High fidelity wastewater surveillance as an early warning tool for diphtheria in the German Ruhr metropolitan area.

INTRODUCTION: Diphtheria is re-emerging as a public health concern in Europe, with recent fatalities underlining the need for strengthened surveillance. In highly vaccinated populations, limited clinical suspicion and reduced routine testing can obscure low-level circulation of toxigenic strains, increasing the risk of silent spread. In addition to classical Corynebacterium diphtheriae infections, the potential mobilization of the diphtheria toxin (tox) gene within and possibly beyond different Corynebacterium species represents a growing threat. METHODS: We developed a high-fidelity wastewat…

Valid and accepted novel bacterial taxa derived from human clinical specimens and taxonomic revisions published in 2022.

Although some nomenclature changes have caused consternation among clinical microbiologists, the discovery of novel taxa and improving classification of existing groups of organisms is exciting and adds to our understanding of microbial pathogenesis. In this mini-review, we present an in-depth summary of novel taxonomic designations and revisions to prokaryotic taxonomy that were published in 2022. Henceforth, these bacteriology taxonomic summaries will appear annually. Several of the novel Gram-positive organisms have been associated with disease, namely, the Corynebacterium kroppenstedtii-li…

The microbiology of chronic rhinosinusitis with and without nasal polyps.

CONCLUSION: Staphylococcus aureus, Streptococcus, Haemophilus, Enterobacter, and Corynebacterium appear to be more frequently associated with patients with chronic rhinosinusitis (CRS) with nasal polyps (CRSwNP) than with patients with CRS without nasal polyps (CRSsNP) or control subjects. Furthermore, the isolation rate of Staphylococcus aureus appears to be much lower in Chinese CRSwNP patients compared with their caucasian counterparts. OBJECTIVES: To compare the predominant bacteriologic profiles in middle meatus of patients with CRSwNP, those with CRSsNP, and control subjects. METHODS: …

Impact of Staphylococcus aureus on pathogenesis in polymicrobial infections.

Polymicrobial infections involving Staphylococcus aureus exhibit enhanced disease severity and morbidity. We reviewed the nature of polymicrobial interactions between S. aureus and other bacterial, fungal, and viral cocolonizers. Microbes that were frequently recovered from the infection site with S. aureus are Haemophilus influenzae, Enterococcus faecalis, Pseudomonas aeruginosa, Streptococcus pneumoniae, Corynebacterium sp., Lactobacillus sp., Candida albicans, and influenza virus. Detailed analyses of several in vitro and in vivo observations demonstrate that S. aureus exhibits cooperative …

Development of a bacterin-toxoid vaccine using a Korean isolate for protection against caseous lymphadenitis in goats.

Caseous lymphadenitis (CLA), a chronic infectious disease caused by Corynebacterium pseudotuberculosis, remains a major concern in small ruminant populations owing to its persistent nature and impact on animal productivity. Despite the availability of commercial vaccines, their efficacy in goats has been inconsistent and field-based evidence supporting their use remains limited. In this study, we report the development and evaluation of a novel bacterin-toxoid vaccine formulated with C. pseudotuberculosis strain 51-12A obtained from a Korean native goat. The toxoid, prepared from culture super…

Bacteriological & epidemiological characteristics of diphtheria cases in & around Delhi -a retrospective study.

BACKGROUND & OBJECTIVE: Diphtheria infections caused by the different toxigenic biotypes of Corynebacterium diphtheriae are endemic in Delhi. Information on biochemical identification, toxigenicity and antimicrobial susceptibility to this bacterium is scanty. This retrospective study was carried out to identify isolated Corynebacteria biochemically, determine their toxigenicity, drug sensitivity and some epidemiological characteristics of diphtheria cases from Delhi and adjoining States for the period 1998-2004. METHODS: A total of 1118 throat and 585 nasal swabs were used to detect human pat…

The emergence of coryneform bacteria as a cause of nosocomial infections in compromised hosts.

Corynebacterium species that are normally abundant on the skin and mucous membranes rarely cause infections and are susceptible to most antibiotics. The report in 1976 of four cases of sepsis at the National Institutes of Health caused by a hitherto undescribed corynebacterium that is highly antibiotic resistant, but uniformly susceptible to vancomycin, alerted the medically oriented scientific community to the emergence of these organisms as a possible new cause of nosocomial infections. Although we have always performed antibiotic susceptibility tests on all microorganisms recovered from nor…

Idiopathic Granulomatous Mastitis, Erythema Nodosum, and Polyarthritis.

Idiopathic granulomatous mastitis (IGM) is an inflammatory-mediated rare disease that can be linked to rare manifestations. Erythema nodosum (EN) and polyarthritis, seen in a multitude of autoinflammatory and autoimmune diseases, have been rarely linked to IGM. Despite the cause of IGM being unclear, Corynebacterium infections are thought to play a role in the pathophysiology of IGM. Unusually, IGM has a relapsing and remitting course, which also applies to its systemic manifestations. As such, we present a case of IGM in a middle-aged lady who was initially thought to have Corynebacterium-con…

Clinical Identification of Two Novel C. kroppenstedtii-like Species Isolated as Pathogens of Granulomatous Lobular Mastitis.

Granulomatous lobular mastitis (GLM) is a rare benign breast inflammatory disease that affects women of childbearing age. Corynebacterium species, especially Corynebacterium kroppenstedtii, was reported as the pathogen of GLM. A recent study showed that the C. kroppenstedtii complex is composed of C. kroppenstedtii and two novel species, C. parakroppenstedtii and C. pseudokroppenstedtii. The study presents seven C. kroppenstedtii-like strains isolated from GLM patients. However, they turned out to be six strains of C. parakroppenstedtii and one strain of C. pseudokroppenstedtii according to 16…

Coryneform bacteria associated with canine otitis externa.

This study aims to investigate the occurrence of coryneform bacteria in canine otitis externa. A combined case series and case-control study was carried out to improve the current knowledge on frequency and clinical significance of coryneform bacteria in samples from canine otitis externa. A total of 16 cases of otitis externa with involvement of coryneform bacteria were recorded at two referral veterinary hospitals in Denmark and the US, respectively. Coryneform bacteria were identified by partial 16S rRNA gene sequencing. Corynebacterium auriscanis was the most common coryneform species (10 …

A case of fatal respiratory diphtheria imported from Poland to Germany: possible link to an undetected imported diphtheria cluster in Poland?

PURPOSE: Diphtheria is a re-emerging vaccine-preventable disease, mainly caused by toxigenic Corynebacterium diphtheriae. METHODS: Here, we report a fatal respiratory diphtheria infection in a Polish woman travelling to Germany possibly linked to a cutaneous diphtheria infection in a homeless man living in the same geographic area. Laboratory diagnostics involving MALDI-TOF MS, tox-gene PCR, Lateral Flow Immuno Assay, a modified Elek test and Next Generation Sequencing (NGS) identified the causative strain as cotrimoxazole-resistant toxigenic Corynebacterium diphtheriae biotype mitis, sequenc…
