Computational screening of oncogenic genetic variations in tumor suppressor proteins driving gastric cancer pathogenesis.

Gastric cancer (GC) is currently the fifth most common cancer globally, often driven by dysregulation of tumor suppressor pathways. While individual studies on genetic variations of proteins are common, a comprehensive systems-level analysis of proteins regulating GC pathways showing both expression dysregulation and high mutation frequency remains unexplored. Therefore, our study aimed to identify critical genes and their pathogenic variations disrupting the tumor-suppressive capacity of the GC pathway. We employed a deep learning-based graph neural network model to identify genes exhibiting

Alpha 2 agonists for sedation to produce better outcomes from critical illness (A2B trial): protocol for a mixed-methods process evaluation of a randomised controlled trial.

INTRODUCTION: An association between deep sedation and adverse short-term outcomes has been demonstrated although this evidence has been inconsistent. The A2B (alpha-2 agonists for sedation in critical care) sedation trial is designed to determine whether the alpha-2 agonists clonidine and dexmedetomidine, compared with usual care, are clinically and cost-effective. The A2B intervention is a complex intervention conducted in 39 intensive care units (ICUs) in the UK. Multicentre organisational factors, variable cultures, perceptions and practices and the involvement of multiple members of the h

Nested and multipart prospective observational studies, flaming fiasco or efficiently economical?: The Brain, Bone, Heart case study.

BACKGROUND: Collecting new data from cross-sectional/survey and cohort observational study designs can be expensive and time-consuming. Nested (hierarchically cocooned within an existing parent study) and/or Multipart (≥ 2 integrally interlinked projects) study designs can expand the scope of a prospective observational research program beyond what might otherwise be possible with available funding and personnel. The Brain, Bone, Heart (BBH) study provides an exemplary case to describe the real-world advantages, challenges, considerations, and insights from these complex designs. MAIN: BBH is

Factors promoting and impeding efforts to deprescribe antidepressants among nursing home residents with dementia- a process evaluation guided by normalization process theory.

BACKGROUND: Despite recommendations against psychotropic medication in older nursing homes residents with behavioral and psychological symptoms of dementia (BPSD), antidepressants and other psychotropic drugs are still prescribed. We performed a cluster-randomized controlled trial to evaluate the effect of a complex intervention aiming to promote the deprescribing of antidepressants in institutionalized older persons with dementia. To understand the underlying mechanisms of trial outcomes, we conducted a process evaluation exploring the interventions implementation, areas of impact, and contex

Rare and common genetic variations in the Keap1/Nrf2 antioxidant response pathway impact thyroglobulin gene expression and circulating levels, respectively.

Nuclear factor, erythroid 2-like 2 (Nrf2) is a transcription factor that has been gaining attention in the field of pharmacology and especially in the chemoprevention of diseases such as cancer, metabolic and neurodegenerative diseases, etc. This is because natural compounds such as sulforaphane, which is found in broccoli sprout extracts, can activate Nrf2. The repertoire of the roles of Nrf2 is ever increasing; besides its traditional antioxidant and cytoprotective effects, Nrf2 can have other functions as a transcription factor. We have recently shown that Nrf2 directly regulates the expres

Microevolution of epidemiological highly relevant non-O157 enterohemorrhagic Escherichia coli of serogroups O26 and O111.

Enterohemorrhagic Escherichia coli (EHEC) are a cause of bloody diarrhea, hemorrhagic colitis (HC) and the potentially fatal hemolytic uremic syndrome (HUS). While O157:H7 is the dominant EHEC serotype, non-O157 EHEC have emerged as serious causes of disease. In Germany, the most important non-O157 O-serogroups causing one third of EHEC infections, including diarrhea as well as HUS, are O26, O103, O111 and O145. Interestingly, we identified EHEC O-serogroups O26 and O111 in one single sequence type complex, STC29, that also harbours atypical enteropathogenic E. coli (aEPEC). aEPEC differ from

Dissemination of pheU- and pheV-located genomic islands among enteropathogenic (EPEC) and enterohemorrhagic (EHEC) E. coli and their possible role in the horizontal transfer of the locus of enterocyte effacement (LEE).

We have recently shown that the locus of enterocyte effacement (LEE) of the bovine enterohemorrhagic E. coli RW1374 (O103:H2) resides within a large pathogenicity island (PAI), integrated in the vicinity of the phenylalanine tRNA gene pheV. Here we describe an additional, but LEE-negative genomic island in RW1374 in the vicinity of another phenylalanine tRNA gene, pheU, the sequence of which is identical to pheV. These two genomic islands revealed identity of the left, but a relative variability of their right end sequences. To investigate the mechanism of LEE-PAI distribution in E. coli, we a

The effect of image reconstruction kernel and density modulus relationship in finite element models of simulated cadaveric acromial loading.

Fractures of the acromion are a common complication following surgical procedures of the shoulder due to changes in joint biomechanics. To improve understanding of acromial stresses and evaluate surgical procedures, image-based finite element models (FEMs) may be used. Image-based FEMs are dependent on accurate volumetric bone mineral density (vBMD), as this relates to mechanical properties in FEMs. Image reconstruction kernel alters vBMD; however, the effect on FEM output during simulated loading of the acromion has not been reported. The objective of this study was to compare predicted force

Transposon activity, local duplications and propagation of structural variants across haplotypes drive the evolution of the Drosophila S2 cell line.

