Trans-Ferulic acid reduces the sedative activity of diazepam in thiopental sodium-induced sleeping mice: A potential GABAergic transmission.

trans-Ferulic acid (TFA), a bioactive compound found in many plants, has been recognized for its diverse pharmacological activities, including potential neurological benefits. Previous studies suggest that TFA exerts anxiolytic effects via GABAergic pathways. This study aimed to investigate the sedative effects of TFA and its possible molecular mechanisms through in vivo and in silico approaches. Adult Swiss mice were randomly divided into six groups (n = 7): control (vehicle), standard (DZP: Diazepam at 3 mg/kg, p.o.), three test groups (TFA at 25, 50, and 75 mg/kg, p.o.), and a combination g

Effects of Dietary Supplementation with a Ferulic Acid-Rich Bioactive Component of Wheat Bran in a Murine Model of Graft-Versus-Host Disease.

Graft-versus-host disease (GvHD) is a common and severe complication following allogeneic hematopoietic stem cell transplantation (HSCT). Its prevention and treatment is a major challenge. Ferulic acid (FA) has anti-inflammatory and antioxidant properties that could be attractive in this setting. Our aim was to evaluate a bioactive ingredient derived from wheat bran (WB), selected for its high concentration of FA, in a murine model of GvHD. The ingredient was obtained via a bioprocess involving hydrolysis and spray-drying. GvHD was induced via HSCT between MHC-mismatched mouse strains. FA trea

Preparative separation of high-purity trans- and cis-ferulic acid from wheat bran by pH-zone-refining counter-current chromatography.

Ferulic acid stereoisomers are the most abundant phenolic acids in cereal bran. However, it is challenging to separate them because of the similar structures and properties. In this study, a preparative separation method of ferulic acid stereoisomers from the crude extract of wheat bran was successfully developed. The method contained a two-step separation, the traditional counter-current chromatography (CCC, hexane: ethyl acetate: methanol: water = 2:5:2:4) was followed with a pH-zone-refining CCC (hexane: ethyl acetate: acetonitrile: water = 2:5:2:2, 10 mmol L-1 trifluoroacetic acid in organ

Enhanced Antioxidant Activity and Elevated Phenolic Content in Hydrolyzed Spent Coffee Ground Extracts for Stabilizing Edible Oils During Storage.

This study investigated how alkaline hydrolysis enhances the antioxidant activity of spent coffee ground (SCG) extracts in stripped soybean oil (SSBO) and stripped flaxseed oil (SFSO) during storage. Hydrolysis of SCG for 2 h markedly increased the concentrations of caffeic acid, p‑coumaric acid, ferulic acid, and total phenolic compounds, followed by gradual declines at 4 and 6 h. Among them, caffeic acid was the most abundant in the hydrolyzed SCG (HSCG) extracts, increasing from 992 to 17,862 µg/g in the ethanol extract and 965 to 20,540 µg/g in the acetone extract during the 2-h hydrolysis

Substrate and inhibitor specificities of the monocarboxylate transporters of single rat heart cells.

We have used the intracellular pH-sensitive fluorescent dye 2',7'-bis(carboxyethyl)-5(6)-carboxyfluorescein (BCECF) to characterize the substrate and inhibitor specificity of monocarboxylate transport into isolated rat heart cells. Further evidence was obtained for the presence of two lactate carriers present in heart cells (Wang et al., Biochem. J. 290: 249-258, 1993) both distinct from the recently cloned monocarboxylate transporter isoform 1 (MCT-1) found in many other cell types. Only one isoform was potently inhibited by alpha-cyano-4-hydroxycinnamate [CHC; inhibitor constant (Ki) 190 mic

Protective activity of caffeic acid and sinapic acid against UVB-induced photoaging in human fibroblasts.

In this study, the protective effects of caffeic acid (CA) and sinapic acid (SA) on photoaging in human skin fibroblasts induced by UVB irradiation (30 mJ/cm2 ) were examined. The results revealed that exposure to UVB irradiation reduced cell viability by approximately 33% compared with the non-UVB irradiated control. However, CA and SA blocked the UVB irradiation-induced cytotoxicity greater than the other phenolic acids. CA and SA also significantly inhibited the release of matrix metalloproteinase 1 (MMP-1) and reduced the expression of MMP-1 mRNA in UVB-irradiated Hs68 cells. Furthermore,

Ferulic acid suppresses glutamate release through inhibition of voltage-dependent calcium entry in rat cerebrocortical nerve terminals.

This study investigated the effects and possible mechanism of ferulic acid, a naturally occurring phenolic compound, on endogenous glutamate release in the nerve terminals of the cerebral cortex in rats. Results show that ferulic acid inhibited the release of glutamate evoked by the K⁺ channel blocker 4-aminopyridine (4-AP). The effect of ferulic acid on the evoked glutamate release was prevented by chelating the extracellular Ca²⁺ ions, but was insensitive to the glutamate transporter inhibitor DL-threo-beta-benzyl-oxyaspartate. Ferulic acid suppressed the depolarization-induced increase in a
