decline in elderly surgery patients. Mechanistically, downregulation of sterol regulatory element-binding protein 2 (SREBP2) in reactive astrocytes of the hippocampal dorsal CA1 (dCA1) reg

Although cortical-thalamic projections contribute to pain modulation, their downstream targets and functional roles are not well defined. Here, we show that glutamatergic neurons in the hindlimb region of primary somatosensory cortex (S1HL Glu ) project to a subset of glutamatergic neurons in the ventral posterolateral thalamic nucleus (VPL Glu ) that are anatomically separated from VPL neurons receiving peripheral afferent inputs. These VPL Glu neurons preferentially innervate S1HL Glu neurons, forming a recurrent glutamatergic corticothalamocortical pathway. Optogenetic and chemogenetic acti

Opioid addiction and misuse are a serious national crisis that affects public health, as well as social and economic welfare. Mortality due to opioid misuse is further exasperated by the combination of opioids with non-opioid respiratory depressants such as xylazine that are resistant to mu opioid receptor antagonists such as naloxone. This study tested the hypothesis that oxytocin can mitigate the severe opioid induced respiratory depression (OIRD) and mortality induced by high doses of fentanyl or the combination of fentanyl with xylazine. Our results show OXT can improve survival and respir

Musashi2 (MSI2) plays a crucial role in neural stem cells and reproductive regulation, yet its mechanism in integrating the neuro-endocrine system remains poorly understood. This study investigates how MSI2 influences ovarian function by using conditional knockout mice for Msi2 in the brain and ovary. Our results show that KRT14/MSI2 neurons in the dentate gyrus (DG) of the hippocampus directly project to the ovary. Deletion of Msi2 in the DG and ovary increases follicle number and litter size, but causes a decline in fertility and follicle reserve after 6 months of age. AAV-mediated specific

Perfusion through leptomeningeal collateral vessels is a likely pivotal factor in the outcome of ischemic stroke patients. Acupuncture has been reported to restore cerebral blood flow (CBF) after acute ischemic stroke, but the underlying mechanisms are poorly understood. This study aimed to examine whether electroacupuncture (EA) could improve CBF following an acute ischemic injury by regulating leptomeningeal collaterals. The clinical outcomes suggest that EA resulted in an increase in the average CBF within the whole brain and gray matter of healthy subjects compared to pre-intervention. The