BACKGROUND: Immortalized cell lines are widely used model systems whose genomes are often highly rearranged and polyploid. However, their genome structure is seldom deciphered and is thus not accounted for during analyses. We therefore used linked short- and long-read sequencing to perform haplotype-level reconstruction of the genome of a Drosophila melanogaster cell line (S2-DRSC) with a complex genome structure. RESULTS: Using a custom implementation (that is designed to use ultra-long reads in complex genomes with nested rearrangements) to call structural variants (SVs), we found that the

Artificial valves to replace the Ureterovesical Junctions: a novel approach to prevent urinary backflow.

Vesicoureteral reflux is a common complication following radical cystectomy, resulting from the removal of the ureterovesical junctions (UVJ). Current bladder reconstruction methods do not address this issue, leading to potential renal complications. In this study, we designed, fabricated, and tested two passive ureteral valve designs - the Butterfly and Cap valves - to ensure unidirectional urine flow from the kidneys to the bladder when inserted into the ureters of an artificial bladder (AB). Finite Element Method simulations guided the optimization of valves. The prototypes were 3D-printed

Finite-element modelling of interactions of needle with tympanic membrane and middle ear.

The tympanic membrane (TM) is one of the most common routes to access the middle ear and inner ear for the treatment of hearing and balance pathologies. Since the TM is a soft thin biological tissue with small dimensions, using needles seems to be among the most practical interventional approaches. In this study, we proposed a finite-element (FE) analysis of needle-TM interactions that combines a 3D model of the TM and other main middle-ear structures in gerbil, and a 2D model of needle insertion into the TM based on the cohesive zone method (CZM). The TM was modelled using a 1st-order Ogden h

Biomechanism of resistance to retinal injury in woodpecker's eyes.

Retinal injury is the most common ocular impairment associated with shaken baby syndrome (SBS), which could lead to vision loss and blindness. However, a woodpecker does not develop retinal hemorrhages or detachment even at a high acceleration of 1,000×g during pecking. To understand the mechanism of retinal injury and its resistance strategy, we put insight into the special ability of the woodpecker to protect the retina against damage under acceleration-deceleration impact. In this study, the structural and mechanical differences on the eyes of the woodpecker and human were analyzed quantita

A porous fibrous hyperelastic damage model for human periodontal ligament: Application of a microcomputerized tomography finite element model.

The periodontal ligament (PDL) is a soft biological tissue that connects the tooth with the trabecular bone of the mandible. It plays a key role in load transmission and is primarily responsible for bone resorption and most common periodontal diseases. Although several numerical studies have analysed the biomechanical response of the PDL, most did not consider its porous fibrous structure, and only a few analysed damage to the PDL. This study presents an innovative numerical formulation of a porous fibrous hyperelastic damage material model for the PDL. The model considers two separate softeni

Effects of recombinant human erythropoietin in infants with very low birth weights.

Anaemia of prematurity, a postnatal fall in haemoglobin concentration and haematocrit, is particularly common in those born at less than 32 weeks of gestation. Experimental and clinical data implicate inadequate erythropoietin production as an important reason. In this study recombinant human erythropoietin (r-HuEpo) was used in an attempt to treat or prevent this anaemia and thereby provide an alternative to erythrocyte transfusions. Premature infants (birth weight < or = 1250 g and gestational age < or = 32 weeks), who were likely to need transfusions, were randomly assigned to receive 4 wee

Molecular convergence of risk variants for congenital heart defects leveraging a regulatory map of the human fetal heart.

Congenital heart defects (CHD) arise in part due to inherited genetic variants that alter genes and noncoding regulatory elements in the human genome. These variants are thought to act during fetal development to influence the formation of different heart structures. However, identifying the genes, pathways, and cell types that mediate these effects has been challenging due to the immense diversity of cell types involved in heart development as well as the superimposed complexities of interpreting noncoding sequences. As such, understanding the molecular functions of both noncoding and coding

Electronic structure and metal-metal bonding in early lanthanide dimers La2, Ce2, and Pr2.

Understanding whether 4f electrons participate directly in lanthanide-lanthanide bonding remains a long-standing question in f-element chemistry. Lanthanide dimers provide the simplest molecular platform for isolating intrinsic metal-metal bonding interactions without ligand-field or oxidation-state complications. Here, we present a fully relativistic multireference investigation of La2, Ce2, and Pr2 to resolve their ground electronic states and elucidate the evolution of bonding across the early lanthanide series. The ground states under spin-orbit coupling are determined to be 0g, 1g, and 2u

AC Electrokinetics-Enhanced Capacitive Aptasensor for Point-of-Care Testing of Acrylamide in Coffee.

Acrylamide (AA) is a common contaminant in foods processed at high temperatures and has attracted significant attention due to its potential neurotoxicity and carcinogenicity. Therefore, the development of a highly sensitive, highly selective sensing technology suitable for on-site detection is of great importance for ensuring food safety. In this study, an aptamer (Apt)-based capacitive AA sensor was developed based on the alternating current electrokinetics (ACEK) effect. The sensor utilizes an aptamer as the biomimetic recognition element, which can specifically recognize AA, thereby enabli
